TLDR
In the presence of insulin, resting 3T3-L1 fibroblasts differentiate into adipocytes after a lag period of 12 to 20 days and indomethacin shortens the lag period dramatically and increases the rate of expression of lipogenic activity.

Understanding Adipocyte Differentiation

TL;DR: Characterization of regulatory regions of adipose-specific genes has led to the identification of the transcription factors peroxisome proliferator-activated receptor-gamma and CCAAT/enhancer binding protein (C/EBP), which play a key role in the complex transcriptional cascade during adipocyte differentiation.

Peroxisome Proliferator-activated Receptors α and γ Are Activated by Indomethacin and Other Non-steroidal Anti-inflammatory Drugs

TL;DR: It is shown that indomethacin binds and activates peroxisome proliferator-activated receptor γ (PPARγ), a ligand-activated transcription factor known to play a pivotal role in adipogenesis, and several other NSAIDs are also PPARγ ligands and induce adipocyte differentiation of C3H10T1/2 cells.

Differentiation of muscle, fat, cartilage, and bone from progenitor cells present in a bone-derived clonal cell population: effect of dexamethasone.

TL;DR: The data suggest that this clonal cell line contains subpopulations of mesenchymal progenitor cells which can, under the influence of glucocorticoid hormones, differentiate in vitro into four distinct cell types.

Development of hormone receptors and hormonal responsiveness in vitro. Insulin receptors and insulin sensitivity in the preadipocyte and adipocyte forms of 3T3-L1 cells.

TL;DR: This new procedure permitted an analysis of insulin binding activity and cellular responsiveness to insulin during adipocyte conversion in vitro and found that cells appeared to be adipocytes by morphological and biochemical criteria and the specific insulin binding was 6to lo-fold greater than in undifferentiated control cells.

Modulation of Rho GTPase Signaling Regulates a Switch between Adipogenesis and Myogenesis

TL;DR: Together, these results identify the Rho GTPase as an essential modulator of IGF-1 signals that direct the adipogenesis-myogenesis cell fate decision.

An established preadipose cell line and its differentiation in culture II. Factors affecting the adipose conversion

TL;DR: Lipogenic and lipolytic hormones and drugs affect the rate of synthesis and accumulation of triglyceride by 3T3-L1 cells, but in contrast to bromodeoxyuridine, these modulating agents do not seem to affect the proportion of cells which undergoes the adipose conversion.

An established pre-adipose cell line and its differentiation in culture

TL;DR: The conversion of 3T3-L1 from pre-adipose to adipose cells therefore involves a process of differentiation which can be studied under cell culture conditions.

Sublines of mouse 3T3 cells that accumulate lipid

TL;DR: Two clonal sublines are isolated from the established mouse fibroblast line 3T3 that accumulate large amounts of triglyceride fat when the cells are in the resting state and are reduced by lipolytic agents.
Journal ArticleDOI

Induction of lipogenesis during differentiation in a "preadipocyte" cell line.

TL;DR: 3T3-L1 fibroblasts differentiate in culture into cells having adipocyte character and the elevated acetyl-CoA carboxylase activity is the product of an increased cellular enzyme level.
Journal ArticleDOI

Cyclic AMP increases the concentration of insulin receptors in cultured fibroblasts and lymphocytes.

TL;DR: The increase in insulin receptors was a specific response to cyclic AMP as the number of receptors for human growth hormone was unaltered and was independent of changes in cell cycle.
