Targeted Therapies for Hepatocellular Carcinoma

TLDR
Identification of oncogenes that mediate tumor progression, and trials that monitor their products as biomarkers, might lead to personalized therapy; reagents that interfere with signaling pathways required for HCC progression might be used to treat selected populations, and thereby maximize the efficacy and cost benefit.

Synergistic inhibition of autophagy and neddylation pathways as a novel therapeutic approach for targeting liver cancer

TL;DR: Blockage of autophagy enhanced the efficacy of MLN4924 in an orthotopic model of human liver cancer, with induction of NOXA and apoptosis in tumor tissues, and provide important preclinical evidence for clinical investigation of synergistic inhibition of neddylation and Autophagy in liver cancer.

STAT3 in hepatocellular carcinoma: new perspectives.

TL;DR: The oncogenic role of STAT3 in cancers has been challenged by several reports that suggest a tumor-suppressive activity, and here the effects of cytokine-activatedSTAT3 in HCC are discussed.

Differential Expression of Drug Uptake and Efflux Transporters in Japanese Patients with Hepatocellular Carcinoma

TL;DR: Results suggest different expression patterns of drug transporters in HCC, which are associated only in part with clinicopathological characteristics and may be promising for individualization and optimization of drug therapy for liver cancer.
Journal ArticleDOI

Klotho endows hepatoma cells with resistance to anoikis via VEGFR2/PAK1 activation in hepatocellular carcinoma.

TL;DR: A novel oncogenic function of Klotho is revealed in promoting anoikis resistance via activating VEGFR2/PAK1 signaling, thus facilitating tumor migration and invasion during hepatoma progression, which could provide a putative molecular mechanism for tumor metastasis.
Journal ArticleDOI

Notch3 inhibition enhances sorafenib cytotoxic efficacy by promoting GSK3b phosphorylation and p21 down-regulation in hepatocellular carcinoma.

TL;DR: It is demonstrated that inhibition of Notch3 signaling prevents HCC-mediate drug resistance and sensitizes HCC cells to sorafenib by overcoming drug resistance.

New Guidelines to Evaluate the Response to Treatment in Solid Tumors

TL;DR: A model by which a combined assessment of all existing lesions, characterized by target lesions and nontarget lesions, is used to extrapolate an overall response to treatment is proposed, which is largely validated by the Response Evaluation Criteria in Solid Tumors Group and integrated into the present guidelines.

Cancer Statistics, 2009

TL;DR: The most recent data on cancer incidence, mortality, and survival from the American Cancer Society (ACS) is presented in this paper, where the authors compare the three major cancer sites in men (lung, prostate, and colon and rectum [colorectum]) and in two major cancers sites in women (breast and colorectal) over a 15-year period.

Management of hepatocellular carcinoma: An update

TL;DR: The American Association for the Study of Liver Diseases (AASLD) practice guidelines on the management of hepatocellular carcinoma (HCC) were updated in 2010 as discussed by the authors.
