TLDR
The quality scores of a protein are displayed in the context of all known protein structures and problematic parts of a structure are shown and highlighted in a 3D molecule viewer in the ProSA-web service.Abstract:
A major problem in structural biology is the recognition of errors in experimental and theoretical models of protein structures. The ProSA program (Protein Structure Analysis) is an established tool which has a large user base and is frequently employed in the refinement and validation of experimental protein structures and in structure prediction and modeling. The analysis of protein structures is generally a difficult and cumbersome exercise. The new service presented here is a straightforward and easy to use extension of the classic ProSA program which exploits the advantages of interactive web-based applications for the display of scores and energy plots that highlight potential problems spotted in protein structures. In particular, the quality scores of a protein are displayed in the context of all known protein structures and problematic parts of a structure are shown and highlighted in a 3D molecule viewer. The service specifically addresses the needs encountered in the validation of protein structures obtained from X-ray analysis, NMR spectroscopy and theoretical calculations. Identification, Activity and Disulfide Connectivity of C-di-GMP Regulating Proteins in Mycobacterium tuberculosis
TL;DR: The homologous protein from Mycobacterium tuberculosis (Rv 1354c) named as MtbDGC is identified and characterized and the pattern of disulfide connectivity obtained by Mass spectrometric analysis is supported.

A thaumarchaeal provirus testifies for an ancient association of tailed viruses with archaea.
Mart Krupovic,Anja Spang,Simonetta Gribaldo,Patrick Forterre,Patrick Forterre,Christa Schleper +5 more
TL;DR: It is shown that the putative provirus is related to tailed bacterial and euryarchaeal viruses and encodes a full complement of proteins that are required to build a tailed virion, suggesting that an association of these viruses with Archaea might be more ancient than previously anticipated.

TL;DR: Sequence and structural analyses suggest that the reduced sphingomyelinase activity observed in some class IIb PLDs is probably due to point mutations which lead to a different substrate preference.

Assessment of the structural and functional impact of in-frame mutations of the DMD gene, using the tools included in the eDystrophin online database.
Aurélie Nicolas,Aurélie Nicolas,Céline Lucchetti-Miganeh,Céline Lucchetti-Miganeh,Rabah Ben Yaou,Jean-Claude Kaplan,Jamel Chelly,Frédérique Barloy-Hubler,Elisabeth Le Rumeur +8 more
TL;DR: Structural and functional modifications of dystrophin caused by in-frame mutations are highlighted and hybrid repeats were reconstituted at the deletion site in some cases, showing that hybrid repeats harbored the typical triple coiled-coil structure of native repeats, which may be correlated with better function in muscle cells.

Synthesis, Biological Evaluation, and Molecular Simulation of Chalcones and Aurones as Selective MAO-B Inhibitors
Nicole Morales-Camilo,Cristian O. Salas,Claudia Sanhueza,Christian Espinosa-Bustos,Silvia Sepúlveda-Boza,Miguel Reyes-Parada,Miguel Reyes-Parada,Fernando D. González-Nilo,Marcos CaroliRezende,Angélica Fierro +9 more
TL;DR: Results show that aurones, which had not been previously reported as MAOi, are MAO‐B inhibitors, and both families inhibited selectively the B isoform of MAO in the micromolar range, offering novel scaffolds for the design of new and potent MAO inhibitors.

Structure of a bacterial multidrug ABC transporter
TL;DR: The observed, outward-facing conformation reflects the ATP-bound state, with the two nucleotide-binding domains in close contact and the two transmembrane domains forming a central cavity—presumably the drug translocation pathway—that is shielded from the inner leaflet of the lipid bilayer and from the cytoplasm, but exposed to the outer leaflet and the extracellular space.

TL;DR: A prototype of a new approach to the folding problem of polypeptide chains based on the analysis of known protein structures, which derives the energy potentials for the atomic interactions of all amino acid residue pairs as a function of the distance between the involved atoms is presented.
