Abstract: Low molecular weight and high molecular weight metal ion binders present in blood plasma are shortly described. The binding of vanadium and ruthenium complexes by these components has received much attention, namely their interactions with human serum albumin and transferrin, and these studies are critically reviewed. The influence of the protein binding on the bioavailability of the prospective drugs, namely on the transport by blood plasma and uptake by cells is also discussed. It is concluded that vanadium compounds are mainly transported in blood by transferrin, but that no study has properly addressed the influence of albumin and transferrin in the vanadium uptake by cells. Ruthenium complexes bind strongly to HSA, most likely at the level of His residues, leading to the formation of stable adducts. If the kinetics of binding to this protein is fast enough, probably they are mainly transported by this serum protein. Nevertheless, at least for a few Ru III -complexes, hTf seems to play an active role in the uptake of ruthenium, while HSA may provide selectivity and higher activity for the compounds due to an enhanced permeability effect.

Abstract: The copper content of recombinant CotA laccase from Bacillus subtilis produced by Escherichia coli cells is shown to be strongly dependent on the presence of copper and oxygen in the culture media. In copper-supplemented media, a switch from aerobic to microaerobic conditions leads to the synthesis of a recombinant holoenzyme, while the maintenance of aerobic conditions results in the synthesis of a copper-depleted population of proteins. Strikingly, cells grown under microaerobic conditions accumulate up to 80-fold more copper than aerobically grown cells. In vitro copper incorporation into apoenzymes was monitored by optical and electron paramagnetic resonance (EPR) spectroscopy. This analysis reveals that copper incorporation into CotA laccase is a sequential process, with the type 1 copper center being the first to be reconstituted, followed by the type 2 and the type 3 copper centers. The copper reconstitution of holoCotA derivatives depleted in vitro with EDTA results in the complete recovery of the native conformation as monitored by spectroscopic, kinetic and thermal stability analysis. However, the reconstitution of copper to apo forms produced in cultures under aerobic and copper-deficient conditions resulted in incomplete recovery of biochemical properties of the holoenzyme. EPR and resonance Raman data indicate that, presumably, folding in the presence of copper is indispensable for the correct structure of the trinuclear copper-containing site.

Abstract: This paper presents a literature review and experimental results on the effect of high incorporation levels of fly ash (FA) and recycled concrete aggregates (RCA), individually and jointly, on the pore system of concrete that remarkably influences its durability. For that purpose, apart from an extensive literature review, three tests were performed, including electrical resistivity (ER) test, which indirectly measures the interconnected porosity of concrete, and water absorption (WA) by capillarity and immersion tests that both depend on the pores number and size but in a different way. A comparison between the experimental results and the literature is also presented to show the main findings and the research needs. The results show that WA increases and ER decreases with increasing incorporation level of RCA, and the opposite occurs with the addition of FA for both tests. The reduction percentage of WA was higher in mixes with both RCA and FA when compared to the sum of reductions in mixes with only RCA or FA. Thus, it is advisable to produce concrete with both mentioned non-traditional materials in terms of WA and ER of concrete. In addition, the benefit of incorporating of FA and RCA in concrete increased even more when superplasticizers was used.
