Journal ArticleDOI
Transcription factor T-bet represses expression of the inhibitory receptor PD-1 and sustains virus-specific CD8 + T cell responses during chronic infection
Charlly Kao,Kenneth J. Oestreich,Michael A. Paley,Alison Crawford,Jill M. Angelosanto,Mohammed Alkhatim A. Ali,Andrew M. Intlekofer,Jeremy M. Boss,Steven L. Reiner,Amy S. Weinmann,E. John Wherry +10 more
TL;DR: It is demonstrated that the transcription factor T-bet regulated the exhaustion of CD8+ T cells and the expression of inhibitory receptors and suggested therapeutic opportunities involving higher T-Bet expression during chronic infection.
Journal ArticleDOI
Cooperativity Between CD8+ T Cells, Non-Neutralizing Antibodies, and Alveolar Macrophages Is Important for Heterosubtypic Influenza Virus Immunity
Brian J. Laidlaw,Vilma Decman,Mohammed Alkhatim A. Ali,Michael C. Abt,Amaya I. Wolf,Laurel A. Monticelli,Krystyna Mozdzanowska,Jill M. Angelosanto,David Artis,Jan Erikson,E. John Wherry +10 more
TL;DR: Overall, the studies suggest that an influenza vaccine capable of eliciting both CD8+ T cells and antibodies specific for highly conserved influenza proteins may be able to provide heterosubtypic protection in humans, and act as the basis for a potential “universal” vaccine.
Journal ArticleDOI
Bystander chronic infection negatively impacts development of CD8(+) T cell memory
Erietta Stelekati,Haina Shin,Travis A. Doering,Douglas V. Dolfi,Carly G. K. Ziegler,Daniel P. Beiting,Lucas Dawson,Jennifer Liboon,David Wolski,Mohammed Alkhatim A. Ali,Peter D. Katsikis,Hao Shen,David S. Roos,W. Nicholas Haining,Georg M. Lauer,E. John Wherry +15 more
TL;DR: In this article, the authors investigated whether bystander chronic infections impact differentiation of memory CD8(+) T cells, the hallmark of protective immunity against intracellular pathogens, and found that exposure to prolonged bystander inflammation impairs the effector to memory transition.
Journal ArticleDOI
In vivo CD8+ T cell CRISPR screening reveals control by Fli1 in infection and cancer.
Zeyu Chen,Eri Arai,Omar Khan,Zhen Zhang,Shin Foong Ngiow,Yuan He,Hua Huang,Sasikanth Manne,Zhendong Cao,Amy E. Baxter,Zhangying Cai,Elizabeth Freilich,Mohammed Alkhatim A. Ali,Josephine R. Giles,Jennifer E. Wu,Allison R. Greenplate,Mohamed A. Hakeem,Qingzhou Chen,Makoto Kurachi,Kito Nzingha,Viktoriya Ekshyyan,Divij Mathew,Zhuoyu Wen,Nancy A. Speck,Alexis Battle,Shelley L. Berger,E. John Wherry,Junwei Shi +27 more
TL;DR: In this paper, the authors developed an in-vivo T-cell CRISPR screening platform and identified a key mechanism restraining TEFF biology through the ETS family TF, Fli1.
Journal ArticleDOI
Inhibitory signaling sustains a distinct early memory CD8 + T cell precursor that is resistant to DNA damage
Jonathan B. Johnnidis,Yuki Muroyama,Shin Foong Ngiow,Zeyu Chen,Sasikanth Manne,Zhangying Cai,Shufei Song,Jesse M. Platt,Jesse M. Platt,Jason M. Schenkel,Jason M. Schenkel,Mohamed S. Abdel-Hakeem,Jean Christophe Beltra,Allison R. Greenplate,Mohammed Alkhatim A. Ali,Kito Nzingha,Josephine R. Giles,Christelle Harly,John Attanasio,Kristen E. Pauken,Kristen E. Pauken,Bertram Bengsch,Bertram Bengsch,Michael A. Paley,Vesselin T. Tomov,Makoto Kurachi,Dario A. A. Vignali,Arlene H. Sharpe,Arlene H. Sharpe,Steven L. Reiner,Avinash Bhandoola,F. Bradley Johnson,E. John Wherry +32 more
TL;DR: In this paper, the authors identified a subset of virus-specific CD8+ T cells that possessed distinct characteristics including expression of CD62L, T cell factor 1 (TCF-1), and Eomesodermin; relative quiescence; expression of activation markers; and features of limited effector differentiation.
