TL;DR: Somatic copy number aberrations (SCNAs) in 1,087 unique medulloblastomas are reported, including recurrent events targeting TGF-β signalling in Group 3, and NF-κB signalling in Groups 4, which suggest future avenues for rational, targeted therapy.
Journal ArticleDOI
Quiescent sox2(+) cells drive hierarchical growth and relapse in sonic hedgehog subgroup medulloblastoma.
Robert Vanner,Marc Remke,Marco Gallo,Hayden J. Selvadurai,Fiona J. Coutinho,Lilian Lee,Michelle Kushida,Renee Head,Sorana Morrissy,Xueming Zhu,Tzvi Aviv,Veronique Voisin,Ian D. Bader,Konrad Hochedlinger,Andrey Korshunov,Michael D. Taylor,Peter B. Dirks +28 more
TL;DR: It is shown that quiescent, therapy-resistant Sox2(+) cells propagate sonic hedgehog subgroup medulloblastoma by a mechanism that mirrors a neurogenic program, creating a reservoir for tumor regrowth and addressing functional heterogeneity within tumors.
Journal ArticleDOI
The RNA-binding SAM domain of Smaug defines a new family of post-transcriptional regulators.
Tzvi Aviv,Tzvi Aviv,Zhen Lin,Stefanie Lau,Laura M. Rendl,Frank Sicheri,Frank Sicheri,Craig A. Smibert +7 more
TL;DR: It is shown here that the SAM domain of Saccharomyces cerevisiae Vts1 binds RNA with the same specificity as Smaug and that VTS1 induces transcript degradation through a mechanism involving the cytoplasmic deadenylase CCR4.
Journal ArticleDOI
Inhibition of Dopamine Receptor D4 Impedes Autophagic Flux, Proliferation, and Survival of Glioblastoma Stem Cells
Sonam Dolma,Hayden J. Selvadurai,Xiaoyang Lan,Lilian Lee,Michelle Kushida,Veronique Voisin,Heather Whetstone,Milly So,Tzvi Aviv,Nicole I. Park,Xueming Zhu,Changjiang Xu,Renee Head,Katherine J. Rowland,Mark Bernstein,Ian D. Clarke,Gary D. Bader,Lea Harrington,John H. Brumell,Mike Tyers,Peter B. Dirks +20 more
TL;DR: A role for neurochemical pathways in governing GBM stem cell proliferation and suggest therapeutic approaches for GBM is demonstrated and a role for dopamine receptor D4 antagonists is demonstrated.
Journal ArticleDOI
Sequence-specific recognition of RNA hairpins by the SAM domain of Vts1p
TL;DR: The 2.0-Å X-ray structure of the Vts1p SAM domain bound to a high-affinity RNA ligand is reported and NNF1 is validated as an endogenous target of VTS1p among 79 transcripts that copurify with Vts 1p.
