TLDR
Although augmented concentrations of ROS and faulty antioxidant defense have been associated to NAFLD and related complications, mechanisms of action and proofs of principle should be highlighted to support the causative role of OxS and to translate its concept into the clinic.Abstract:
Significance: Nonalcoholic fatty liver disease (NAFLD), characterized by liver triacylglycerol build-up, has been growing in the global world in concert with the raised prevalence of cardiometabolic disorders, including obesity, diabetes, and hyperlipemia. Redox imbalance has been suggested to be highly relevant to NAFLD pathogenesis. Recent Advances: As a major health problem, NAFLD progresses to the more severe nonalcoholic steatohepatitis (NASH) condition and predisposes susceptible individuals to liver and cardiovascular disease. Although NAFLD represents the predominant cause of chronic liver disorders, the mechanisms of its development and progression remain incompletely understood, even if various scientific groups ascribed them to the occurrence of insulin resistance, dyslipidemia, inflammation, and apoptosis. Nevertheless, oxidative stress (OxS) more and more appears as the most important pathological event during NAFLD development and the hallmark between simple steatosis and NASH manif...read more

Polygonatum sibiricum polysaccharides protect against obesity and non-alcoholic fatty liver disease in rats fed a high-fat diet
TL;DR: In this article , a high-fat diet-induced rat model of obesity and nonalcoholic fatty liver disease (NAFLD) was established to explore the potential protective effect of polygonatum sibiricum polysaccharides and the mechanisms behind it.

Liraglutide improves hepatic steatosis and metabolic dysfunctions in a 3-week dietary mouse model of nonalcoholic steatohepatitis
Thibaut Duparc,François Briand,Charlotte Trenteseaux,Jules Merian,Guillaume Combes,Souad Najib,Thierry Sulpice,Laurent O. Martinez +7 more
TL;DR: This study provides a novel 3-week dietary model of mice that rapidly develop NASH features, and this model will be suitable for evaluating the therapeutic efficacy of compounds in preclinical drug development for NASH.

Evaluation of Hepatoprotective Activity of the Crude Extract and Solvent Fractions of Clutia abyssinica ( Euphorbiaceae ) Leaf Against CCl 4 -Induced Hepatotoxicity in Mice
TL;DR: Clutia abyssinica is endowed with hepatoprotective activity, probably mediated via its antioxidant activity, and can be taken as one candidate for the development of hepatoprotsective agents because of its good safety profile.

Heme oxygenase-1 induction by hemin prevents oxidative stress-induced acute cholestasis in the rat.
Pamela Lucía Martín,Paula Ceccatto,María Valeria Razori,Daniel E. Francés,Sandra Mónica María Arriaga,Gerardo B. Pisani,Alejandra I. Martínez,Enrique J. Sánchez Pozzi,Marcelo G. Roma,Cecilia Lorena Basiglio +9 more
TL;DR: In vivo HO-1 induction protects the liver from acute oxidative injury, thus preventing consequent cholestasis, and reveals an important role for the induction of HO- 1 and the consequently elevated levels of BR in preserving biliary secretory function under OS conditions, thus representing a novel therapeutic tool to limit the cholESTatic injury that bears an oxidative background.

Impact of Sodium Glucose Cotransporter 2 Inhibitors on Nonalcoholic Fatty Liver Disease Complicated by Diabetes Mellitus
TL;DR: Several SGLT2 inhibitors are reviewed that are capable of protecting individuals with NAFLD from severe complications by inhibiting de novo lipogenesis, oxidative responses, inflammation induction, and hepatocyte death.

Global epidemiology of nonalcoholic fatty liver disease—Meta‐analytic assessment of prevalence, incidence, and outcomes
Zobair M. Younossi,Zobair M. Younossi,Aaron B. Koenig,Dinan Abdelatif,Yousef Fazel,Linda Henry,Mark Wymer,Mark Wymer +7 more
TL;DR: As the global epidemic of obesity fuels metabolic conditions, the clinical and economic burden of NAFLD will become enormous, and random‐effects models were used to provide point estimates of prevalence, incidence, mortality and incidence rate ratios.

Nonalcoholic fatty liver disease.
TL;DR: Nonalcoholic fatty liver disease is associated with an increased risk of all-cause death, probably because of complications of insulin resistance such as vascular disease, as well as due to cirrhosis and hepatocellular carcinoma, which occurs in a minority of patients.

Mitochondrial formation of reactive oxygen species.
TL;DR: This review describes the main mitochondrial sources of reactive species and the antioxidant defences that evolved to prevent oxidative damage in all the mitochondrial compartments and discusses various physiological and pathological scenarios resulting from an increased steady state concentration of mitochondrial oxidants.
