Serwas,Hiroyuki Yoda,Alan Ashworth,Sourav Bandyopadhyay,Trevor Bivona,Gabriel Eades,Stefan Oberlin,Neil Tay,Yuhao Wang,Jonathan S. Weissman +124 more
TL;DR: A framework is described for this expanded list of cancer targets, providing novel opportunities for clinical translation and indicating that the diversity of therapeutic targets engendered by non-oncogene dependencies is much larger than the list of recurrently mutated genes.
Journal ArticleDOI
Synthesis of (±)-Actinophyllic Acid and Analogs: Applications of Cascade Reactions and Diverted Total Synthesis
Brett A. Granger,Ivan T. Jewett,Ivan T. Jewett,Jeffrey D. Butler,Bruce K. Hua,Claire E. Knezevic,Elizabeth I. Parkinson,Paul J. Hergenrother,Stephen F. Martin +8 more
TL;DR: The synthesis features a novel cascade of reactions of N-stabilized carbocations with π-nucleophiles to create the tetracyclic core of actinophyllic acid in a single chemical operation, and one key intermediate was modified to furnish several novel compounds having potentially promising anticancer activity.
Journal ArticleDOI
Synergistic Effects of Stereochemistry and Appendages on the Performance Diversity of a Collection of Synthetic Compounds
Bruno Melillo,Bruno Melillo,Jochen Zoller,Jochen Zoller,Bruce K. Hua,Bruce K. Hua,Oscar Verho,Oscar Verho,Jannik C. Borghs,Jannik C. Borghs,Jannik C. Borghs,Shawn D. Nelson,Shawn D. Nelson,Micah Maetani,Micah Maetani,Mathias Wawer,Paul A. Clemons,Stuart L. Schreiber,Stuart L. Schreiber,Stuart L. Schreiber +19 more
TL;DR: It is shown that stereoisomerism and appendage diversification can produce effects of similar magnitude, and that the concurrent use of these strategies results in a broader sampling of biological activity.
Journal ArticleDOI
Real-Time Biological Annotation of Synthetic Compounds
Christopher J. Gerry,Bruce K. Hua,Mathias J. Wawer,Jonathan P. Knowles,Shawn D. Nelson,Oscar Verho,Sivaraman Dandapani,Bridget K. Wagner,Paul A. Clemons,Kevin I. Booker-Milburn,Zarko V. Boskovic,Stuart L. Schreiber +11 more
TL;DR: The use of high-dimensional “multiplex” assays, which are capable of measuring thousands of cellular features in one experiment, are proposed to annotate rapidly and inexpensively the biological activities of newly synthesized compounds, indicating that real-time biological annotation can inform optimization efforts and library syntheses by illuminating trends relating to biological activity that would be difficult to predict if only chemical structure were considered.
Journal ArticleDOI
Nuisance compounds in cellular assays
Jayme L. Dahlin,Douglas S. Auld,Ina Rothenaigner,Steve Haney,Jonathan Z. Sexton,J. Willem M. Nissink,Jarrod Walsh,Jonathan A. Lee,John Strelow,Francis S. Willard,Lori Ferrins,Jonathan B. Baell,Jonathan B. Baell,Michael A. Walters,Bruce K. Hua,Bruce K. Hua,Kamyar Hadian,Bridget K. Wagner +17 more
TL;DR: In this paper, the authors summarize the collective academic, government, and industry experiences regarding cellular nuisance compounds in more complex cell-based assays and suggest best practices to efficiently address these compounds in complex biological settings.
