HSP90 at the hub of protein homeostasis: emerging mechanistic insights

TLDR
Comprehensive understanding of how HSP90 functions promises not only to provide new avenues for therapeutic intervention, but to shed light on fundamental biological questions.
Abstract
Heat shock protein 90 (HSP90) is a highly conserved molecular chaperone that facilitates the maturation of a wide range of proteins (known as clients). Clients are enriched in signal transducers, including kinases and transcription factors. Therefore, HSP90 regulates diverse cellular functions and exerts marked effects on normal biology, disease and evolutionary processes. Recent structural and functional analyses have provided new insights on the transcriptional and biochemical regulation of HSP90 and the structural dynamics it uses to act on a diverse client repertoire. Comprehensive understanding of how HSP90 functions promises not only to provide new avenues for therapeutic intervention, but to shed light on fundamental biological questions.

Does the cranial mesenchyme contribute to neural fold elevation during neurulation

TL;DR: The evidence implicating the cranial mesenchyme in providing a driving force for neural fold elevation is reviewed to evaluate whether there are sufficient data to support this hypothesis.

Analysis of serum Hsp90 as a potential biomarker of β cell autoimmunity in type 1 diabetes.

TL;DR: Elevated Hsp90 levels are present in youth with β cell autoimmunity, but are not able to distinguish youth or adult type 1 diabetes progressors from nonprogressors in samples collected 12 months prior to diabetes development.

Hsp90 as a therapeutic target in endocrinology: current evidence

TL;DR: The ability of heat shock protein 90 to modulate many growth and signal- ing pathways simultaneously makes it an attractive target in the field of cancer therapeutics and provided the initial impetus for significant efforts over the past decade to identify Hsp90 inhibitors, several of which are now showing promise in the clinic for cancer treatment.

De novo transcriptome sequencing of Isaria cateniannulata and comparative analysis of gene expression in response to heat and cold stresses.

TL;DR: The transcriptome of I. cateniannulata is sequenced and de novo assembled and the expression patterns of 18 randomly selected candidate DEGs resulting from quantitative real-time PCR (qRT-PCR) were consistent with their transcriptome analysis results.

The Role of HSP90 Chaperones in Stability and Plasticity of Ontogenesis of Plants under Normal and Stressful Conditions (Arabidopsis thaliana)

TL;DR: The role of H SP90 in growth maintenance and morphogenesis stability, developmental plasticity, and mechanisms of stress tolerance in plants was shown and a model for the interaction of the HSP90 functions under normal and stressful conditions is presented.

Mechanism of Activation of the Raf-Erk Signaling Pathway by Oncogenic Mutations of B-Raf

TL;DR: The high activity mutants signal to ERK by directly phosphorylating MEK, whereas the impaired activity mutants stimulate MEK by activating endogenous C-RAF, possibly via an allosteric or transphosphorylation mechanism.

HSP90 and the chaperoning of cancer.

TL;DR: Pharmacologically 'bribing' the essential guard duty of the chaperone HSP90 (heat-shock protein of 90 kDa) seems to offer a unique anticancer strategy of considerable promise.

Hsp90 as a capacitor for morphological evolution

TL;DR: It is reported that when Drosophila Hsp90 is mutant or pharmacologically impaired, phenotypic variation affecting nearly any adult structure is produced, with specific variants depending on the genetic background and occurring both in laboratory strains and in wild populations.
