TL;DR: Variants associated with cholesterol metabolism and type 1 diabetes showed similar phenomena, indicating that large-scale eQTL mapping provides insight into the downstream effects of many trait-associated variants.
Journal ArticleDOI
Innate immune activity conditions the effect of regulatory variants upon monocyte gene expression.
Benjamin P. Fairfax,Peter Humburg,Seiko Makino,Vivek Naranbhai,Daniel Wong,Evelyn Lau,Luke Jostins,Katharine Plant,Robert Andrews,Chris J. McGee,Julian C. Knight +10 more
TL;DR: This work mapped interindividual variation in gene expression as a quantitative trait, defining expression quantitative trait loci (eQTLs) and found trans associations to the major histocompatibility complex are dependent on context, paralleling the expression of class II genes.
Journal ArticleDOI
Genetic association analyses implicate aberrant regulation of innate and adaptive immunity genes in the pathogenesis of systemic lupus erythematosus
James Bentham,David L. Morris,Deborah S. Cunninghame Graham,Christopher L. Pinder,Philip Tombleson,Timothy W. Behrens,Javier Martin,Benjamin P. Fairfax,Julian C. Knight,Lingyan Chen,Joseph M. TL;DR: This study comprised 7,219 cases and 15,991 controls of European ancestry, constituting a new genome-wide association study, a meta-analysis with a published GWAS and a replication study, which mapped 43 susceptibility loci, including ten new associations.

TL;DR: It is observed that cis-eQTLs can be detected for 88% of the studied genes, but that they have a different genetic architecture compared to disease-associated variants, limiting the ability to use cis- eZTLs to pinpoint causal genes within susceptibility loci.
Journal ArticleDOI
Genetics of gene expression in primary immune cells identifies cell type–specific master regulators and roles of HLA alleles
Benjamin P. Fairfax,Seiko Makino,Jayachandran Radhakrishnan,Katharine Plant,Stephen Leslie,Alexander T. Dilthey,Peter R. Ellis,Cordelia Langford,Fredrik O. Vannberg,Fredrik O. Vannberg,Julian C. Knight +10 more
TL;DR: It is shown that mapping gene expression in defined primary cell populations identifies new cell type–specific trans-regulated networks and provides insights into the genetic basis of disease susceptibility.
