TLDR
The highest transformation efficiency was obtained with the disarmed hypervirulent strain AGL1 (Ti plasmid TiBo542), for which the percentage of explants forming kanamycin (Km)-resistant calli was double that obtained with each of the other three strains.Abstract:
The efficiency of Agrobacterium tumefaciens transformation of the model legume Medicago truncatula cv. Jemalong (genotype 2HA) was evaluated for strains LBA 4404, C58pMP90, C58pGV2260 and AGL1. Binary vectors carrying promoter-gus/gfp reporter gene fusions and the nptII gene as selectable marker were used for plant in vitro transformation/regeneration. The highest transformation efficiency was obtained with the disarmed hypervirulent strain AGL1 (Ti plasmid TiBo542), for which the percentage of explants forming kanamycin (Km)-resistant calli was double that obtained with each of the other three strains. In addition, we were able to reduce the time necessary for plant regeneration using AGL1, with 24% of the explants generating Km-resistant transgenic plantlets within only 4–5 months of culture. Transgene expression in planta was analysed and found to be conserved in the T1 descendents.read more

A remorin protein interacts with symbiotic receptors and regulates bacterial infection.
Benoit Lefebvre,Ton Timmers,Malick Mbengue,Sandra Moreau,Christine Hervé,Katalin Tóth,Joana Bittencourt-Silvestre,Dörte Klaus,Laurent Deslandes,Laurence Godiard,Jeremy D. Murray,Michael K. Udvardi,Sylvain Raffaele,Sébastien Mongrand,Julie V. Cullimore,Pascal Gamas,Andreas Niebel,Thomas Ott +17 more
TL;DR: It is shown that this oligomeric remorin protein attaches to the host plasma membrane surrounding the bacteria and controls infection and release of rhizobia into the host cytoplasm.

TL;DR: Transcription factors are DNA-binding proteins that interact with other transcriptional regulators, including chromatin remodeling/modifying proteins, to recruit or block access of RNA polymerases to the DNA template.

TL;DR: A nodule fate map was produced that showed, among other things, that intracellular release of rhizobia in primordium cells and meristem daughter cells are regulated in a different manner.

TL;DR: In vivo studies have shown that gaps within the bacterial file are a common feature during the early stages of IT development, and that segments of the file are able to slide collectively down the thread.

TL;DR: Findings indicate that M. truncatula and possibly other related legumes have complex mechanisms of gene regulation involving specific and common small RNAs operating post-transcriptionally.

The promoter of TL-DNA gene 5 controls the tissue-specific expression of chimaeric genes carried by a novel type of Agrobacterium binary vector
TL;DR: It was found that the gene 5 promoter is active in a tissue-specific fashion whereas this is not the case for the NOS promoter, providing the first documented instance of a gene derived from a procaryotic host the expression of which is apparently regulated by plant growth factors.

A binary plant vector strategy based on separation of vir- and T-region of the Agrobacterium tumefaciens Ti-plasmid
TL;DR: The interaction of two compatible plasmids, one containing the vir-region, the other carrying the T-DNA on a wide host-range replicon are reported, which allows introduction of the manipulated T- DNA into plant cells.
