Abstract: The present invention allows the screening of previously established genebanks or libraries by proxy for genes encoding secreted, partially secreted, or cell surface-displayed polypeptides of industrial interest, such as enzymes, receptors, cytokines, peptide hormones etc. that would not likely have been isolated using conventional screening assays. A method for isolating genes encoding secreted, partially secreted, or cell surface displayed polypeptides from existing gene libraries is described in which the endogenous secretion signal sequences are detected using an in vitro polynucleotide insertion reaction where the inserted polynucleotide comprises a promoter-less and secretion signal-less secretion reporter.

Abstract: Enzymatic bio-polishing offers a finish for pill prevention. The present study demonstrates that a cutinase can be used for bio-polishing of polyester fabrics and can be combined with a compatible cellulase to treat polyester and cotton blended fabrics. Two different cutinases were investigated with 100% polyester woven fabric. One cutinase and one cellulase were tested separately and in combination with 50%/50% polyester/cotton blend knit fabric. Following enzymatic treatment, weight loss, high-performance liquid chromatography (HPLC) analysis of the treatment liquor and pilling note were evaluated. An improvement in pilling note for both polyester and polyester blends was demonstrated. Additionally, HPLC analysis of the treatment liquors indicated polyester hydrolysis due to cutinase activity which correlated well with pilling note results

Abstract: Heteroblasty, the temporal development of the meristem, can produce diverse leaf shapes within a plant. Gevuina avellana, a tree from the South American temperate rainforest shows strong heteroblasty affecting leaf shape, transitioning from juvenile simple leaves to highly pinnate adult leaves. Light availability within the forest canopy also modulates its leaf size and complexity. Here we studied how the interaction between the light environment and the heteroblastic progression of leaves is coordinated in this species. We used RNA-seq on the Illumina platform to compare the range of transcriptional responses in leaf primordia of G. avellana at different heteroblastic stages and growing under different light environments. We found a steady up-regulation of SQUAMOSA PROMOTER BINDING PROTEIN LIKE (SPL), NAC, YUCCA and AGAMOUS-LIKE genes associated with increases in age, leaf complexity, and light availability. In contrast, expression of TCP, TPR and KNOTTED1 homeobox genes showed a sustained down-regulation. Additionally, genes involved in auxin synthesis/transport and jasmonate activity were differentially expressed, indicating an active regulation of processes controlled by these hormones. Our large-scale transcriptional analysis of the leaf primordia of G. avellana sheds light on the integration of internal and external cues during heteroblastic development in this species.

Abstract: An experimental investigation of the spray drying behavior of droplets containing excipients and carrier materials used in the pharmaceutical and biotechnological industries has been conducted. Specifically, rice starch suspensions with different amounts of TiO2, maltodextrin, dextrin, NaCl and Na2SO4 are dried. The drying rate is measured, and the morphology formation is mapped to obtain a more fundamental understanding of the drying process, which is very useful when designing product formulations. In the pilot spray dryer, droplet generation is based on the JetCutter technology and the droplets are dried under well-defined temperature and flow conditions. The droplets are sampled during drying to determine the drying rate, and the dried particles are collected for morphology analysis.
