Therapeutic effect of JAK1/2 blockade on the manifestations of hemophagocytic lymphohistiocytosis in mice

TL;DR: It is demonstrated that clinically relevant doses of the JAK1/2 inhibitor ruxolitinib suppresses the harmful consequences of macrophage overactivation characterizing HLH in 2 murine models and could be readily translated into the clinic for the treatment of primary, and perhaps even secondary, forms of HLH.

TL;DR: G germline loss-of-function mutations in HAVCR2, which encodes the immune modulator TIM-3, are found in individuals with subcutaneous panniculitis-like T cell lymphomas and hemophagocytic lymphohistiocytosis, a life-threatening inflammatory condition.
