Observing the cell in its native state: Imaging subcellular dynamics in multicellular organisms
Tsung-Li Liu,Srigokul Upadhyayula,Daniel E. Milkie,Ved P. Singh,Kai Wang,Ian A. Swinburne,Kishore R. Mosaliganti,Zach M. Collins,Tom W. Hiscock,Jamien Shea,Abraham Q. Kohrman,Taylor N Medwig,Daphné Dambournet,Ryan Forster,Brian Cunniff,Brian Cunniff,Yuan Ruan,Hanako Yashiro,Steffen Scholpp,Steffen Scholpp,Elliot M. Meyerowitz,Dirk Hockemeyer,David G. Drubin,Benjamin L. Martin,David Q. Matus,Minoru Koyama,Sean G. Megason,Tom Kirchhausen,Eric Betzig +28 more
TL;DR: AO-LLSM takes high-resolution live-cell imaging of subcellular processes from the confines of the coverslip to the more physiologically relevant 3D environment within whole transparent organisms and creates new opportunities to study the phenotypic diversity of intracellular dynamics, extracellular communication, and collective cell behavior across different cell types, organisms, and developmental stages.
Journal ArticleDOI
Cortical column and whole-brain imaging with molecular contrast and nanoscale resolution
Ruixuan Gao,Ruixuan Gao,Ruixuan Gao,Shoh Asano,Shoh Asano,Srigokul Upadhyayula,Igor Pisarev,Daniel E. Milkie,Tsung-Li Liu,Ved P. Singh,Austin R. Graves,Grace H. Huynh,Yongxin Zhao,John A. Bogovic,Jennifer Colonell,Carolyn Ott,Christopher T. Zugates,Susan Tappan,Alfredo Rodriguez,Kishore R. Mosaliganti,Shu-Hsien Sheu,H. Amalia Pasolli,Song Pang,C. Shan Xu,Sean G. Megason,Harald F. Hess,Jennifer Lippincott-Schwartz,Adam W. Hantman,Gerald M. Rubin,Tom Kirchhausen,Stephan Saalfeld,Yoshinori Aso,Edward S. Boyden,Eric Betzig +33 more
TL;DR: An approach for high-resolution tracing of neurons, their subassemblies, and their molecular constituents over large volumes is introduced, which can enable high-throughput comparative studies of neural development, circuit stereotypy, and structural correlations to neural activity or behavior.
Journal ArticleDOI
Specified Neural Progenitors Sort to Form Sharp Domains after Noisy Shh Signaling
Fengzhu Xiong,Andrea R. Tentner,Peng Huang,Arnaud Gelas,Kishore R. Mosaliganti,Lydie Souhait,Nicolas Rannou,Ian A. Swinburne,Nikolaus D. Obholzer,Paul D. Cowgill,Alexander F. Schier,Sean G. Megason +11 more
TL;DR: This work finds that specified progenitors of different fates are spatially mixed following heterogeneous Sonic Hedgehog signaling responses, and reveals that cell sorting acts to correct imprecision of spatial patterning by noisy inductive signals.
Journal ArticleDOI
ACME: automated cell morphology extractor for comprehensive reconstruction of cell membranes.
TL;DR: By using the ACME software to study somite formation, the method was able to segment touching cells with high accuracy and reliably quantify changes in morphogenetic parameters such as cell shape and size, and the arrangement of epithelial and mesenchymal cells.
Journal ArticleDOI
Interplay of Cell Shape and Division Orientation Promotes Robust Morphogenesis of Developing Epithelia
Fengzhu Xiong,Wenzhe Ma,Tom W. Hiscock,Kishore R. Mosaliganti,Andrea R. Tentner,Kenneth A. Brakke,Nicolas Rannou,Arnaud Gelas,Lydie Souhait,Ian A. Swinburne,Nikolaus D. Obholzer,Sean G. Megason +11 more
TL;DR: This work identifies an evolvable design logic that enables robust cell-level regulation of tissue-level development and predicts that cell shape is robust to changes of tissue surface area, cell volume, and cell number, which is confirmed in vivo.
