Abstract: Salmonella enterica subspecies 1 serovar Typhimurium is a principal cause of human enterocolitis. For unknown reasons, in mice serovar Typhimurium does not provoke intestinal inflammation but rather targets the gut-associated lymphatic tissues and causes a systemic typhoid-like infection. The lack of a suitable murine model has limited the analysis of the pathogenetic mechanisms of intestinal salmonellosis. We describe here how streptomycin-pretreated mice provide a mouse model for serovar Typhimurium colitis. Serovar Typhimurium colitis in streptomycin-pretreated mice resembles many aspects of the human infection, including epithelial ulceration, edema, induction of intercellular adhesion molecule 1, and massive infiltration of PMN/CD18(+) cells. This pathology is strongly dependent on protein translocation via the serovar Typhimurium SPI1 type III secretion system. Using a lymphotoxin beta-receptor knockout mouse strain that lacks all lymph nodes and organized gut-associated lymphatic tissues, we demonstrate that Peyer's patches and mesenteric lymph nodes are dispensable for the initiation of murine serovar Typhimurium colitis. Our results demonstrate that streptomycin-pretreated mice offer a unique infection model that allows for the first time to use mutants of both the pathogen and the host to study the molecular mechanisms of enteric salmonellosis.

Abstract: More than 3000 per- and polyfluoroalkyl substances (PFASs) are, or have been, on the global market, yet most research and regulation continues to focus on a limited selection of rather well-known long-chain PFASs, particularly perfluorooctanesulfonate (PFOS), perfluorooctanoic acid (PFOA) and their precursors. Continuing to overlook the vast majority of other PFASs is a major concern for society. We provide recommendations for how to proceed with research and cooperation to tackle the vast number of PFASs on the market and in the environment.

Abstract: Amyloids are highly organized cross–β-sheet–rich protein or peptide aggregates that are associated with pathological conditions including Alzheimer’s disease and type II diabetes. However, amyloids may also have a normal biological function, as demonstrated by fungal prions, which are involved in prion replication, and the amyloid protein Pmel17, which is involved in mammalian skin pigmentation. We found that peptide and protein hormones in secretory granules of the endocrine system are stored in an amyloid-like cross–β-sheet–rich conformation. Thus, functional amyloids in the pituitary and other organs can contribute to normal cell and tissue physiology.

Abstract: In the context of phase 5 of the Coupled Model Intercomparison Project, most climate simulations use prescribed atmospheric CO2 concentration and therefore do not interactively include the effect of carbon cycle feedbacks. However, the representative concentration pathway 8.5 (RCP8.5) scenario has additionally been run by earth system models with prescribed CO2 emissions. This paper analyzes the climate projections of 11 earth system models (ESMs) that performed both emission-driven and concentration-driven RCP8.5 simulations. When forced by RCP8.5 CO2 emissions, models simulate a large spread in atmospheric CO2; the simulated 2100 concentrations range between 795 and 1145 ppm. Seven out of the 11 ESMs simulate a larger CO2 (on average by 44 ppm, 985 ± 97 ppm by 2100) and hence higher radiative forcing (by 0.25 W m−2) when driven by CO2 emissions than for the concentration-driven scenarios (941 ppm). Abstract: Symbolic model checking with Binary Decision Diagrams (BDDs) has been successfully used in the last decade for formally verifying finite state systems such as sequential circuits and protocols. Since its introduction in the beginning of the 90's, it has been integrated in the quality assurance process of several major hardware companies. The main bottleneck of this method is that BDDs may grow exponentially, and hence the amount of available memory re- stricts the size of circuits that can be verified efficiently. In this article we survey a technique called Bounded Model Checking (BMC), which uses a propositional SAT solver rather than BDD manipulation techniques. Since its introduction in 1999, BMC has been well received by the industry. It can find many logical er- rors in complex systems that can not be handled by competing techniques, and is therefore widely perceived as a complementary technique to BDD-based model checking. This observation is supported by several independent comparisons that have been published in the last few years.
