Journal ArticleDOI
Immune dysregulation in human subjects with heterozygous germline mutations in CTLA4
Hye Sun Kuehn,Weiming Ouyang,Bernice Lo,Elissa K. Deenick,Elissa K. Deenick,Julie E. Niemela,Danielle T. Avery,Jean Nicolas Schickel,Dat Q. Tran,Jennifer Stoddard,Yu Zhang,David M. Frucht,Bogdan Dumitriu,Phillip Scheinberg,Les R. Folio,Cathleen Frein,Susan Price,Christopher Koh,Theo Heller,Christine M. Seroogy,Anna Huttenlocher,V. Koneti Rao,Helen C. Su,David E. Kleiner,Luigi D. Notarangelo,Yajesh Rampertaap,Kenneth N. Olivier,Joshua J McElwee,Jason D. Hughes,Stefania Pittaluga,Joao Bosco Oliveira,Eric Meffre,Thomas A. Fleisher,Steven M. Holland,Michael J. Lenardo,Stuart G. Tangye,Stuart G. Tangye,Gulbu Uzel +37 more
TL;DR: The findings support the idea that CTLA4 tells the immune system when enough is enough, and Inherited human CTLA 4 haploinsufficiency demonstrates a critical quantitative role for CTLA-4 in governing T and B lymphocyte homeostasis.
Journal ArticleDOI
Dominant-activating germline mutations in the gene encoding the PI(3)K catalytic subunit p110δ result in T cell senescence and human immunodeficiency
Carrie L. Lucas,Hye Sun Kuehn,Fang Zhao,Julie E. Niemela,Elissa K. Deenick,Umaimainthan Palendira,Danielle T. Avery,Leen Moens,Jennifer L. Cannons,Matthew Biancalana,Jennifer Stoddard,Weiming Ouyang,David M. Frucht,V. Koneti Rao,T. Prescott Atkinson,Anahita Agharahimi,Ashleigh A. Hussey,Les R. Folio,Kenneth N. Olivier,Thomas A. Fleisher,Stefania Pittaluga,Steven M. Holland,Jeffrey I. Cohen,Joao Bosco Oliveira,Stuart G. Tangye,Pamela L. Schwartzberg,Michael J. Lenardo,Gulbu Uzel +27 more
TL;DR: T cells from patients with heterozygous for three different germline, gain-of-function mutations in PIK3CD exhibited increased phosphorylation of the kinase Akt and hyperactivation of the metabolic checkpoint kinase mTOR, enhanced glucose uptake and terminal effector differentiation in vitro.
Journal ArticleDOI
BAFF selectively enhances the survival of plasmablasts generated from human memory B cells
Danielle T. Avery,Susan L. Kalled,Julia I. Ellyard,Christine Ambrose,Sarah A. Bixler,Marilyn Thien,Robert Brink,Fabienne Mackay,Philip D. Hodgkin,Stuart G. Tangye +9 more
TL;DR: BAFF may enhance humoral immunity in vivo by promoting survival of ISCs via a BCMA-dependent mechanism and have wide-ranging implications for the treatment of human immunodeficiencies as well as autoimmune diseases.
Journal ArticleDOI
Cytokine-Mediated Regulation of Human B Cell Differentiation into Ig-Secreting Cells: Predominant Role of IL-21 Produced by CXCR5+ T Follicular Helper Cells
Vanessa L. Bryant,Vanessa L. Bryant,Vanessa L. Bryant,Cindy S. Ma,Cindy S. Ma,Danielle T. Avery,Danielle T. Avery,Ying Li,Kim L. Good,Kim L. Good,Kim L. Good,Lynn M. Corcoran,Rene de Waal Malefyt,Stuart G. Tangye,Stuart G. Tangye,Stuart G. Tangye +15 more
TL;DR: The findings suggest that IL-21 produced by TFH cells during the primary as well as the subsequent responses to T cell-dependent Ag makes a major contribution to eliciting and maintaining long-lived humoral immunity.
Journal ArticleDOI
B cell-intrinsic signaling through IL-21 receptor and STAT3 is required for establishing long-lived antibody responses in humans
Danielle T. Avery,Elissa K. Deenick,Elissa K. Deenick,Cindy S. Ma,Cindy S. Ma,Santi Suryani,Santi Suryani,Nicholas Simpson,Gary Y. Chew,Tyani D. Chan,Tyani D. Chan,Umamainthan Palendira,Umamainthan Palendira,Jacinta Bustamante,Stéphanie Boisson-Dupuis,Stéphanie Boisson-Dupuis,Sharon Choo,Karl Bleasel,Jane Peake,Cecile King,Cecile King,Martyn A. French,Dan Engelhard,Sami Al-Hajjar,Saleh Al-Muhsen,Klaus Magdorf,Joachim Roesler,Peter D. Arkwright,Pravin Hissaria,D. Sean Riminton,Melanie Wong,Robert Brink,Robert Brink,Robert Brink,David A. Fulcher,Jean-Laurent Casanova,Jean-Laurent Casanova,Matthew C. Cook,Stuart G. Tangye,Stuart G. Tangye +39 more
TL;DR: STAT3-activating cytokines such as IL-21 underpin Ag-specific humoral immune responses and provide a mechanism for the functional antibody deficit in STAT3-deficient patients.
