TL;DR: It is demonstrated that autophagy, which is often disabled in cancer, is dispensable for chemotherapy-induced cell death but required for its immunogenicity, and increased extracellular ATP concentrations improve the efficacy of antineoplastic chemotherapies when Autophagy is disabled.
Journal ArticleDOI
Regulation of Autophagy by Cytosolic Acetyl-Coenzyme A
Guillermo Mariño,Federico Pietrocola,Tobias Eisenberg,Yongli Kong,Shoaib Ahmad Malik,Aleksandra Andryushkova,Sabrina Schroeder,Tobias Pendl,Alexandra Harger,Mireia Niso-Santano,Naoufal Zamzami,Marie Scoazec,Silvère Durand,David Enot,Álvaro F. TL;DR: It is shown that nutrient starvation causes rapid depletion of AcCoA, and multiple distinct manipulations designed to increase or reduce cytosolic Ac CoA led to the suppression or induction of autophagy, respectively, both in cultured human cells and in mice, delineating Ac coenzyme A-centered pharmacological strategies that allow for the therapeutic manipulation ofautophagy.
Journal ArticleDOI

TL;DR: Results indicate that PDXK expression levels constitute a biomarker for risk stratification among patients with NSCLC, in line with a general role of vitamin B6 in stress responses.
Journal ArticleDOI
Erlotinib antagonizes ABC transporters in acute myeloid leukemia
Elodie Lainey,Marie Sebert,Sylvain Thepot,Marie Scoazec,Cyrielle Bouteloup,Carole Leroy,Stéphane de Botton,Lorenzo Galluzzi,Pierre Fenaux,Guido Kroemer +9 more
TL;DR: It is shown that both erlotinib and gefitinib inhibit transmembrane transporters of the ATP-binding cassette (ABC) family, including P-glycoprotein (P-gp), multidrug resistance-associated proteins (MRPs) and breast cancer resistance protein (BCRP), in acute myeloid leukemia (AML) cells that do not overexpress these pumps.
Book ChapterDOI
Metabolomic Profiling of Cultured Cancer Cells
TL;DR: A simple procedure to determine the complete complement of intracellular metabolites in cultured malignant cells by HPLC coupled to Q-TOF HRMS, which is expected to provide reliable metabolomic data on a variety of cellular samples.
