Targeted Therapies for Hepatocellular Carcinoma

TLDR
Identification of oncogenes that mediate tumor progression, and trials that monitor their products as biomarkers, might lead to personalized therapy; reagents that interfere with signaling pathways required for HCC progression might be used to treat selected populations, and thereby maximize the efficacy and cost benefit.

Overexpression of Dickkopf-1 predicts poor prognosis for patients with hepatocellular carcinoma after orthotopic liver transplantation by promoting cancer metastasis and recurrence

TL;DR: DKK1 was an independent prognostic indicator for overall survival and cumulative recurrence in this cohort of HCC patients post-OLT and DKK1 overexpression may be potential molecular therapeutic targets for liver cancer.

Targeting Hepatocellular Carcinoma: What did we Discover so Far?

TL;DR: Mutations of the TP53 gene, as well as a deregulation between the expression of pro- and anti-apoptotic proteins of the BCL-2 family are observed, which represent a wide variety of mutations in HCC.
Journal ArticleDOI

Swainsonine inhibits growth and potentiates the cytotoxic effect of paclitaxel in hepatocellular carcinoma in vitro and in vivo.

TL;DR: Results suggest that swainsonine may be an important agent against hepatocellular carcinoma (HCC) via directly inhibiting HCC cell growth and enhancing the responsiveness of HCC cells to paclitaxel.

Clinical features associated with radiological response to sorafenib in unresectable hepatocellular carcinoma: a large multicenter study in Japan.

TL;DR: Investigation of the factors predicting a good response to sorafenib in Japanese patients with unresectable hepatocellular carcinoma found no established predictive factors.

Design of siRNA Therapeutics from the Molecular Scale

TL;DR: Design considerations for siRNA therapeutics, identifying criteria for choosing therapeutic targets, producing highly-active siRNA sequences, and designing an optimized delivery vehicle are discussed, providing logical guidelines for generating novel siRNAs.

New Guidelines to Evaluate the Response to Treatment in Solid Tumors

TL;DR: A model by which a combined assessment of all existing lesions, characterized by target lesions and nontarget lesions, is used to extrapolate an overall response to treatment is proposed, which is largely validated by the Response Evaluation Criteria in Solid Tumors Group and integrated into the present guidelines.

Cancer Statistics, 2009

TL;DR: The most recent data on cancer incidence, mortality, and survival from the American Cancer Society (ACS) is presented in this paper, where the authors compare the three major cancer sites in men (lung, prostate, and colon and rectum [colorectum]) and in two major cancers sites in women (breast and colorectal) over a 15-year period.

Management of hepatocellular carcinoma: An update

TL;DR: The American Association for the Study of Liver Diseases (AASLD) practice guidelines on the management of hepatocellular carcinoma (HCC) were updated in 2010 as discussed by the authors.
