Journal ArticleDOI
MET overexpression turns human primary osteoblasts into osteosarcomas.
Salvatore Patanè,Sofia Avnet,Nadia Coltella,Barbara Costa,Simone Sponza,Martina Olivero,Elisa Vigna,Luigi Naldini,Nicola Baldini,Riccardo Ferracini,Simona Corso,Silvia Giordano,Paolo M. Comoglio,Maria Flavia Di Renzo +13 more
TL;DR: Experimental evidence is provided that overexpression of the MET oncogene causes and sustains the full-blown transformation of osteoblasts and that it is essential for the maintenance of the cancer phenotype.
Journal ArticleDOI
CD4+ T Cells from IPEX Patients Convert into Functional and Stable Regulatory T Cells by FOXP3 Gene Transfer
Laura Passerini,Eva Rossi Mel,Claudia Sartirana,Georgia Fousteri,Attilio Bondanza,Luigi Naldini,Maria Grazia Roncarolo,Rosa Bacchetta +7 more
TL;DR: It is shown that when CD4+ T cells are converted into Treg cells after lentivirus-mediated FOXP3 gene transfer, the resulting CD4FOXP3 T cell population displays stable phenotype and suppressive function, especially when naïve T cells is converted.
Journal ArticleDOI
Liver-Directed Lentiviral Gene Therapy in a Dog Model of Hemophilia B
Alessio Cantore,Marco Ranzani,Cynthia C. Bartholomae,Monica Volpin,Patrizia Della Valle,Francesca Sanvito,Lucia Sergi Sergi,Pierangela Gallina,Fabrizio Benedicenti,Dwight A. Bellinger,Robin A. Raymer,Elizabeth P. Merricks,Francesca Bellintani,Samia Martin,Claudio Doglioni,Armando D'Angelo,Thierry VandenDriessche,Marinee Chuah,Marinee Chuah,Manfred Schmidt,Timothy C. Nichols,Eugenio Montini,Luigi Naldini +22 more
TL;DR: It is shown that gene therapy using lentiviral vectors targeting the expression of a canine factor IX transgene in hepatocytes was well tolerated and provided a stable long-term production of coagulation factor IX in dogs with hemophilia B.
Journal ArticleDOI
Lentiviral Vector-Mediated Gene Transfer in T Cells from Wiskott–Aldrich Syndrome Patients Leads to Functional Correction
Logic Dupre,Sara Trifari,Antonia Follenzi,Francesco Marangoni,Teresa Lain de Lera,Antonio Bernad,Silvana Martino,Shigeru Tsuchiya,Claudio Bordignon,Luigi Naldini,Luigi Naldini,Alessandro Aiuti,Maria Grazia Roncarolo +12 more
TL;DR: The observation that lentiviral vector-mediated gene transfer results in correction of T cell defects in vitro supports their application for gene therapy in WAS patients.
Journal ArticleDOI
CRISPR germline engineering—the community speaks
Katrine Bosley,Michael R. Botchan,Annelien L. Bredenoord,Dana Carroll,R. Alta Charo,Emmanuelle Charpentier,Ronald A. Cohen,Jacob E. Corn,Jennifer A. Doudna,Guoping Feng,Henry T. Greely,Rosario Isasi,Weihzi Ji,Jin-Soo Kim,Bartha Maria Knoppers,Edward Lanphier,Jinsong Li,Robin Lovell-Badge,G. Steven Martin,Jonathan D. Moreno,Luigi Naldini,Martin F. Pera,Anthony C.F. Perry,J. Craig Venter,Feng Zhang,Qi Zhou +25 more
TL;DR: Nature Biotechnology asks selected members of the international community to comment on the ethical issues raised by the prospect of CRISPR-Cas9 engineering of the human germline.
