Universal sample preparation method for proteome analysis

Clinical resistance is a complex phenomenon in major human cancers involving multifactorial mechanisms, and hypoxia is one of the key components that affect the cellular expression program and lead to therapy resistance. The present study aimed to summarize the role of hypoxia in cancer therapy by regulating the tumor microenvironment (TME) and to highlight the potential of hypoxia-targeted therapy. Relevant published studies were retrieved from PubMed, Web of Science, and Embase using keywords such as hypoxia, cancer therapy, resistance, TME, cancer, apoptosis, DNA damage, autophagy, p53, and other similar terms. Recent studies have shown that hypoxia is associated with poor prognosis in patients by regulating the TME. It confers resistance to conventional therapies through a number of signaling pathways in apoptosis, autophagy, DNA damage, mitochondrial activity, p53, and drug efflux. Hypoxia targeting might be relevant to overcome hypoxia-associated resistance in cancer treatment.

Role of hypoxia in cancer therapy by regulating the tumor microenvironment

In an in vitro culture system of human cancer cells MCF-7 and its adriamycin-resistant line MCF-7/Ad or KB and its vincristine-resistant line KBv200, tetrandrine was found to exhibit significantly selective anticancer activity against the drug-resistant cancer cell MCF-7/Ad. In addition, essentially complete reversal effects of tetrandrine on drug resistance were observed in MCF/Ad or KBv200.

This study was performed to investigate the chemical constituents in the twigs and leaves of Harrisonia perforate. Six compounds were isolated from the 95% EtOH extract of the twigs and leaves of Harrisonia perforate by silica gel, ODS, Sephadex LH-20 column chromatographies and preparative HPLC. On the basis of chemical properties and spectra data, these compounds were identified as harriperfin E (1), kihadanin A (2), kihadanin B (3), 6α-acetoxyobacunol acetate (4), gardaubryone C (5), and β-sitosterol methyl ether (6), respectively. Compound 1 is a new chromone, and compounds 2-6 are isolated from this plant for the first time.

It has been known that the VEGF/VEGFR signaling pathway plays an important role in angiogenesis and it is overactive in many types of solid tumors. Since tumor angiogenesis is vital for tumor cell survival, proliferation, migration, and invasion, antiangiogenic therapy has been extensively explored. Currently, a lot of tumor angiogenesis inhibitors (TAIs) have appeared in the market or in different stages of clinical trials, and several of them have been commonly applied in cancer treatment alone, or in combination with other chemotherapeutic agents (CTAs) or radiotherapy. This chapter will introduce several of the angiogenesis inhibitors, their antiangiogenic pharmacological mechanisms, new indications, and clinical trial results in chemotherapy, with a focus on bevacizumab, ramucirumab, sorafenib, and sunitinib.
