Insertional oncogenesis in 4 patients after retrovirus-mediated gene therapy of SCID-X1
Salima Hacein-Bey-Abina,Alexandrine Garrigue,Gary P. Wang,Jean Soulier,Annick Lim,Estelle Morillon,Emmanuelle Clappier,Laure Caccavelli,Eric Delabesse,Kheira Beldjord,Vahid Asnafi,Elizabeth Macintyre,Liliane Dal Cortivo,Isabelle Radford,Nicole Brousse,François Sigaux,Despina Moshous,Julia Hauer,Arndt Borkhardt,Bernd H. Belohradsky,Uwe Wintergerst,Maria C. Velez,Lily E. Leiva,Ricardo U. Sorensen,NM Wulffraat,Stéphane Blanche,Frederic D. Bushman,Alain Fischer,Marina Cavazzana-Calvo +28 more
TL;DR: These findings functionally specify a genetic network that controls growth in T cell progenitors and led to sustained remission in 3 of the 4 cases of T cell leukemia, but failed in the fourth.
Journal ArticleDOI
Vector integration is nonrandom and clustered and influences the fate of lymphopoiesis in SCID-X1 gene therapy
Annette Deichmann,Salima Hacein-Bey-Abina,Manfred Schmidt,Manfred Schmidt,Alexandrine Garrigue,Martijn H. Brugman,Jingqiong Hu,Hanno Glimm,Gabor Gyapay,Bernard Prum,Christopher Fraser,Nicolas Fischer,Kerstin Schwarzwaelder,Kerstin Schwarzwaelder,Maria Luise Siegler,Dick de Ridder,Dick de Ridder,Karin Pike-Overzet,Steven J. Aronow,Christophe Hue,Claudia Prinz,Manuela Wissler,Chuck Klanke,Jean Weissenbach,Ian E. Alexander,Alain Fischer,Christof von Kalle,Marina Cavazzana-Calvo +34 more
TL;DR: The results obtained from a large-scale mapping of retroviral integration sites isolated from cells of 9 patients with X-linked SCID (SCID-X1) treated with a retrovirus-based gene therapy protocol help to elucidate the relationship between vector insertion and long-term in vivo selection of transduced cells in human patients with SCID- X1.
Journal ArticleDOI
Therapeutic effect of JAK1/2 blockade on the manifestations of hemophagocytic lymphohistiocytosis in mice
Sophia Maschalidi,Sophia Maschalidi,Fernando E. Sepulveda,Fernando E. Sepulveda,Alexandrine Garrigue,Alexandrine Garrigue,Alain Fischer,Geneviève de Saint Basile,Geneviève de Saint Basile,Geneviève de Saint Basile +9 more
TL;DR: It is demonstrated that clinically relevant doses of the JAK1/2 inhibitor ruxolitinib suppresses the harmful consequences of macrophage overactivation characterizing HLH in 2 murine models and could be readily translated into the clinic for the treatment of primary, and perhaps even secondary, forms of HLH.
Journal ArticleDOI
Germline HAVCR2 mutations altering TIM-3 characterize subcutaneous panniculitis-like T cell lymphomas with hemophagocytic lymphohistiocytic syndrome
Tenzin Gayden,Fernando E. Sepulveda,Dong-Anh Khuong-Quang,Dong-Anh Khuong-Quang,Jonathan Pratt,Elvis Terci Valera,Elvis Terci Valera,Alexandrine Garrigue,Susan Kelso,Susan Kelso,Frank Sicheri,Frank Sicheri,Leonie G. Foulkes,Alain Fischer,Alain Fischer,Alain Fischer,Bénédicte Neven,Bénédicte Neven,David Michonneau,Geneviève de Saint Basile,Geneviève de Saint Basile,Nada Jabado,Nada Jabado +56 more
TL;DR: G germline loss-of-function mutations in HAVCR2, which encodes the immune modulator TIM-3, are found in individuals with subcutaneous panniculitis-like T cell lymphomas and hemophagocytic lymphohistiocytosis, a life-threatening inflammatory condition.
Journal ArticleDOI
A human postnatal lymphoid progenitor capable of circulating and seeding the thymus
Emmanuelle Six,Emmanuelle Six,Delphine Bonhomme,Delphine Bonhomme,Marta Monteiro,Marta Monteiro,Kheira Beldjord,Kheira Beldjord,Monika Jurkowska,Corinne Cordier-Garcia,Alexandrine Garrigue,Alexandrine Garrigue,Liliane Dal Cortivo,Benedita Rocha,Benedita Rocha,Alain Fischer,Alain Fischer,Alain Fischer,Marina Cavazzana-Calvo,Marina Cavazzana-Calvo,Isabelle André-Schmutz,Isabelle André-Schmutz +21 more
TL;DR: It is demonstrated that these Lin−CD34+CD10+CD24− progenitors have a very low myeloid potential but can generate B, T, and natural killer lymphocytes and coexpress recombination activating gene 1, terminal deoxynucleotide transferase, PAX5, interleukin 7 receptor α, and CD3ε.
