Several dectin‐1 crystal structures are reported, including a high‐resolution structure and a 2.8 Å resolution structure in which a short soaked natural β‐glucan is trapped in the crystal lattice, which suggest potential mechanisms of defense against fungal pathogens.
Abstract
The murine molecule dectin-1 (known as the β-glucan receptor in humans) is an immune cell surface receptor implicated in the immunological defense against fungal pathogens. Sequence analysis has indicated that the dectin-1 extracellular domain is a C-type lectin-like domain, and functional studies have established that it binds fungal β-glucans. We report several dectin-1 crystal structures, including a high-resolution structure and a 2.8 A resolution structure in which a short soaked natural β-glucan is trapped in the crystal lattice. In vitro characterization of dectin-1 in the presence of its natural ligand indicates higher-order complex formation between dectin-1 and β-glucans. These combined structural and biophysical data considerably extend the current knowledge of dectin-1 structure and function, and suggest potential mechanisms of defense against fungal pathogens.

Zymosan-induced leukocyte and cytokine changes in pigs: a new model for streamlined drug testing against severe COVID-19

TL;DR: Within a 6 h experiment, the model enables consecutive reproduction of a symptom triad that is characteristic of late-stage COVID-19, and the presented relatively simple large animal model may advance the R&D of drugs against these conditions.

TL;DR: Results suggest that WSC3 might act as a structural component of the fungal cell wall to maintain cell wall integrity while FGB1 is acting as universal suppressor of MAMP-triggered ROS production.

Characterization of fungal exposure and dectin-1 expression in healthy horses and horses with severe asthma.

TL;DR: Equine dectin-1 was localized in tissues and cells, and functional isoforms were upregulated significantly in BALF after stabling, suggesting exposure to fungi could contribute to airway inflammation by increasing dectIn-1functional isoforms, and exposure to indoor molds should be avoided.
