The functional roles of mammalian DNA polymerase.

TLDR
A full understanding of the function of the eucaryotic DNA polymerases now depends on the clarification of the intimate mechanisms of DNA synthesis in various modes as discussed by the authors, and identification of the accessory proteins, cofactors, and replicative DNA structures within a given replication complex, as has been accomplished in certain procaryotes (53), may help in this task.

Use of a low-speed, iso-density percoll centrifugation method to increase the viability of isolated rat hepatocyte preparations.

TL;DR: The enriched parenchymal cells have been shown to be superior to untreated cells by a number of criteria including: preparation homogeneity, cell morphology, maintenance of cytochrome P-450, hormonal responsiveness, and the DNA repair capability after treatment with benzo[a]pyrene or 2-acetylaminofluorene.
Journal ArticleDOI

DNA polymerase γ, the mitochondrial replicase

TL;DR: The two-subunit holoenzyme is an efficient and processive polymerase, which exhibits high fidelity in nucleotide selection and incorporation while proofreading errors with its intrinsic 3′ → 5′ exonuclease.

The mutational specificity of DNA polymerase-beta during in vitro DNA synthesis. Production of frameshift, base substitution, and deletion mutations.

TL;DR: The frequency and specificity of mutations produced in vitro by eucaryotic DNA polymerase-beta have been determined in a forward mutation assay using a 250-base target sequence in M13mp2 DNA, indicating that purified DNA polymerases alone are capable of producing deletions.

The Translesion DNA Polymerase ζ Plays a Major Role in Ig and bcl-6 Somatic Hypermutation

TL;DR: Inhibition of the pol zeta REV3 catalytic subunit by specific phosphorothioate-modified oligonucleotides impaired Ig and bcl-6 hypermutation and UV damage-induced DNA mutagenesis, without affecting cell cycle or viability.
Journal ArticleDOI

The mutational specificity of DNA polymerases-alpha and -gamma during in vitro DNA synthesis.

TL;DR: A correlation between the accuracy and processivity of DNA polymerization and the most accurate DNA polymerase also incorporates the most nucleotides per association with the primer-template, while the least accurate enzyme (pol-beta) is the least processive is demonstrated.

On the mechanism of DNA replication in mammalian chromosomes

TL;DR: It is found that the long fibers of which chromosomal DNA is composed are made up of many tandemly joined sections in each of which DNA is replicated at a fork-like growing point.

DNA polymerase associated with human hepatitis B antigen.

TL;DR: DNA polymerase activity was detected in each of eight preparations of concentrated human hepatitis B antigen (HBAg) rich in Dane particles prepared by high-speed centrifugation of antigen-positive human plasma and in none of seven control preparations prepared in the same way from HBAg-negative plasma.

Benzo(a)pyrene diol epoxides as intermediates in nucleic acid binding in vitro and in vivo.

TL;DR: Evidence has been obtained that a specific isomer of a diol epoxide derivative of benzo(a)pyrene, (+/-)-7 beta,8alpha-dihydroxy-9alpha, 10alpha-epoxy-7,8,9,10-tetrahydrobenzo( a)pyene, is an intermediate in the binding of benzos(a).pyrene to RNA in cultured bovine bronchial mucosa.
