TL;DR: In this article, the authors apply the Sabatier principle to an interfacial enzyme reaction and find that the results can be rationalized in so-called volcano plots that are emblematic of the principle.
Abstract: The Sabatier principle states that optimal catalysis occurs when interactions between catalyst and substrate are of intermediary strength. Although qualitative in nature, this concept has proven extremely useful within (nonbiochemical) heterogeneous catalysis. In the current work, we show that the principle may be applied to an interfacial enzyme reaction. Specifically, we studied the breakdown of cellulose by different cellulases (wild types and variants) and found that the results could be rationalized in so-called volcano plots that are emblematic of the principle. This implies that the rate of the complex enzymatic reaction can be described by a single parameter (binding strength), and we show how this may help elucidating e.g. rate-controlling steps and relationships of substrate load and enzymatic efficacy. On a more general level, we propose that the Sabatier principle may be widely applicable to interfacial enzyme processes and hence open an avenue to the application within biocatalysis of some of...

10 Mar 2004TL;DR: In this paper, a pre-treatment at an elevated temperature below the initial gelatinization temperature of said granular starch followed by simultaneous saccharification and fermentation, and optionally recovery of ethanol is described.
Abstract: The present invention relates to processes for production of an alcohol product from granular starch comprising a pre-treatment at an elevated temperature below the initial gelatinization temperature of said granular starch followed by simultaneous saccharification and fermentation, and optionally recovery of ethanol.

TL;DR: It is found that confinement-driven stabilization is not only dependent on protein size and cage radius, but also on the specific topology, and the choice of the confining potential is shown to have an effect on the observed stabilization and the scaling behavior of the stabilization with respect to the cage size.

26 Mar 2009TL;DR: Peptide aldehydes or ketone derivatives are particularly efficient for stabilizing subtilisin-type proteases in aqueous compositions such as liquid detergents, including peptide compounds with OH-substituted phenylalanine aldehyde as the C-terminal residue as mentioned in this paper.
