Prediction of pyrazinamide resistance in Mycobacterium tuberculosis using structure-based machine learning approaches

TL;DR: An approach to improve the sensitivity/specificity of pyrazinamide resistance prediction in genetics-based clinical microbiology workflows, highlights novel mutations for future biochemical investigation, and is a proof of concept for using this approach in other drugs such as bedaquiline.

TL;DR: Multi-label random forest models are compared with single-label Random Forest models for both predicting phenotypic resistance from whole genome sequences and identifying important mutations for better prediction of four first-line drugs in a dataset of Mycobacterium tuberculosis isolates.
