TL;DR: This paper performed a meta-analysis of >15 million genetic variants in 21,399 cases and 95,464 controls from populations of European, African, Japanese and Latino ancestry, followed by replication in 32,059 cases and 228,628 controls from 18 studies.

Atopic dermatitis increases the effect of exposure to peanut antigen in dust on peanut sensitization and likely peanut allergy
Helen A. Brough,Andrew H. Liu,Scott H. Sicherer,Kerry Makinson,Abdel Douiri,Sara J. Brown,Alick Stephens,W.H. Irwin McLean,Victor Turcanu,Robert A. Wood,Stacie M. Jones,Wesley Burks,Peter Dawson,Donald Stablein,Hugh A. Sampson,Gideon Lack +15 more
TL;DR: In this article, an exposure-response relationship between exposure to peanut antigen in household dust and peanut skin prick test (SPT) sensitization and likely allergy was found, and an increase in 4 log 2 EPE units increased the odds of peanut SPT sensitization (1.71-fold, 95% CI, 1.13-to 2.59-fold; P ǫ =.01) and likely peanut allergy (PA; 2.10-fold), and 95% à 1.20-to 3.67-fold à Ã
Journal ArticleDOI
Skin microbiome before development of atopic dermatitis: Early colonization with commensal staphylococci at 2 months is associated with a lower risk of atopic dermatitis at 1 year.
Elizabeth A. Kennedy,Jennifer Connolly,Jonathan O'b Hourihane,Padraic G. Fallon,Padraic G. Fallon,W.H. Irwin McLean,Deirdre M. Murray,Jay-Hyun Jo,Julia A. Segre,Heidi H. Kong,Alan D. Irvine,Alan D. Irvine +11 more
TL;DR: This study suggests that 12‐month‐old infants with AD were not colonized with S aureus before having AD, and commensal staphylococci were significantly less abundant in infants affected at month 12, suggesting that this genus might protect against the later development of AD.
Journal ArticleDOI
Skin barrier dysfunction measured by transepidermal water loss at 2 days and 2 months predates and predicts atopic dermatitis at 1 year
Maeve M. Kelleher,Audrey DunnGalvin,Jonathan O'b Hourihane,Deirdre M. Murray,Linda E. Campbell,W.H. Irwin McLean,Alan D. Irvine,Alan D. Irvine +7 more
TL;DR: Impairment of skin barrier function at birth and at 2 months precedes clinical AD, and these findings have implications for the optimal timing of interventions for the prevention of AD.
Journal ArticleDOI
Genome-wide Comparative Analysis of Atopic Dermatitis and Psoriasis Gives Insight into Opposing Genetic Mechanisms

Elder,James T. Elder,Stephan Weidinger,Heather J. Cordell,Sara J. Brown +40 more
TL;DR: The results show that atopic dermatitis and psoriasis have distinct genetic mechanisms with opposing effects in shared pathways influencing epidermal differentiation and immune response.
