This protocol provides an overview of the comparative CT method for quantitative gene expression studies and various examples to present quantitative gene Expression data using this method.Abstract:
Two different methods of presenting quantitative gene expression exist: absolute and relative quantification. Absolute quantification calculates the copy number of the gene usually by relating the PCR signal to a standard curve. Relative gene expression presents the data of the gene of interest relative to some calibrator or internal control gene. A widely used method to present relative gene expression is the comparative C(T) method also referred to as the 2 (-DeltaDeltaC(T)) method. This protocol provides an overview of the comparative C(T) method for quantitative gene expression studies. Also presented here are various examples to present quantitative gene expression data using this method.read more

EDB fibronectin specific peptide for prostate cancer targeting
Zheng Han,Zhuxian Zhou,Xiaoyue Shi,Junpeng Wang,Xiaohui Wu,Da Sun,Yinghua Chen,Hui Zhu,Cristina Magi-Galluzzi,Zheng-Rong Lu +9 more
TL;DR: The development of a small peptide specific to EDB-FN for targeting prostate cancer is reported, which shows stronger binding to human prostate tumor specimens with a higher Gleason score compared to those with a lower score, with no binding in benign prostatic hyperplasia.

Rac2 controls tumor growth, metastasis and M1-M2 macrophage differentiation in vivo.
Shweta Joshi,Alok R. Singh,Muamera Zulcic,Lei Bao,Karen Messer,Trey Ideker,Janusz Dutkowski,Donald L. Durden +7 more
TL;DR: It is demonstrated that the small GTPase, Rac2 controls macrophage M1 to M2 differentiation and the metastatic phenotype in vivo and a novel molecular mechanism by which signals transmitted from the extracellular matrix via the α4β1 integrin and MCSF receptor lead to the activation of Rac2 and potentially regulate macrophages M2 differentiate.

Transcription factor WRKY22 promotes aluminum tolerance via activation of OsFRDL4 expression and enhancement of citrate secretion in rice (Oryza sativa)
Ge Zi Li,Ge Zi Li,Zhan Qi Wang,Kengo Yokosho,Bing Ding,Wei Fan,Qiao Qiao Gong,Gui Xin Li,Yun Rong Wu,Jian Li Yang,Jian Feng Ma,Shao Jian Zheng +11 more
TL;DR: It is concluded that OsWRKY22 promotes Al-induced increases in OsFRDL4 expression, thus enhancing Al- induced citrate secretion and Al tolerance in rice.

Interspecies NASH disease activity whole-genome profiling identifies a fibrogenic role of PPARα-regulated dermatopontin
Philippe Lefebvre,Fanny Lalloyer,Eric Baugé,Michał Pawlak,Céline Gheeraert,Hélène Dehondt,Jonathan Vanhoutte,Eloise Woitrain,Nathalie Hennuyer,Claire Mazuy,Marie Bobowski-Gérard,Francesco Paolo Zummo,Bruno Derudas,Ann Driessen,Guy Hubens,Luisa Vonghia,Wilhelmus J. Kwanten,Peter Michielsen,Thomas Vanwolleghem,Jérôme Eeckhoute,An Verrijken,Luc Van Gaal,Sven Francque,Bart Staels +23 more
TL;DR: Liver fibrotic histological damages are characterized by the deregulated expression of a restricted set of inflammation- and ECM-related genes, and dermatopontin may be a valuable target to reverse the hepatic fibrotics process.

Fluoxetine treatment affects the inflammatory response and microglial function according to the quality of the living environment.
Silvia Alboni,S. Poggini,Stefano Garofalo,Giampaolo Milior,Hassan El Hajj,Cynthia Lecours,Isabelle Girard,Steven Gagnon,Samuel Boisjoly-Villeneuve,Nicoletta Brunello,David P. Wolfer,Cristina Limatola,Marie-Ève Tremblay,Laura Maggi,Igor Branchi +14 more
TL;DR: Results show that the effects of fluoxetine treatment on inflammation and microglial function, as compared to vehicle, were dependent on the quality of the living environment and may be crucial to devise pharmacological strategies aimed at enhancing antidepressant efficacy by means of controlling environmental conditions.

A new mathematical model for relative quantification in real-time RT-PCR.
TL;DR: This study enters into the particular topics of the relative quantification in real-time RT-PCR of a target gene transcript in comparison to a reference gene transcript and presents a new mathematical model that needs no calibration curve.

Cluster analysis and display of genome-wide expression patterns
TL;DR: A system of cluster analysis for genome-wide expression data from DNA microarray hybridization is described that uses standard statistical algorithms to arrange genes according to similarity in pattern of gene expression, finding in the budding yeast Saccharomyces cerevisiae that clustering gene expression data groups together efficiently genes of known similar function.
