Abstract: Cognitive load theory was introduced in the 1980s as an instructional design theory based on several uncontroversial aspects of human cognitive architecture. Our knowledge of many of the characteristics of working memory, long-term memory and the relations between them had been well-established for many decades prior to the introduction of the theory. Curiously, this knowledge had had a limited impact on the field of instructional design with most instructional design recommendations proceeding as though working memory and long-term memory did not exist. In contrast, cognitive load theory emphasised that all novel information first is processed by a capacity and duration limited working memory and then stored in an unlimited long-term memory for later use. Once information is stored in long-term memory, the capacity and duration limits of working memory disappear transforming our ability to function. By the late 1990s, sufficient data had been collected using the theory to warrant an extended analysis resulting in the publication of Sweller et al. (Educational Psychology Review, 10, 251–296, 1998). Extensive further theoretical and empirical work have been carried out since that time and this paper is an attempt to summarise the last 20 years of cognitive load theory and to sketch directions for future research.

Abstract: The Y-maze can be used to assess short term memory in mice. Spontaneous alternation, a measure of spatial working memory, can be assessed by allowing mice to explore all three arms of the maze and is driven by an innate curiosity of rodents to explore previously unvisited areas. A mouse with intact working memory, and hence intact prefrontal cortical functions, will remember the arms previously visited and show a tendency to enter a less recently visited arm. Spatial reference memory, which is underlined by the hippocampus, can also be tested by placing the test mice into the Y-maze with one arm closed off during training. After an intertrial interval of for example 1 h, the mouse should remember which arm it has not explored previously and should visit this arm more often. This chapter describes the pre-test conditions, the materials required and the protocol for conducting and interpreting the results of these two related tests.

Abstract: Mental imagery can be advantageous, unnecessary and even clinically disruptive. With methodological constraints now overcome, research has shown that visual imagery involves a network of brain areas from the frontal cortex to sensory areas, overlapping with the default mode network, and can function much like a weak version of afferent perception. Imagery vividness and strength range from completely absent (aphantasia) to photo-like (hyperphantasia). Both the anatomy and function of the primary visual cortex are related to visual imagery. The use of imagery as a tool has been linked to many compound cognitive processes and imagery plays both symptomatic and mechanistic roles in neurological and mental disorders and treatments. Mental imagery plays a role in a variety of cognitive processes such as memory recall. In this review, Joel Pearson discusses recent insights into the neural mechanisms that underlie visual imagery, how imagery can be objectively and reliably measured, and how it affects general cognition.

Abstract: There is broad agreement that working memory is closely related to attention. This article delineates several theoretical options for conceptualizing this link, and evaluates their viability in light of their theoretical implications and the empirical support they received. A first divide exists between the concept of attention as a limited resource, and the concept of attention as selective information processing. Theories conceptualizing attention as a resource assume that this resource is responsible for the limited capacity of working memory. Three versions of this idea have been proposed: Attention as a resource for storage and processing, a shared resource for perceptual attention and memory maintenance, and a resource for the control of attention. The first of these three is empirically well supported, but the other two are not. By contrast, when attention is understood as a selection mechanism, it is usually not invoked to explain the capacity limit of working memory - rather, researchers ask how different forms of attention interact with working memory, in two areas. The first pertains to attentional selection of the contents of working memory, controlled by mechanisms of filtering out irrelevant stimuli, and removing no-longer relevant representations from working memory. Within working memory contents, a single item is often selected into the focus of attention for processing. The second area pertains to the role of working memory in cognitive control. Working memory contributes to controlling perceptual attention - by holding templates for targets of perceptual selection - and controlling action - by holding task sets to implement our current goals.

Abstract: Traversing sensory environments requires keeping relevant information in mind while simultaneously processing new inputs. Visual information is kept in working memory via feature-selective responses in early visual cortex, but recent work has suggested that new sensory inputs obligatorily wipe out this information. Here we show region-wide multiplexing abilities in classic sensory areas, with population-level response patterns in early visual cortex representing the contents of working memory alongside new sensory inputs. In a second experiment, we show that when people get distracted, this leads to both disruptions of mnemonic information in early visual cortex and decrements in behavioral recall. Representations in the intraparietal sulcus reflect actively remembered information encoded in a transformed format, but not task-irrelevant sensory inputs. Together, these results suggest that early visual areas play a key role in supporting high-resolution working memory representations that can serve as a template for comparison with incoming sensory information.

Abstract: Beta oscillations (∼13 to 30 Hz) have been observed during many perceptual, cognitive, and motor processes in a plethora of brain recording studies. Although the function of beta oscillations (hereafter "beta" for short) is unlikely to be explained by any single monolithic description, we here discuss several convergent findings. In prefrontal cortex (PFC), increased beta appears at the end of a trial when working memory information needs to be erased. A similar "clear-out" function might apply during the stopping of action and the stopping of long-term memory retrieval (stopping thoughts), where increased prefrontal beta is also observed. A different apparent role for beta in PFC occurs during the delay period of working memory tasks: it might serve to maintain the current contents and/or to prevent interference from distraction. We confront the challenge of relating these observations to the large literature on beta recorded from sensorimotor cortex. Potentially, the clear-out of working memory in PFC has its counterpart in the postmovement clear-out of the motor plan in sensorimotor cortex. However, recent studies support alternative interpretations. In addition, we flag emerging research on different frequencies of beta and the relationship between beta and single-neuron spiking. We also discuss where beta might be generated: basal ganglia, cortex, or both. We end by considering the clinical implications for adaptive deep-brain stimulation.

Abstract: Recently it has been proposed that information in working memory (WM) may not always be stored in persistent neuronal activity but can be maintained in 'activity-silent' hidden states, such as synaptic efficacies endowed with short-term synaptic plasticity. To test this idea computationally, we investigated recurrent neural network models trained to perform several WM-dependent tasks, in which WM representation emerges from learning and is not a priori assumed to depend on self-sustained persistent activity. We found that short-term synaptic plasticity can support the short-term maintenance of information, provided that the memory delay period is sufficiently short. However, in tasks that require actively manipulating information, persistent activity naturally emerges from learning, and the amount of persistent activity scales with the degree of manipulation required. These results shed insight into the current debate on WM encoding and suggest that persistent activity can vary markedly between short-term memory tasks with different cognitive demands.

Abstract: Neurocognitive heterogeneity is increasingly recognized as a valid phenomenon in ADHD, with most estimates suggesting that executive dysfunction is present in only about 33%–50% of these children. However, recent critiques question the veracity of these estimates because our understanding of executive functioning in ADHD is based, in large part, on data from single tasks developed to detect gross neurological impairment rather than the specific executive processes hypothesized to underlie the ADHD phenotype. The current study is the first to comprehensively assess heterogeneity in all three primary executive functions in ADHD using a criterion battery that includes multiple tests per construct (working memory, inhibitory control, set shifting). Children ages 8–13 (M = 10.37, SD = 1.39) with and without ADHD (N = 136; 64 girls; 62% Caucasian/Non-Hispanic) completed a counterbalanced series of executive function tests. Accounting for task unreliability, results indicated significantly improved sensitivity and specificity relative to prior estimates, with 89% of children with ADHD demonstrating objectively-defined impairment on at least one executive function (62% impaired working memory, 27% impaired inhibitory control, 38% impaired set shifting; 54% impaired on one executive function, 35% impaired on two or all three executive functions). Children with working memory deficits showed higher parent- and teacher-reported ADHD inattentive and hyperactive/impulsive symptoms (BF10 = 5.23 × 104), and were slightly younger (BF10 = 11.35) than children without working memory deficits. Children with vs. without set shifting or inhibitory control deficits did not differ on ADHD symptoms, age, gender, IQ, SES, or medication status. Taken together, these findings confirm that ADHD is characterized by neurocognitive heterogeneity, while suggesting that contemporary, cognitively-informed criteria may provide improved precision for identifying a smaller number of neuropsychologically-impaired subtypes than previously described.

Abstract: In the present meta-analysis we examined the near- and far-transfer effects of training components of children's executive functions skills: working memory, inhibitory control, and cognitive flexibility. We found a significant near-transfer effect (g+ = 0.44, k = 43, p < .001) showing that the interventions in the primary studies were successful in training the targeted components. However, we found no convincing evidence of far-transfer (g+ = 0.11, k = 17, p = .11). That is, training a component did not have a significant effect on the untrained components. By showing the absence of benefits that generalize beyond the trained components, we question the practical relevance of training specific executive function skills in isolation. Furthermore, the present results might explain the absence of far-transfer effects of working memory training on academic skills (Melby-Lervag & Hulme, 2013; Sala & Gobet, 2017). Abstract: Working memory involves storing and/or manipulating previously encoded information over a short-term delay period, which is typically followed by a behavioral response based on the remembered information. Although working memory tasks often engage dorsolateral prefrontal cortex, few studies have investigated whether their subprocesses are localized to different cortical depths in this region, and none have done so in humans. Here we use high-resolution functional MRI to interrogate the layer specificity of neural activity during different periods of a delayed-response task in dorsolateral prefrontal cortex. We detect activity time courses that follow the hypothesized patterns: namely, superficial layers are preferentially active during the delay period, specifically in trials requiring manipulation (rather than mere maintenance) of information held in working memory, and deeper layers are preferentially active during the response. Results demonstrate that layer-specific functional MRI can be used in higher-order brain regions to noninvasively map cognitive processing in humans.

Abstract: The relationships between cognitive impairment that exist during the clinical course of multiple sclerosis (MS) remain poorly described. The effect of disease duration has been studied in a few longitudinal cohorts and some cross-sectional studies that suggest that cognitive deficits tend to extend with disease duration. However, the effect of disease duration seems to be confounded by the effect of age. At the pre-clinical stage, cognitive deficits have been observed in patients with radiologically isolated syndromes, and their profile is similar than in clinically isolated syndromes (CIS) and relapsing-remitting MS (RRMS). The frequency of cognitive impairment tends to be higher in RRMS than in CIS. In these phenotypes, slowness of information processing speed (IPS) and episodic verbal and visuo-spatial memory deficits are frequently observed, but executive functions, and in particular verbal fluency, could also be impaired. More frequent and severe deficits are reported in SPMS than in RRMS with more severe deficits for memory tests, working memory and IPS. Similarly to what is observed in SPMS, patients with primary progressive MS (PPMS) present with a wide range of cognitive deficits in IPS, attention, working memory, executive functions, and verbal episodic memory with more tests and domains impaired than RRMS patients. Altogether these data suggested that not only the duration of the disease and age play an important role in the cognitive profile of patients, but also the phenotype itself, probably because of its specific pathological mechanism.

Abstract: Verbal Working memory (vWM) capacity measures the ability to maintain and manipulate verbal information for a short period of time. The specific neural correlates of this construct are still a matter of debate. The aim of this study was to conduct a coordinate-based meta-analysis of 42 fMRI studies on visual vWM in healthy subjects (n=795, males=459, females=325, unknown=11; age range: 18-75). The studies were obtained after an exhaustive literature search on PubMed, Scopus, Web of Science, and Brainmap database. We analyzed regional activation differences during fMRI tasks with the anisotropic effect-size version of seed-based d mapping software (ES-SDM). The results were further validated by performing jackknife sensitivity analyses and heterogeneity analyses. We investigated the effect of numerous relevant influencing factors by fitting corresponding linear regression models. We isolated consistent activation in a network containing fronto-parietal areas, right cerebellum, and basal ganglia structures. Regarding lateralization, the results pointed towards a bilateral frontal activation, a left-lateralization of parietal regions and a right-lateralization of the cerebellum, indicating that the left-hemisphere concept of vWM should be reconsidered. We also isolated activation in regions important for response inhibition, emphasizing the role of attentional control in vWM. Moreover, we found a significant influence of mean reaction time, load and age on activation associated with vWM. Activation in left medial frontal gyrus, left precentral gyrus and left precentral gyrus turned out to be positively associated with mean reaction time whereas load was associated with activation across the PFC, fusiform gyrus, parietal cortex and parts of the cerebellum. In the latter case activation was mainly detectable in both hemispheres whereas the influence of age became manifest predominantly in the left hemisphere. This led us to conclude that future vWM studies should take these factors into consideration.

Abstract: Schizophrenia is a severe neuropsychiatric disorder with persistence of symptoms throughout adult life in most of the affected patients. This unfavorable course is associated with multiple episodes and residual symptoms, mainly negative symptoms and cognitive deficits. The neural diathesis-stress model proposes that psychosocial stress acts on a pre-existing vulnerability and thus triggers the symptoms of schizophrenia. Childhood trauma is a severe form of stress that renders individuals more vulnerable to developing schizophrenia; neurobiological effects of such trauma on the endocrine system and epigenetic mechanisms are discussed. Childhood trauma is associated with impaired working memory, executive function, verbal learning, and attention in schizophrenia patients, including those at ultra-high risk to develop psychosis. In these patients, higher levels of childhood trauma were correlated with higher levels of attenuated positive symptoms, general symptoms, and depressive symptoms; lower levels of global functioning; and poorer cognitive performance in visual episodic memory end executive functions. In this review, we discuss effects of specific gene variants that interact with childhood trauma in patients with schizophrenia and describe new findings on the brain structural and functional level. Additive effects between childhood trauma and brain-derived neurotrophic factor methionine carriers on volume loss of the hippocampal subregions cornu ammonis (CA)4/dentate gyrus and CA2/3 have been reported in schizophrenia patients. A functional magnetic resonance imaging study showed that childhood trauma exposure resulted in aberrant function of parietal areas involved in working memory and of visual cortical areas involved in attention. In a theory of mind task reflecting social cognition, childhood trauma was associated with activation of the posterior cingulate gyrus, precuneus, and dorsomedial prefrontal cortex in patients with schizophrenia. In addition, decreased connectivity was shown between the posterior cingulate/precuneus region and the amygdala in patients with high levels of physical neglect and sexual abuse during childhood, suggesting that disturbances in specific brain networks underlie cognitive abilities. Finally, we discuss some of the questionnaires that are commonly used to assess childhood trauma and outline possibilities to use recent biostatistical methods, such as machine learning, to analyze the resulting datasets.

Abstract: Visual working memory enables us to hold onto past sensations in anticipation that these may become relevant for guiding future actions. Yet laboratory tasks have treated visual working memories in isolation from their prospective actions and have focused on the mechanisms of memory retention rather than utilization. To understand how visual memories become used for action, we linked individual memory items to particular actions and independently tracked the neural dynamics of visual and motor selection when memories became used for action. This revealed concurrent visual-motor selection, engaging appropriate visual and motor brain areas at the same time. Thus we show that items in visual working memory can invoke multiple, item-specific, action plans that can be accessed together with the visual representations that guide them, affording fast and precise memory-guided behavior.

Abstract: In order to efficiently process incoming visual information, selective attention acts as a filter that enhances relevant and suppresses irrelevant information. In this study, we used an event-related potential (ERP) approach with systematic lateralization to investigate enhancement and suppression during encoding of information into visual working memory (WM) separately. We used a change detection task in which observers had to memorize some items while ignoring other items. We found that the to-be-ignored items elicited a PD component in the ERP, suggesting that irrelevant information is actively suppressed from WM. The PD amplitude increased with distractor load and decreased with the ability to group distractors according to Gestalt principles. This suggests that the PD can be used as an indicator of how efficiently items can be suppressed from entering WM. Furthermore, while lateral memory-targets elicited a "traditional" CDA (starting ~300 ms), lateral memory-distractors elicited a sustained positivity contralateral to memory-distractors (CDAp, starting ~400 ms). In sum the results suggest that inhibition of irrelevant information is an important factor for efficient WM and is reflected in spontaneous (PD) and sustained suppression (CDAp).

Abstract: Introduction Brain processes of working memory involve oscillatory activities at multiple frequencies in local and long-range neural networks. The current study addressed the specific roles of alpha oscillations during memory encoding and retention, supporting the hypothesis that multiple functional mechanisms of alpha oscillations exist in parallel. Method We recorded magnetoencephalography (MEG) in 25 healthy young adults, who performed a variant of a Sternberg working memory task. A sequential list of five consonant letters was visually presented and was followed after a 2.0 s retention interval by a probe of a pair of two letters from the study list. Participants responded whether the probe pair was in same or reversed order in the list. Result Reaction time (RT) was shortest for the first letters in the list, increased with increasing serial position, and shorter for the last position. RT was substantially longer for the probe in reversed order. Time-frequency analysis of the MEG revealed event-related desynchronization (ERD) of alpha oscillations during the encoding interval and an alpha power increase (ERS) during memory retention. Alpha ERD during encoding occurred at 10 Hz and ERS during retention at 12 Hz, suggesting different alpha mechanisms. Analysis of alpha coherence and alpha-gamma cross-spectral coupling, applied to MEG beamformer source activity, revealed connectivity across brain areas. Additionally, alpha-gamma coupling identified centers of local computation. The connectivity between occipital and frontotemporal areas was correlated with alpha ERS during memory retention. Cross-frequency coupling between alpha phase and gamma amplitude depicted a hierarchy of information flow from frontal to temporal and occipital brain areas. Conclusion Alpha decrease during encoding indicates an active state of visual processing, while subsequent ERS indicates inhibition of further visual input for protecting the memory, and phasic timing of temporal and occipital gamma oscillations is related to a long-range working memory networks.

Abstract: Everyday life is defined by goal states that are continuously reprioritized based on available, often affective information. To pursue these goals, individuals need to process and maintain goal-relevant information, while ignoring potentially salient information that distracts resources from these goals. Empirically, this ability has typically been operationalized as working memory (WM) capacity. A growing body of research is investigating the impact of information's affective salience on WM capacity. In the present review we address this question by exploring the potential differential impact of affective compared with neutral information on WM, and the underlying neural substrates. One-hundred and 65 studies (N = 7,433) were included in the meta-analysis. Results showed negligible to small (d = -.07-.20) effects of affective information on behavioral measures of WM in healthy individuals (n = 4,936) that varied as a function of valence and task-relevance. Heterogeneity analyses were significant, demonstrating the need to identify further study-specific factors and individual differences that moderate affective WM. At the neural level (33 studies; n = 683), processing affective versus neutral material during WM tasks was associated with more frequent recruitment of the vlPFC, the amygdala, and the temporo-occipital cortex. In contrast to healthy individuals, across behavioral studies those suffering from mental health problems (n = 2,041) showed impaired WM accuracy (d = -0.21) in the presence of affective material. These findings highlight the importance of integrating behavioral and neural levels of analysis.
