Abstract: A cAMP dependent protein kinase (PKA) was identified in the dinoflagellate Amphidinium operculum. In vitro kinase activity towards kemptide, a PKA-specific substrate, was not detectable in crude lysates. However, fractionation of dinoflagellate extracts by gel filtration chromatography showed PKA-like activity toward kemptide at approximately 66 kDa. These findings suggest that possible low molecular mass inhibitors in crude lysates were removed by the gel filtration chromatography. Pre-incubation of extracts with cAMP prior to chromatography resulted in an apparent molecular mass shift in the in vitro kinase assay to 40 kDa. An in-gel kinase assay reflected activity of the free catalytic subunit at approximately 40 kDa. Furthermore, western blotting with an antibody to the human PKA catalytic subunit confirmed a catalytic subunit with a mass of approximately 40 kDa. Results from this study indicate that the PKA in A. operculatum has a catalytic subunit of similar size to that in higher eukaryotes, but with a holoenzyme of a size suggesting a dimeric, rather than tetrameric structure.

Abstract: Optimizing complex displays is difficult because there are many alternative ways of mapping the data to its graphical representation. In this paper we report on a study employing a method we call “interactive design space hill climbing”. This involves first parameterizing the mapping from data to display. Next, using an interactive interface, designers and domain experts attempt to construct good designs by interactively changing parameters settings based on a random starting point. In our study we applied this method to the problem of 2D flow visualization: users adjusted 22 different sliders under each of 11 mappings to try to create an optimal display of a flow field from an ocean flow model. The results suggest that some variables should have settings in a narrow range. We conclude that employing designers with a human in the loop hill climbing interface can be a good overall solution for complex visual display designs in cases where a relatively simple parameterization is possible.

Abstract: Coastal regions are vulnerable to storm surge and flooding due to tropical and extratropical storms. It is necessary to build robust resiliency of the coastal communities to these hazards. The main objectives of operational surge and inundation forecast and coastal warning systems are to protect life and to sustain economic prosperity. The National Oceanic and Atmospheric Administration of the United States has initiated an integrated effort through pilot demonstration projects, and model-based ocean and coastal forecasting systems, to build improved operational warnings and forecasts capability for storm surge and inundation. This note describes the overall strategy and progress to date, with an emphasis on forecasting extratropical storm surge.
