TL;DR: Using genome-wide association data from three studies and targeted replication association analyses in up to 18,554 independent participants, it is shown that common SNPs at 18 loci are reproducibly associated with concentrations of low-density cholesterol, high-density lipoprotein (HDL) cholesterol, and/or triglycerides.

Caulfield,Alberto Zanchetti,Anna F. Dominiczak +82 more
TL;DR: The newly discovered UMOD locus for hypertension has the potential to give new insights into the role of uromodulin in BP regulation and to identify novel drugable targets for reducing cardiovascular risk.
Journal ArticleDOI
Common Missense Variant in the Glucokinase Regulatory Protein Gene Is Associated With Increased Plasma Triglyceride and C-Reactive Protein but Lower Fasting Glucose Concentrations
Marju Orho-Melander,Olle Melander,Candace Guiducci,Pablo Perez-Martinez,Pablo Perez-Martinez,Dolores Corella,Charlotta Roos,Ryan Tewhey,Mark J. Rieder,Jennifer L. Hall,Gonçalo R. Abecasis,E. Ordovas,Sekar Kathiresan +42 more
TL;DR: These findings point to a molecular mechanism in humans by which higher triglycerides and CRP can be coupled with lower plasma glucose concentrations and position GCKR in central pathways regulating both hepatic triglyceride and glucose metabolism.

TL;DR: An analysis of combined discovery and follow-up data identified SNPs significantly associated with BP at p < 8.56 × 10(-7) at four further loci and highlighted the utility of studying SNPs and samples that are independent of those studied previously even when the sample size is smaller than that in previous studies.
Journal ArticleDOI
Plasma lipid composition and risk of developing cardiovascular disease.
Céline Fernandez,Marianne Sandin,Julio L. Sampaio,Peter Almgren,Krzysztof Narkiewicz,Michal Hoffmann,Thomas Hedner,Björn Wahlstrand,Kai Simons,Andrej Shevchenko,Peter James,Olle Melander +11 more
TL;DR: This study suggests that CVD development is preceded by reduced levels of LPC16∶0, LPC20∶4 and some specific TAG species and by increased levels of SM38∶2 and indicates that certain lipid species are intermediate phenotypes between genetic susceptibility and overt CVD.
