TL;DR: In this article, a collaborative GWAS involving 9,772 cases of European descent collected by 23 research groups working in 15 different countries, they have replicated almost all of the previously suggested associations and identified at least a further 29 novel susceptibility loci.
Journal ArticleDOI
Meta-analysis of genome scans and replication identify CD6, IRF8 and TNFRSF1A as new multiple sclerosis susceptibility loci
Philip L. De Jager,Philip L. De Jager,Xiaoming Jia,Joanne Wang,Paul I.W. de Bakker,Linda Ottoboni,Linda Ottoboni,Neelum T. TL;DR: It is reported that the susceptibility allele near IRF8, which encodes a transcription factor known to function in type I Interferon signaling, is associated with higher mRNA expression of interferon-response pathway genes in subjects with MS.
Journal ArticleDOI
Cutting Edge: TREM-2 Attenuates Macrophage Activation
Isaiah R. Turnbull,Susan Gilfillan,Marina Cella,Taiki Aoshi,Mark J. Miller,Laura Piccio,M. O. Hernandez,Marco Colonna +7 more
TL;DR: It is shown that TREM-2 is expressed on newly differentiated and alternatively activated macrophages and functions to restrain macrophage activation and completely accounts for the increased cytokine production previously reported by DAP12−/−macrophages.
Journal ArticleDOI
Chemokines trigger immediate beta2 integrin affinity and mobility changes: differential regulation and roles in lymphocyte arrest under flow.
Gabriela Constantin,Meytham Majeed,Cinzia Giagulli,Laura Piccio,Ji Yun Kim,Eugene C. Butcher,Carlo Laudanna +6 more
TL;DR: It is shown that the chemokines SLC, ELC, and SDF-1alpha rapidly induce lateral mobility and transient increase of affinity of the beta2 integrin LFA-1 that can coordinately determine the vascular arrest of circulating lymphocytes under physiologic conditions.
Journal ArticleDOI
The Microglial Innate Immune Receptor TREM2 Is Required for Synapse Elimination and Normal Brain Connectivity.
Fabia Filipello,Raffaella Morini,Irene Corradini,Valerio Zerbi,Alice Canzi,Bernadeta Michalski,Marco Erreni,Marija Markicevic,Chiara Starvaggi-Cucuzza,Karel Otero,Laura Piccio,Francesca Cignarella,Fabio Perrucci,Matteo Tamborini,Marco Genua,Lawrence Rajendran,Elisabetta Menna,Stefania Vetrano,Margaret Fahnestock,Rosa C. Paolicelli,Michela Matteoli +20 more
TL;DR: It is shown that TREM2 is essential for microglia‐mediated synaptic refinement during the early stages of brain development, and also reduced in autistic patients, suggesting that the receptor may be involved in neurodevelopmental diseases.
