Abstract: The basic amino acids lysine (Lys) and arginine (Arg) play important roles in membrane protein activity, the sensing of membrane voltages, and the actions of antimicrobial, toxin, and cell-penetrating peptides. These roles are thought to stem from the strong interactions and disruptive influences of these amino acids on lipid membranes. In this study, we employ fully atomistic molecular dynamics simulations to observe, quantify, and compare the interactions of Lys and Arg with saturated phosphatidylcholine membranes of different thickness. We make use of both charged (methylammonium and methylguanidinium) and neutral (methylamine and methylguanidine) analogue molecules, as well as Lys and Arg side chains on transmembrane helix models. We find that the free energy barrier experienced by a charged Lys crossing the membrane is strikingly similar to that of a charged Arg (to within 2 kcal/mol), despite the two having different chemistries, H-bonding capability, and hydration free energies that differ by ∼10 k...

Abstract: Using differential display (DD), we discovered a new member of the serine protease family of protein-cleaving enzymes, named protease M. The gene is most closely related by sequence to the kallikreins, to prostate-specific antigen (PSA), and to trypsin. The diagnostic use of PSA in prostate cancer suggested that a related molecule might be a predictor for breast or ovarian cancer. This, in turn, led to studies designed to characterize the protein and to screen for its expression in cancer. The isolation of protease M by DD, the cloning and sequencing of the cDNA, and the comparison of the predicted protein structure with related proteins are described, as are methods to produce recombinant proteins and polyclonal antibody preparations. Protease M expression was examined in mammary, prostate, and ovarian cancer, as well as normal, cells and tissues. Stable transfectants expressing the protease M gene were produced in mammary carcinoma cells. Protease M was localized by fluorescent in situ hybridization analysis to chromosome 19q13.3, in a region to which other kallikreins and PSA also map. The gene is expressed in the primary mammary carcinoma lines tested but not in the corresponding cell lines of metastatic origin. It is strongly expressed in ovarian cancer tissues and cell lines. The enzyme activity could not be established, because of difficulties in producing sufficient recombinant protein, a common problem with proteases. Transfectants were selected that overexpress the mRNA, but the protein levels remained very low. Protease M expression (mRNA) may be a useful marker in the detection of primary mammary carcinomas, as well as primary ovarian cancers. Other medical applications are also likely, based on sequence relatedness to trypsin and PSA.

Abstract: A conductometric H2, NO2, and hydrocarbon gas sensor based on single-crystalline zinc oxide (ZnO) nanobelts has been developed. The nanobelt sensitive layer was deposited using a radio frequency (RF) magnetron sputterer. The microcharacterization study reveals that the nanobelts have a single crystal hexagonal structure with average thickness and width of about 10 and 50nm, respectively. The sensor was exposed to H2, NO2 and propene gases at operating temperatures between 150 degC and 450 degC. The study showed that optimum operating temperatures for the sensor are in the range of 300 degC-400degC for H2, 300 degC-350 degC for NO2, and 350 degC-420 degC for propene sensing

Abstract: The electrochemical N2 reduction reaction (NRR) offers a direct pathway to produce NH3 from renewable energy. However, aqueous NRR suffers from both low Faradaic efficiency (FE) and low yield rate. The main reason is the more favored H+ reduction to H2 in aqueous electrolytes. Here we demonstrate a highly selective Ru/MoS2 NRR catalyst on which the MoS2 polymorphs can be controlled to suppress H+ reduction. A NRR FE as high as 17.6% and NH3 yield rate of 1.14 × 10–10 mol cm–2 s–1 are demonstrated at 50 °C. Theoretical evidence supports a hypothesis that the high NRR activity originates from the synergistic interplay between the Ru clusters as N2 binding sites and nearby isolated S-vacancies on the 2H-MoS2 as centers for hydrogenation; this supports formation of NH3 at the Ru/2H-MoS2 interface.

Abstract: Introduction Higher midlife body mass index (BMI) is suggested to increase the risk of dementia, but weight loss during the preclinical dementia phase may mask such effects. Methods We examined this hypothesis in 1,349,857 dementia-free participants from 39 cohort studies. BMI was assessed at baseline. Dementia was ascertained at follow-up using linkage to electronic health records (N = 6894). We assumed BMI is little affected by preclinical dementia when assessed decades before dementia onset and much affected when assessed nearer diagnosis. Results Hazard ratios per 5-kg/m 2 increase in BMI for dementia were 0.71 (95% confidence interval = 0.66–0.77), 0.94 (0.89–0.99), and 1.16 (1.05–1.27) when BMI was assessed 10 years, 10-20 years, and >20 years before dementia diagnosis. Conclusions The association between BMI and dementia is likely to be attributable to two different processes: a harmful effect of higher BMI, which is observable in long follow-up, and a reverse-causation effect that makes a higher BMI to appear protective when the follow-up is short.
