Abstract
A major problem in structural biology is the recognition of errors in experimental and theoretical models of protein structures. The ProSA program (Protein Structure Analysis) is an established tool which has a large user base and is frequently employed in the refinement and validation of experimental protein structures and in structure prediction and modeling. The analysis of protein structures is generally a difficult and cumbersome exercise. The new service presented here is a straightforward and easy to use extension of the classic ProSA program which exploits the advantages of interactive web-based applications for the display of scores and energy plots that highlight potential problems spotted in protein structures. In particular, the quality scores of a protein are displayed in the context of all known protein structures and problematic parts of a structure are shown and highlighted in a 3D molecule viewer. The service specifically addresses the needs encountered in the validation of protein structures obtained from X-ray analysis, NMR spectroscopy and theoretical calculations. ProSA-web is accessible at https://prosa.services.came.sbg.ac.at.

A novel mutation of the high-temperature requirement A serine peptidase 1 (HTRA1) gene in a Chinese family with cerebral autosomal recessive arteriopathy with subcortical infarcts and leukoencephalopathy (CARASIL).

TL;DR: Three-dimensional models showed that the change from proline to leucine could attenuate the hydrogen bond between S284 and S287 residues, which might affect function of serine protease HTRA1.

In-silico screening and experimental validation reveal L-Adrenaline as anti-biofilm molecule against biofilm-associated protein (Bap) producing Acinetobacter baumannii.

TL;DR: The finding suggests that L-Adrenaline can be developed to inhibit biofilm formation by carbapenem resistant strain of Acinetobacter baumannii.

Designing a multi-epitopic vaccine against the enterotoxigenic Bacteroides fragilis based on immunoinformatics approach.

TL;DR: The outer membrane toxin producing proteins found exclusively in the toxigenic B. fragilis were systematically ventured through the in-silico approaches to predict a multi-epitopic chimeric vaccine construct and proved to be exclusive.

Eeyarestatin compounds selectively enhance Sec61-mediated Ca2+ leakage from the endoplasmic reticulum

TL;DR: It is shown that ES1 impairs Ca2+ homeostasis by enhancing theCa2+ leakage from mammalian ER by interacting with the lateral gate of Sec61α, ES1 and ES24 likely capture Sec61 complexes in a Ca2-permeable, open state, in which Sec61 complex allow Ca2+.

Identification of lead molecules against potential drug target protein MAPK4 from L. donovani: An in-silico approach using docking, molecular dynamics and binding free energy calculation.

TL;DR: The current study reveals the identification of natural inhibitors of L. donovani mitogen-activated protein kinase-4 (LdMPK4) and identifies two compounds Genistein (GEN) and Chrysin (CHY), which can be considered as lead molecules for targeting Ld MPK4 enzyme and could emerge as potential LdMPk4 inhibitors.

Structure of a bacterial multidrug ABC transporter

TL;DR: The observed, outward-facing conformation reflects the ATP-bound state, with the two nucleotide-binding domains in close contact and the two transmembrane domains forming a central cavity—presumably the drug translocation pathway—that is shielded from the inner leaflet of the lipid bilayer and from the cytoplasm, but exposed to the outer leaflet and the extracellular space.
