TL;DR: A hierarchical model for undertaking a meta‐analysis of multiple studies, in which it is not necessary that the same genetic markers are measured in each study, provides an overall estimate of the causal relationship between the phenotype and the outcome, and an assessment of its heterogeneity across studies.
Journal ArticleDOI
Canonical correlation analysis for gene-based pleiotropy discovery.
TL;DR: Improved statistical power is found in the detection of previously reported genetic associations, and a number of novel pleiotropic associations between genetic variants and phenotypes are identified.
Journal ArticleDOI
Lipid profile, plasma apolipoproteins, and pre-eclampsia risk in the GenPE case-control study.
Norma C. Serrano,Elizabeth Guío-Mahecha,Doris Cristina Quintero-Lesmes,Silvia Becerra-Bayona,María C Paez,Monica Beltran,Victor M. Herrera,Lydia J. Leon,David J. Williams,Juan P. Casas +9 more
TL;DR: Higher levels of triglycerides, ApoE and the ApoB/ApoA1 ratio are associated with an increased risk of pre-eclampsia, particularly for those cases with an early onset.
Journal ArticleDOI
Multilocus Bayesian meta-analysis of gene-disease associations.
Paul J. Newcombe,Claudio J. Verzilli,Juan P. Casas,Aroon D. Hingorani,Liam Smeeth,John C. Whittaker +5 more
TL;DR: A Bayesian approach is presented that exploits prior information on underlying haplotypes to allow multi-marker analysis incorporating data from all relevant studies of a gene or region, irrespective of the markers typed.
Journal ArticleDOI
A ‘Polypill’ Aimed At Preventing Cardiovascular Disease Could Prove Highly Cost-Effective For Use In Latin America
Leonelo E. Bautista,Lina M. Vera-Cala,Daniel Ferrante,Victor M. Herrera,J. Jaime Miranda,Rafael N. Pichardo,José R. Sánchez Abanto,Catterina Ferreccio,Egle Silva,Myriam Oróstegui Arenas,Julio A. Chirinos,Josefina Medina-Lezama,Cynthia M. Pérez,N Schapochnik,Juan P. Casas +14 more
TL;DR: The polypill would be very cost-effective even in the country with the lowest gross national income, however, policy makers must weigh the value of intervention with the polypill against other interventions, as well as their country's willingness and ability to pay for the intervention.
