Journal ArticleDOI
Modification and de novo design of non-ribosomal peptide synthetases using specific assembly points within condensation domains.
Kenan A. J. Bozhueyuek,Annabell Linck,Andreas Tietze,Janik Kranz,Frank Wesche,Sarah Nowak,Florian Fleischhacker,Yan-Ni Shi,Peter Grün,Helge B. Bode +9 more
TL;DR: A new fusion point inside the condensation domains of NRPSs is described that results in the development of the exchange unit condensation domain (XUC) concept, which enables the efficient production of peptides, even containing non-natural amino acids, in yields up to 280 mg l−1.
Journal ArticleDOI
Lignans and aromatic glycosides from Piper wallichii and their antithrombotic activities
Yan-Ni Shi,Yi-Ming Shi,Lian Yang,Xing-Cong Li,Jin-Hua Zhao,Yan Qu,Hong-Tao Zhu,Dong Wang,Rong-Rong Cheng,Chong-Ren Yang,Min Xu,Ying-Jun Zhang +11 more
TL;DR: It is demonstrated that lignans, phenylpropanoid and biphenyl found in Piper wallichii may be responsible for antithrombotic effect of the titled plant.
Journal ArticleDOI
Promoter Activation in Δhfq Mutants as an Efficient Tool for Specialized Metabolite Production Enabling Direct Bioactivity Testing
Edna Bode,Antje K. Heinrich,Merle Hirschmann,Desalegne Abebew,Yan-Ni Shi,Tien Duy Vo,Frank Wesche,Yi-Ming Shi,Peter Grün,Svenja Simonyi,Nadine Keller,Yvonne Engel,Sebastian L Wenski,Reuel Bennet,Sophie Beyer,Iris Bischoff,Anthony Buaya,Sophie C. Brandt,Ibrahim Cakmak,Harun Cimen,Simone Eckstein,Denia Frank,Robert Fürst,Martin Gand,Gerd Geisslinger,Selcuk Hazir,Marina Henke,Ralf Heermann,Virginie Lecaudey,Wilhelm Schäfer,Susanne Schiffmann,Anja Schüffler,Rebecca Schwenk,Marisa Skaljac,Eckhard Thines,Marco Thines,Thomas Ulshöfer,Andreas Vilcinskas,Thomas A. Wichelhaus,Helge B. Bode +39 more
TL;DR: This easyPACId approach (easy Promoter Activated Compound Identification) facilitates NP identification due to low interference from other NPs and allows direct bioactivity testing of supernatants containing secreted NPs, without laborious purification.
Journal ArticleDOI
Global analysis of biosynthetic gene clusters reveals conserved and unique natural products in entomopathogenic nematode-symbiotic bacteria
Yi-Ming Shi,Merle Hirschmann,Yan-Ni Shi,Shabbir Ahmed,Desalegne Abebew,Nicholas J. Tobias,Peter Grün,Jan J Crames,Laura Pöschel,Wolfgang Kuttenlochner,Christian Richter,Jennifer Herrmann,Rolf Müller,Aunchalee Thanwisai,Sacha J. Pidot,Timothy P. Stinear,Michael Groll,Yonggyun Kim,Helge B. Bode +18 more
TL;DR: In this paper , the pangenome and sequence similarity networks were used to identify 1,000 BGCs belonging to 176 families and identified the most conserved families are denoted by 11 BGC classes, which are eukaryotic proteasome inhibitors, virulence factors against insects, metallophores and insect immunosuppressants.
Journal ArticleDOI
Integrating genomics and metabolomics for scalable non-ribosomal peptide discovery
Bahar Behsaz,Edna Bode,Alexey Gurevich,Yan-Ni Shi,Florian Grundmann,Deepa D. Acharya,Andrés Mauricio Caraballo-Rodríguez,Amina Bouslimani,Morgan Panitchpakdi,Annabell Linck,Changhui Guan,Julia Oh,Pieter C. Dorrestein,Helge B. Bode,Helge B. Bode,Pavel A. Pevzner,Hosein Mohimani +16 more
TL;DR: NRPminer as discussed by the authors is a modification-tolerant tool for NRP discovery from large (meta)genomic and mass spectrometry datasets, which is able to identify many NRPs from different environments.
