Abstract: This paper is a structural and tectonic approach to the emplacement and deformation of granitoids. The main methods available in structural geology are briefly reviewed and this emphasises that (a) a wealth of data, particularly strain and shear sense, which pertain to these problems, can be determined in and around plutons; (b) given the nature, unlike many other crustal rock types, of granites to crystallise from isotropic through weakly anisotropic crystal suspension fluids, that deformation which has occurred in these states may not be well preserved; and (c) it is entirely possible, using this methodology, to separate deformation resulting from externally originating tectonic stresses from that which is associated with internal magma-related stresses. It is also recommended that the genetically-based Cloosian classification of granite fabrics and structures into “primary” (magmatic flow/magmatic flow current) and “secondary”, be abandoned and that a more observationally-based approach which classifies granite deformation fabrics and structures according to their time of occurrence relative to the crystallisation state of the congealing magma, be adopted (i.e. pre-full crystallisation deformation and crystal plastic strain deformation).Examples of recent, structurally based, studies of emplacement mechanisms of plutons within tectonic settings are described and these show that, in general, space for magma can be created by the combination of tectonically-created cavities and internal magma-related buoyancy. This occurs in both transcurrent and extensional tectonic settings and there is no reason to doubt that it can happen in compressive-contractional regimes. It is concluded that transient and permanent space creation, such as may be exploited by available magmas, is a typical feature of the tectonically stressed and deforming lithosphere and this, in combination with the natural buoyancy and ascending tendency of magmas, can generate the varied emplacement mechanisms of granites.

Abstract: Triclosan (5-chloro-2-(2,4-dichlorophenoxy) phenol) has been used for more than 30 years as a general antibacterial and antifungal agent, and is found in formulations as diverse as toothpastes, cosmetics, antiseptic soaps, carpets, plastic kitchenware and toys. It has recently been suggested that triclosan blocks lipid biosynthesis by specifically inhibiting the enzyme enoyl-acyl carrier protein reductase (ENR)1. We have carried out a structural analysis and inhibition experiments on a complex of ENR from the bacterium Escherichia coli with triclosan and NAD+. We find that triclosan acts as a site-directed, very potent inhibitor of the enzyme by mimicking its natural substrate.

Abstract: Cardiac resynchronization therapy (CRT) has emerged as an attractive intervention to improve left ventricular mechanical function by changing the sequence of electrical activation. Unfortunately, many patients receiving CRT do not benefit but are subjected to device complications and costs. Thus, there is a need for better selection criteria. Current criteria for CRT eligibility include a QRS duration ≥120 ms. However, QRS morphology is not considered, although it can indicate the cause of delayed conduction. Recent studies have suggested that only patients with left bundle branch block (LBBB) benefit from CRT, and not patients with right bundle branch block or nonspecific intraventricular conduction delay. The authors review the pathophysiologic and clinical evidence supporting why only patients with complete LBBB benefit from CRT. Furthermore, they review how the threshold of 120 ms to define LBBB was derived subjectively at a time when criteria for LBBB and right bundle branch block were mistakenly reversed. Three key studies over the past 65 years have suggested that 1/3 of patients diagnosed with LBBB by conventional electrocardiographic criteria may not have true complete LBBB, but likely have a combination of left ventricular hypertrophy and left anterior fascicular block. On the basis of additional insights from computer simulations, the investigators propose stricter criteria for complete LBBB that include a QRS duration ≥140 ms for men and ≥130 ms for women, along with mid-QRS notching or slurring in ≥2 contiguous leads. Further studies are needed to reinvestigate the electrocardiographic criteria for complete LBBB and the implications of these criteria for selecting patients for CRT.
