Modeling HNF1B-associated monogenic diabetes using human iPSCs reveals an early stage impairment of the pancreatic developmental program.
Ranna El-Khairi,Evelyn Olszanowski,Evelyn Olszanowski,Daniele Muraro,Daniele Muraro,Pedro Madrigal,Pedro Madrigal,Katarzyna Tilgner,Mariya Chhatriwala,Mariya Chhatriwala,Sapna Vyas,Crystal Y. Chia,Crystal Y. Chia,Ludovic Vallier,Ludovic Vallier,Santiago A. Rodríguez-Seguí,Santiago A. Rodríguez-Seguí +16 more
TL;DR: In this paper, the authors used a well-controlled human induced pluripotent stem cell pancreatic differentiation model to elucidate the molecular mechanisms underlying HNF1B-associated diabetes.

Limited oxygen availability in standard cell culture alters metabolism and function in terminally-differentiated cells
Joyce Yin Hsing Tan,Sam Virtue,Dougall M. Norris,Olivia J. Conway,Ming Yang,Christopher Gribben,Fatima Lugtu,Ioannis Kamzolas,James R. Krycer,Richard J. Mills,Conceição Pereira,Martin Dale,Amber S. Fisher-Wellman,Christian Frezza,Antonio Vidal-Puig,Daniel J. Fazakerley +26 more
TL;DR: In this paper , the importance of cellular oxygen consumption is often underappreciated, with rates of oxygen consumption often sufficient to cause hypoxia at cell monolayers, and the functional benefits of increasing pericellular oxygen were transferable to other cellular systems including hPSC-derived hepatocytes and cardiac organoids.
Journal ArticleDOI
A novel differentiation system to produce hepatocytes for disease modelling and drug screening
Patent
In vitro production of cholangiocytes
TL;DR: In this article, Foregut stem cells (FSCs) are cultured in a hepatic induction medium comprising bone morphogenetic protein (BMP) and a TGFβ signalling inhibitor to produce a population of hepatoblasts.
