Global Epigenomic Reconfiguration During Mammalian Brain Development
Ryan Lister,Ryan Lister,Eran A. Mukamel,Joseph R. Nery,Mark A. Urich,Clare A. Puddifoot,Nicholas D. Johnson,Jacinta Lucero,Yun Huang,Andrew J. Dwork,Matthew D. Schultz,Matthew D. Schultz,Miao Yu,Julian Tonti-Filippini,Holger Heyn,Shijun Hu,Joseph C. Wu,Anjana Rao,Manel Esteller,Chuan He,Fatemeh Haghighi,Terrence J. Sejnowski,Terrence J. Sejnowski,M. Margarita Behrens,Joseph R. Ecker +24 more
TL;DR: The results extend the knowledge of the unique role of DNA methylation in brain development and function, and offer a new framework for testing the role of the epigenome in healthy function and in pathological disruptions of neural circuits.
Journal ArticleDOI
Large-scale structure of genomic methylation patterns
Robert A. Rollins,Fatemeh Haghighi,John R. Edwards,Rajdeep Das,Michael Q. Zhang,Jingyue Ju,Timothy H. Bestor +6 more
TL;DR: The enrichment of Regulatory sequences in the relatively small unmethylated compartment suggests that cytosine methylation constrains the effective size of the genome through the selective exposure of regulatory sequences.
Journal ArticleDOI
Chromatin and sequence features that define the fine and gross structure of genomic methylation patterns
John R. Edwards,Anne H. O'Donnell,Robert A. Rollins,Heather E. Peckham,Clarence Lee,Maria H. Milekic,Benjamin Chanrion,Yutao Fu,Tao Su,Hanina Hibshoosh,Jay A. Gingrich,Fatemeh Haghighi,Robert C. Nutter,Timothy H. Bestor +13 more
TL;DR: It is concluded that methylation is the default state of most CpG dinucleotides in the mammalian genome and that a combination of local dinucleotide frequencies, the interaction of repeated sequences, and the presence or absence of histone variants or modifications shields a population of C pG sites from DNA methyltransferases that lack intrinsic sequence specificity.
Journal ArticleDOI
Age-related sperm DNA methylation changes are transmitted to offspring and associated with abnormal behavior and dysregulated gene expression
Maria H. Milekic,Yurong Xin,Anne H. O'Donnell,Kevin K. Kumar,M Bradley-Moore,Dolores Malaspina,Holly Moore,Daniela Brunner,Yongchao Ge,John R. Edwards,Stéphane Paul,Fatemeh Haghighi,Jay A. Gingrich +12 more
TL;DR: It is demonstrated that DNA methylation abnormalities arising in the sperm of old fathers are a plausible mechanism to explain some of the risks that APA poses to resulting offspring.
Journal ArticleDOI
Epigenetic regulation of serotonin transporter expression and behavior in infant rhesus macaques.
Erin L. Kinnally,John P. Capitanio,Rudolph Leibel,Liyong Deng,Charles A. LeDuc,Fatemeh Haghighi,J. John Mann +6 more
TL;DR: Higher 5‐HTT CpG methylation, but not rh5‐HTTLPR genotype, exacerbated the effects of early life stress on behavioral stress reactivity in infants, which may contribute to the risk for mood disorders observed in ‘high‐risk’5‐ HTTLPR carriers.
