Journal ArticleDOI

The role of cytochrome P450 enzymes in hepatic and extrahepatic human drug toxicity

TL;DR: Avoidance of toxicity may be possible in rare instances by prediction of individual susceptibility or by designing new chemical entities that are metabolised by a range of enzymes (both cytochromes P450 and others) and do not undergo bioactivation.
Journal ArticleDOI

Liquid chromatography-electrospray mass spectrometry determination of carbamazepine, oxcarbazepine and eight of their metabolites in human plasma.

TL;DR: A specific and sensitive liquid chromatography-electrospray ionization mass spectrometry method was described for the simultaneous determination of carbamazepine, oxcarbazepine and eight of their metabolites in human plasma and found suitable for the analysis of plasma samples collected during therapeutic drug monitoring of patients treated with CBZ or OXCBZ.
Journal ArticleDOI

Immunological principles of adverse drug reactions: the initiation and propagation of immune responses elicited by drug treatment.

TL;DR: The purpose of this review is to classify drug hypersensitivity reactions in terms of their clinical presentation, and to consider recent advances in the understanding of the chemical, biochemical and cellular immunological mechanisms of hypersensitivity.
Journal ArticleDOI

Cytochrome P450 1B1: a major P450 isoenzyme in human blood monocytes and macrophage subsets

TL;DR: The expression of several CYPs in monocytes/macrophages suggests that these cells may be important in the metabolism of small molecular weight compounds, which play a role in allergic contact dermatitis and drug reactions.
Book ChapterDOI

The danger hypothesis applied to idiosyncratic drug reactions.

TL;DR: No one model fits the characteristics of all idiosyncratic drug reactions; however, the danger model provides a new perspective and suggests avenues of research that have the potential to increase the ability to predict and prevent such reactions significantly.

Studies on the sensitization of animals with simple chemical compounds

TL;DR: Experiments on the sensitization of guinea pigs with simple chemical compounds are described, with positive effects obtained by the administration of small quantities with 1:2:4 chlorodinitrobenzene, p-nitrosodimethylaniline, and a number of other aromatic compounds.
Journal ArticleDOI

Immune-complex glomerulopathy in patients treated with captopril

TL;DR: Renal biopsies were performed on 13 patients treated with captopril and findings indicated an early stage of membranous glomerulopathy (MGP), which may be associated with the development of immune-complex glomerULopathy.
Journal ArticleDOI

Antibodies to the surface of halothane-altered rabbit hepatocytes in patients with severe halothane-associated hepatitis.

TL;DR: Control studies using serum from subjects repeatedly exposed to halothane without the development of liver damage, and from patients with viral and toxic liver injury have confirmed the specificity of these findings to serve Halothane-associated liver injury.
Journal ArticleDOI

Biochemical Mechanisms in Chemical-Induced Lung Injury: Roles of Metabolic Activation

TL;DR: In this paper, the role of metabolic activation in Chemical-Induced Lung Injury was discussed and a detailed review of the biochemical mechanism of lung injury was presented. But this paper focused on lung injury.
