TL;DR: The ability to interrogate ex vivo response to PD-1 blockade using murine- and patient-derived organotypic tumor spheroids (MDOTS/PDOTS) is demonstrated, which represents a novel platform to evaluate ICB using established murine models as well as clinically relevant patient specimens.

TL;DR: Drug-induced apoptosis in EGFR-mutant non-small cell lung cancer is prevented by blockade of ERK1/2 reactivation following EGFR TKI treatment by combined EGFR/MEK inhibition, which uncovers cells that survive by entering a senescence-like dormant state characterized by high YAP/TEAD activity.
