V. Piddock,Ian H. Gilbert,Heinz E. Moser,Rolf Müller +54 more
TL;DR: In this paper, the authors present a strategic blueprint to substantially improve our ability to discover and develop new antibiotics, and propose both short-term and long-term solutions to overcome the most urgent limitations in the various sectors of research and funding.
Journal ArticleDOI
Active-site residue, domain and module swaps in modular polyketide synthases
Francesca Del Vecchio,Hrvoje Petković,Steven G. Kendrew,Lindsey Low,Barrie Wilkinson,Rachel E. Lill,Jesus Cortes,Jesus Cortes,Brian A.M. Rudd,Brian A.M. Rudd,J. Staunton,Peter F. Leadlay +11 more
TL;DR: A cassette system for convenient construction of hybrid modular PKSs based on the tylosin PKS in Streptomyces fradiae is described and its use in domain and module swaps is demonstrated.
Journal ArticleDOI
Mutasynthesis of Rapamycin Analogues through the Manipulation of a Gene Governing Starter Unit Biosynthesis
Matthew Alan Gregory,Hrvoje Petković,Rachel E. Lill,Steven J. Moss,Barrie Wilkinson,Sabine Gaisser,Peter F. Leadlay,Rose Mary Sheridan +7 more
Journal ArticleDOI
Genetics of Streptomyces rimosus, the Oxytetracycline Producer
Hrvoje Petković,John Cullum,Daslav Hranueli,Iain S. Hunter,Natasa Peric-Concha,Jasenka Pigac,Arinthip Thamchaipenet,Dušica Vujaklija,Paul F. Long +8 more
TL;DR: From a genetic standpoint, Streptomyces rimosus is arguably the best-characterized industrial streptomycete as the producer of oxytetracycline and other tetracyCline antibiotics.
Journal ArticleDOI
Roles of rapH and rapG in Positive Regulation of Rapamycin Biosynthesis in Streptomyces hygroscopicus
Enej Kuščer,Nigel Coates,Iain R. Challis,Matthew Alan Gregory,Barrie Wilkinson,Rose Mary Sheridan,Hrvoje Petković +6 more
TL;DR: Based on DNA sequence analysis, the products of genes rapH and rapG showed high similarities with two different families of transcriptional activators, LAL and AraC, respectively, and indicated an essential role of the RapG regulator for rapamycin biosynthesis and a supportive role of RapH.
