TLDR
In the past decade, it is has become clear that Membrane Contact Sites (MCSs) have a much broader range of critical roles in cells than was initially thought as mentioned in this paper, and functions for MCSs in intracellular signalling such as autophagy, lipid metabolism, membrane dynamics, cellular stress responses and organelle trafficking and biogenesis have been reported.Abstract:
Organelles compartmentalize eukaryotic cells, enhancing their ability to respond to environmental and developmental changes. One way in which organelles communicate and integrate their activities is by forming close contacts, often called 'membrane contact sites' (MCSs). Interest in MCSs has grown dramatically in the past decade as it is has become clear that they are ubiquitous and have a much broader range of critical roles in cells than was initially thought. Indeed, functions for MCSs in intracellular signalling (particularly calcium signalling, reactive oxygen species signalling and lipid signalling), autophagy, lipid metabolism, membrane dynamics, cellular stress responses and organelle trafficking and biogenesis have now been reported.read more

Quality control of the mitochondrial proteome.
TL;DR: To ensure proper mitochondrial function, cells use multiple mechanisms of quality control that survey mitochondrial protein biogenesis, import and folding, and allow mitochondria to adapt to the changing needs as well as to respond to stresses that compromise proteostasis.

LRRK2 mediates tubulation and vesicle sorting from lysosomes
Luis Bonet-Ponce,Alexandra Beilina,Chad D. Williamson,Eric Lindberg,Jillian H. Kluss,Sara Saez-Atienzar,Natalie Landeck,Ravindran Kumaran,Adamantios Mamais,Christopher K. E. Bleck,Yan Li,Mark R. Cookson +11 more
TL;DR: A new process orchestrated by LRRK2 is described, which is named LYTL (LYsosomal Tubulation/sorting driven by L RRK2), by which lysosomal tubulation is used to release vesicles from lysOSomes.

Oxidative eustress: On constant alert for redox homeostasis.
TL;DR: In the open metabolic system, redox-related signaling requires continuous monitoring and fine-tuning of the steady-state redox set point, which operates within a physiological range that has been called the "Homeodynamic Space", the "Goldilocks Zone" or the "Golden Mean".

Getting around the cell: physical transport in the intracellular world.
TL;DR: The interplay of diffusion, molecular motors, and flows supports the intracellular transport needs that underlie a broad variety of biological phenomena.

FFAT motif phosphorylation controls formation and lipid transfer function of inter-organelle contacts.
Thomas Di Mattia,Arthur Martinet,Souade Ikhlef,Alastair G. McEwen,Yves Nominé,Corinne Wendling,Pierre Poussin-Courmontagne,Laetitia Voilquin,Pascal Eberling,Frank Ruffenach,Jean Cavarelli,John A Slee,Tim P. Levine,Guillaume Drin,Catherine Tomasetto,Fabien Alpy +15 more
TL;DR: This study reveals that phosphorylation acts as a general switch for inter‐organelle contacts and produces a novel prediction algorithm, which expands the repertoire of candidate proteins with a Phospho‐FFAT that are able to create membrane contact sites.

Inositol trisphosphate, a novel second messenger in cellular signal transduction.
TL;DR: Diacylglycerol operates within the plane of the membrane to activate protein kinase C, whereas inositol trisphosphate is released into the cytoplasm to function as a second messenger for mobilizing intracellular calcium.

Regulation of cell death: the calcium-apoptosis link.
TL;DR: The dual role of Ca2+ in living organisms is discussed in this paper, where it has been shown that cellular Ca 2+ overload, or perturbation of intracellular Ca2 + compartmentalization, can cause cytotoxicity and trigger either apoptotic or necrotic cell death.

Triglyceride accumulation protects against fatty acid-induced lipotoxicity
Laura L. Listenberger,Xianlin Han,Sarah Lewis,Sylvaine Cases,Robert V. Farese,Daniel S. Ory,Jean E. Schaffer +6 more
TL;DR: This work demonstrates in cultured cells that the relative toxicity of two common dietary long chain fatty acids is related to channeling of these lipids to distinct cellular metabolic fates, and supports a model of cellular lipid metabolism in which unsaturated fatty acids serve a protective function against lipotoxicity though promotion of triglyceride accumulation.

ER Tubules Mark Sites of Mitochondrial Division
Jonathan R. Friedman,Laura L. Lackner,Matthew West,Jared R. DiBenedetto,Jodi Nunnari,Gia K. Voeltz +5 more
TL;DR: It was found that mitochondrial division occurred at positions where ER tubules contacted mitochondria and mediated constriction before Drp1 recruitment, suggesting that ERtubules may play an active role in defining the position of mitochondrial division sites.

BAX and BAK Regulation of Endoplasmic Reticulum Ca2+: A Control Point for Apoptosis
Luca Scorrano,Scott A. Oakes,Joseph T. Opferman,Emily H. Cheng,Mia D. Sorcinelli,Tullio Pozzan,Stanley J. Korsmeyer +6 more
TL;DR: BAX and BAK operate in both the ER and mitochondria as an essential gateway for selected apoptotic signals, and targeting of BAX to mitochondria selectively restored apoptosis to “BH3-only” signals.
