The 2.9 A resolution crystal structure of Escherichia coli catabolite gene activator protein (CAP) complexed with cyclic AMP reveals two distinct structural domains separated by a cleft. The smaller carboxy-terminal domain is presumed to bind DNA while the amino-terminal domain is seen to bind cyclic AMP. Model building studies suggest that CAP binds to left-handed B-type DNA, contracting its major groove via two alpha-helices. It is possible that the CAP conversion of right- to left-handed DNA in a closed supercoil, is what activates transcription by RNA polymerase.read more

TL;DR: The three-dimensional structure of 3-(adenin-9-yl)propionhistamide hydrochloride, as a model of interactions between the protonated imidazolyl group and adenine moiety, has been determined by X-ray crystallography and shows that a role of the essential histidine in RNAase reaction is a switch between capture and ejection of the substrate, which depends upon proton uptake and its release, respectively.

TL;DR: It is shown that when the lacP class II promoter mutation L305 was transferred on an F-prime into P. mirabilis, its effect on lac expression was much as in E. coli, and when Flac bearing the IQ1 up-promoter mutation was introduced into P., the basal level decreased by three orders of magnitude, in accord with poor expression of lacI+ being responsible for the usual low induction ratio.
