Abstract: Large earthquakes produce crustal deformation that can be quantified by geodetic measurements, allowing for the determination of the slip distribution on the fault. We used data from Global Positioning System (GPS) networks in Central Chile to infer the static deformation and the kinematics of the 2010 moment magnitude ( M w) 8.8 Maule megathrust earthquake. From elastic modeling, we found a total rupture length of ~500 kilometers where slip (up to 15 meters) concentrated on two main asperities situated on both sides of the epicenter. We found that rupture reached shallow depths, probably extending up to the trench. Resolvable afterslip occurred in regions of low coseismic slip. The low-frequency hypocenter is relocated 40 kilometers southwest of initial estimates. Rupture propagated bilaterally at about 3.1 kilometers per second, with possible but not fully resolved velocity variations.

Abstract: Autophagy is the degradative process by which eukaryotic cells digest their own components using acid hydrolases within the lysosome. Originally thought to function almost exclusively in providing starving cells with nutrients taken from their own cellular constituents, autophagy is in fact involved in numerous cellular events including differentiation, turnover of macromolecules and organelles, and defense against parasitic invaders. During the last 10-20 years, molecular components of the autophagic machinery have been discovered, revealing a complex interactome of proteins and lipids, which, in a concerted way, induce membrane formation to engulf cellular material and target it for lysosomal degradation. Here, our emphasis is autophagy in protists. We discuss experimental and genomic data indicating that the canonical autophagy machinery characterized in animals and fungi appeared prior to the radiation of major eukaryotic lineages. Moreover, we describe how comparative bioinformatics revealed that this canonical machinery has been subject to moderation, outright loss or elaboration on multiple occasions in protist lineages, most probably as a consequence of diverse lifestyle adaptations. We also review experimental studies illustrating how several pathogenic protists either utilize autophagy mechanisms or manipulate host-cell autophagy in order to establish or maintain infection within a host. The essentiality of autophagy for the pathogenicity of many parasites, and the unique features of some of the autophagy-related proteins involved, suggest possible new targets for drug discovery. Further studies of the molecular details of autophagy in protists will undoubtedly enhance our understanding of the diversity and complexity of this cellular phenomenon and the opportunities it offers as a drug target.

Abstract: It has been previously found that abscisic acid (ABA) participates in the activation of grapevine leaf tissue defense against potentially damaging effects of solar ultraviolet-B radiation (UV-B), apparently by triggering biosynthesis of phenols that filter the harmful radiation and act as antioxidants. The present work studies the effect of solar UV-B and exogenously applied ABA on berry growth, sugar accumulation, and phenol (anthocyanin and nonanthocyanin) profiles across berry development and ripening of Vitis vinifera L. cv. Malbec in a vineyard at 1450 m of altitude. The grapevines were exposed to relatively high UV-B irradiation (normal sunlight; +UV-B) and also to a reduced UV-B treatment (filter exclusion; -UV-B). These two UV-B treatments were combined with weekly spray applications to the leaves and berries of 1 mM ABA (+ABA) or H(2)O (-ABA). Reduction of UV-B delayed berry development and maturation, whereas the +UV-B and +ABA combined treatment hastened berry sugar and phenol accumulation. +UV-B/+ABA treatments also reduced berry growth and decreased sugar per berry without affecting sugar concentration (°Brix) at harvest. Berry skin ABA levels were higher in the +UV-B and +ABA combined treatment, which also hastened the onset of ripening up to 20 days. Berry skin ABA levels then decreased toward harvest, implying a possible role for ABA in the control of ripening in this nonclimacteric fruit. Under both +UV-B and +ABA treatments berry skin phenols were additively increased with a change in anthocyanin and nonanthocyanin profiles and increases in the proportion of phenols with high antioxidant capacity.

Abstract: The identification of melatonin in plants has inspired new investigations to understand its biological function and which endogenous and external factors control its levels in these organisms. Owing to the therapeutical and nutraceutical properties of melatonin, it should be important to develop reliable analytical methods for its quantification in vegetal matrices containing this indoleamine, such as grape and wine. The main objectives of the present study were to test whether melatonin levels fluctuate during the day in berry skins of Vitis vinifera L. cv Malbec, thereby possibly relating its abundance to its putative antioxidant function, to determine whether daylight reaching clusters negatively controls melatonin levels, and to evaluate whether total polyphenols and anthocyanins also change through a 24-hr period. Grapes were harvested throughout the day/night to determine the moment when high levels of these components are present in grapes. The presence of melatonin in grapes was evaluated by high-performance liquid chromatography/electrospray ionization tandem mass spectrometry. It is shown for the first time that melatonin levels fluctuate during the day/night cycle in plants grown under field conditions in a fruit organ of the species Vitis vinifera. We also determined that the diurnal decay of melatonin in berry skins is induced by sunlight, because covered bunches retained higher melatonin levels than exposed ones, thus explaining at least part of the basis of its daily fluctuation. Evidence of melatonin's antioxidant role in grapes is also suggested by monitoring malondialdehyde levels during the day.

Abstract: Rapid and consistent genotyping is an important requirement for cultivar identification in many crop species. Among them grapevine cultivars have been the subject of multiple studies given the large number of synonyms and homonyms generated during many centuries of vegetative multiplication and exchange. Simple sequence repeat (SSR) markers have been preferred until now because of their high level of polymorphism, their codominant nature and their high profile repeatability. However, the rapid application of partial or complete genome sequencing approaches is identifying thousands of single nucleotide polymorphisms (SNP) that can be very useful for such purposes. Although SNP markers are bi-allelic, and therefore not as polymorphic as microsatellites, the high number of loci that can be multiplexed and the possibilities of automation as well as their highly repeatable results under any analytical procedure make them the future markers of choice for any type of genetic identification. We analyzed over 300 SNP in the genome of grapevine using a re-sequencing strategy in a selection of 11 genotypes. Among the identified polymorphisms, we selected 48 SNP spread across all grapevine chromosomes with allele frequencies balanced enough as to provide sufficient information content for genetic identification in grapevine allowing for good genotyping success rate. Marker stability was tested in repeated analyses of a selected group of cultivars obtained worldwide to demonstrate their usefulness in genetic identification. We have selected a set of 48 stable SNP markers with a high discrimination power and a uniform genome distribution (2-3 markers/chromosome), which is proposed as a standard set for grapevine (Vitis vinifera L.) genotyping. Any previous problems derived from microsatellite allele confusion between labs or the need to run reference cultivars to identify allele sizes disappear using this type of marker. Furthermore, because SNP markers are bi-allelic, allele identification and genotype naming are extremely simple and genotypes obtained with different equipments and by different laboratories are always fully comparable.

Abstract: [1] Static offsets produced by the February 27, 2010 Mw = 8.8 Maule, Chile earthquake as measured by GPS and InSAR constrain coseismic slip along a section of the Andean megathrust of dimensions 650 km (in length) × 180 km (in width). GPS data have been collected from both campaign and continuous sites sampling both the near-field and far field. ALOS/PALSAR data from several ascending and descending tracks constrain the near-field crustal deformation. Inversions of the geodetic data for distributed slip on the megathrust reveal a pronounced slip maximum of order 15 m at ∼15–25 km depth on the megathrust offshore Lloca, indicating that seismic slip was greatest north of the epicenter of the bilaterally propagating rupture. A secondary slip maximum appears at depth ∼25 km on the megathrust just west of Concepcion. Coseismic slip is negligible below 35 km depth. Estimates of the seismic moment based on different datasets and modeling approaches vary from 1.8 to 2.6 × 1022 N m. Our study is the first to model the static displacement field using a layered spherical Earth model, allowing us to incorporate both near-field and far-field static displacements in a consistent manner. The obtained seismic moment of 1.97 × 1022 N m, corresponding to a moment magnitude of 8.8, is similar to that obtained by previous seismic and geodetic inversions.

Abstract: Subduction of the Nazca plate beneath South America has driven the growth of the Andes Mountains. Subduction has routinely generated earthquakes larger than magnitude 8.0 along the western margin of the mountain belt 1 , but the potential size of less frequent earthquakes in the eastern, backarc margin is unknown. Continued support of the high Andean Plateau at the centre of the Andes can be explained only if deformation of the backarc margin is ongoing 2‐4 . Here we present GPS data that record surface motions in the Subandean ranges that are part of the backarc margin. We find that the velocity of surface movement decreases sharply from west to east across the Subandean ranges. We suggest that a subhorizontal fault underlying the ranges slips freely at depth in the west, but is locked for up to 100 km in shallower sections further east. Analysis of fault scarps formed where the subhorizontal fault intersects the surface indicates that the fault has generated repeated large earthquakes. We suggest that rupture of the entire locked section of the fault could generate an earthquake of magnitude 8.7‐8.9. We attribute the large seismic potential to the unusual width of the Subandean ranges, and suggest that deformation of the Subandean ranges, at a rate unmatched by erosion, causes the mountain range to widen. The central Andean Plateau (AP) is a bilaterally symmetric plateau of 4;000m average elevation (Fig. 1). Although AP uplift scenarios are currently debated 5,6 it is generally accepted that the backarc orogenic wedge on the AP’s east flank, comprising the Eastern Cordillera (EC), Inter-Andean zone (IAZ), and Subandes (SA), has contracted by more than 250km since 45Myr ago (refs 79). Abstract: This paper describes a simple and inexpensive procedure to produce thin-films of poly(dimethylsiloxane). Such films were characterized by a variety of techniques (ellipsometry, nuclear magnetic resonance, atomic force microscopy, and goniometry) and used to investigate the adsorption kinetics of three model proteins (fibrinogen, collagen type-I, and bovine serum albumin) under different conditions. The information collected from the protein adsorption studies was then used to investigate the adhesion of human dermal microvascular endothelial cells. The results of these studies suggest that these films can be used to model the surface properties of microdevices fabricated with commercial PDMS. Moreover, the paper provides guidelines to efficiently attach cells in BioMEMS devices.

Abstract: Most rare species appear to be specialists in plant-pollinator networks. This observation could result either from real ecological processes or from sampling artifacts. Several methods have been proposed to overcome these artifacts, but they have the limitation of being based on visitation data, causing interactions involving rare visitor species to remain undersampled. We propose the analysis of food composition in bee trap nests to assess the reliability of network specialization estimates. We compared data from a plant-pollinator network in the Monte Desert of Villavicencio Nature Reserve, Argentina, sampled by visit observation, and data from trap nests sampled at the same time and location. Our study shows that trap nest sampling was good for estimating rare species degree. The rare species in the networks appear to be more specialized than they really are, and the bias in the estimation of the species degree increases with the rareness. The low species degree of these rare species in the visitation networks results from insufficient sampling of the rare interactions, which could have important consequences for network structure.

Abstract: In Argentina, human fascioliasis has never been adequately analysed, although having a physiography, climate, animal prevalences and lymnaeids similar to those of countries where the disease is endemic such as Bolivia, Peru and Chile. We performed a literature search identifying 58 reports accounting for 619 cases, involving 13 provinces, their majority (97.7%) from high altitudes, in central mountainous areas and Andean valleys, concentrated in Cordoba (430 cases), Catamarca (73), San Luis (29) and Mendoza (28), the remaining provinces being rarely affected. This distribution does not fit that of animal fascioliasis. Certain aspects (higher prevalence in females in a local survey, although a trend non-significant throughout Argentina) but not others (patient's age 3-95 years, mean 37.1 years) resemble human endemics in Andean countries, although the lack of intensity studies and surveys in rural areas does not allow for an adequate evaluation. Human infection occurs mainly in January-April, when higher precipitation and temperatures interact with field activities during summer holidays. A second June peak may be related to Easter holidays. The main risk factor appears to be wild watercress ingestion (214) during recreational, weekend outings or holiday activities, explaining numerous family outbreaks involving 63 people and infection far away from their homes. Diagnosis mainly relied on egg finding (288), followed by serology (82), intradermal reaction (63), surgery (43), and erratic fluke observation (6). The number of fascioliasis-hydatidosis co-infected patients (14) is outstanding. Emetine appears as the drug most used (186), replaced by triclabendazole in recent years (21). Surgery reports are numerous (27.0%). A long delay in diagnosis (average almost 3.5 years) and high lithiasis proportion suggest that many patients are frequently overlooked and pose a question mark about fascioliasis detection in the country. High seroprevalences found in recent random surveys suggest human endemic situations. This analysis highlights that human fascioliasis may have been overlooked in the past and its real epidemiological situation in high risk rural, mainly altitudinal areas, may currently be underestimated. Results provide a valuable baseline on which to design appropriate multidisciplinary studies on humans, animals and lymnaeids to assess up to which level and in which areas, human fascioliasis may represent a health problem in Argentina.

Abstract: Summary Autophagy has been implicated as part of the innate immune system against different intracellular microorganisms. Mycobacterium marinum is the causative agent of the fish-tank granuloma and has been widely used as an alternative model to study pathogenic mycobacteria. In this report, we show an active interaction of M. marinum with the autophagic protein LC3, an event that requires pathogen viability and bacterial protein synthesis. Interestingly, M. marinum lacking the region of difference 1 (RD1) is unable to recruit LC3, indicating that a functional ESX-1 secretion system is an absolute requirement for this process. In addition, phagocytosis of the bacteria is also a condition for the LC3 rearrangement induced by M. marinum. We present evidence that this pathogen resides temporarily in a LC3-decorated compartment with late endocytic features but mostly devoid of lysosomal enzymes or degradative properties. In addition our results indicate that autophagy induction by rapamycin treatment leads to maturation of the M. marinum-containing compartment.

Abstract: Coxiella burnetii is a Gram-negative obligate intracellular bacterium. After internalization, this bacterium replicates in a large parasitophorous vacuole that has features of both phagolysosomes and autophagosomal compartments. We have previously demonstrated that early after internalization Coxiella phagosomes interact with both the endocytic and the autophagic pathways. In this report, we present evidence that the Coxiella-replicative vacuoles (CRVs) also interact with the secretory pathway. Rab1b is a small GTPase responsible for the anterograde transport between the endoplasmic reticulum and the Golgi apparatus. We present evidence that Rab1b is recruited to the CRV at later infection times (i.e., after 6 h of infection). Interestingly, knockdown of Rab1b altered vacuole growth, indicating that this protein was required for the proper biogenesis of the CRV. In addition, overexpression of the active GTPase-defective mutant (GFP-Rab1b Q67L) affected the development of the Coxiella-replicative compartment inhibiting bacterial growth. On the other hand, disruption of the secretory pathway by brefeldin A treatment or by overexpression of Sar1 T39N, a defective dominant-negative mutant of Sar1, affected the typical spaciousness of the CRVs. Taken together, our results show for the first time that the Coxiella-replicative niche also intercepts the early secretory pathway.

Abstract: This paper informs the characterization by 16SrDNA partial sequence analysis of an endophytic diazotrophic bacterium isolated from roots of the halophyte shrub Prosopis strombulifera. The bacterium produced ABA, IAA, GA1, GA3 and jasmonic acid in chemically-defined culture medium as assessed by GC-EIMS. The results emphasize the role of phytohormones produced by endophytic bacteria in the association host-beneficial microorganisms, especially under conditions of adverse environments.

Abstract: We present the results of XRF analysis for 428 obsidian samples from archaeological sites in the Argentinian province of Mendoza and from central Chile. The archaeological samples come from different environments and have dates that range from 9000 to 300 BP. The results indicate that all known sources were utilized; however, the beginnings and the intensities of their exploitation were variable. On the contrary, strong differences appear, especially between the Cordilleran and the non-Cordillera sources. We suggest that this pattern is mainly related to differences in the accessibility, quality and abundance of the obsidian in the sources.

Abstract: The most frequent case of horizontal transfer in plants involves a group I intron in the mitochondrial gene cox1, which has been acquired via some 80 separate plant-to-plant transfer events among 833 diverse angiosperms examined. This homing intron encodes an endonuclease thought to promote the intron's promiscuous behavior. A promising experimental approach to study endonuclease activity and intron transmission involves somatic cell hybridization, which in plants leads to mitochondrial fusion and genome recombination. However, the cox1 intron has not yet been found in the ideal group for plant somatic genetics - the Solanaceae. We therefore undertook an extensive survey of this family to find members with the intron and to learn more about the evolutionary history of this exceptionally mobile genetic element. Although 409 of the 426 species of Solanaceae examined lack the cox1 intron, it is uniformly present in three phylogenetically disjunct clades. Despite strong overall incongruence of cox1 intron phylogeny with angiosperm phylogeny, two of these clades possess nearly identical intron sequences and are monophyletic in intron phylogeny. These two clades, and possibly the third also, contain a co-conversion tract (CCT) downstream of the intron that is extended relative to all previously recognized CCTs in angiosperm cox1. Re-examination of all published cox1 genes uncovered additional cases of extended co-conversion and identified a rare case of putative intron loss, accompanied by full retention of the CCT. We infer that the cox1 intron was separately and recently acquired by at least three different lineages of Solanaceae. The striking identity of the intron and CCT from two of these lineages suggests that one of these three intron captures may have occurred by a within-family transfer event. This is consistent with previous evidence that horizontal transfer in plants is biased towards phylogenetically local events. The discovery of extended co-conversion suggests that other cox1 conversions may be longer than realized but obscured by the exceptional conservation of plant mitochondrial sequences. Our findings provide further support for the rampant-transfer model of cox1 intron evolution and recommend the Solanaceae as a model system for the experimental analysis of cox1 intron transfer in plants.

Abstract: This study evaluates the antioxidant and the anti-inflammatory properties of garlic (G) and onion (O) in fructose-fed rats (FFR). Thirty-day-old male Wistar rats were assigned to control (C), F (10% fructose in drinking water), F+T (tempol 1 mM as control antioxidant), F+G, and F+O. Aqueous G and O extracts were administered orally in doses of 150 and 400 mg/kg/d respectively, and along with tempol, were given during the last 8 weeks of a 14-week period. At the end of the study, FFR had developed insulin resistance, aortic NADPH oxidase activity, increased SBP, plasma TBARS and vascular cell adhesion molecule-1 (VCAM-1) expression in mesenteric arteries, and a decrease in heart endothelial nitric oxide synthase (eNOS). Garlic and onion administration to F rats reduced oxidative stress, increased eNOS activity, and also attenuated VCAM-1 expression. These results provide new evidence showing the anti-inflammatory and antioxidant effect of these vegetables.

Abstract: Carbon allocation within grapevines may affect berry growth and development. The plant hormones gibberellins (GAs) and abscisic acid (ABA) control various processes across the plant life and both have been involved in assimilate production and transport in different species. Hence, this work examined the distribution of sugars (sucrose, fructose, and glucose) and starch in grapevines at veraison after foliar applications of GA3, ABA, and an inhibitor of GA biosynthesis, paclobutrazol (PBZ). The results demonstrated that GA3 increased total grapevine mass, with carbon allocated to the whole grapevine (as structural and soluble carbohydrates). Both GA3 and ABA increased monosaccharide (glucose and fructose) levels in berries (up to tenfold) and roots (up to threefold). However, GA3 increased the net carbon fixation whereas ABA did not. PBZ diminished most growth parameters except grapevine mass, and allocated more carbohydrates to roots (up to threefold more sucrose and starch). Such results indicate that GAs promote net carbon fixation and transport, whereas ABA as a stress signal only enhances sugar transport; notwithstanding the two hormones promoted carbon allocation toward roots and berries.

Abstract: Perturbation of renal tubular antioxidants and overproduction of reactive oxygen species may amplify the proinflammatory state of renal obstruction, culminating in oxidative stress and tubular loss. Here, we analyzed the heat shock protein 70 (Hsp70) response and the function and signal transduction of NF-E2-related protein 2 (Nrf2) transcription factor on oxidative stress modulation in obstruction. Rats were subjected to unilateral ureteral obstruction or sham operation and kidneys harvested at 5, 7, 10, and 14 days after obstruction. Hsp70 expression and Nrf2 activity and its downstream target gene products were assessed. After 10 and 14 days of obstruction, enhanced lipid peroxidation through higher thiobarbituric acid reactive substances levels and increased oxidative stress resulted in reduced total antioxidant activity and enhanced nicotinamide adenine dinucleotide phosphate reduced (NADPH) oxidase activity were demonstrated. This was accompanied by decreased inducible Hsp70 expression and a progressive reduction of nuclear Nrf2 and its target gene products glutathione S-transferase A2 (GSTA2) and NADPH/quinone oxidoreductase 1 (NQO1), whereas the Nrf2 repressor Kelch-like ECH-associated protein-1 (Keap1) was upregulated. By contrast, on early obstruction for 7 days, lack of increased oxidative markers associated with higher inducible Hsp70 protein levels and a rapid nuclear accumulation of Nrf2, Keap1 downregulation, and mRNA induction of the identified Nrf2-dependent genes, NQO1 and GSTA2, were shown. For these results, we suggest that the magnitude of cytoprotection in early obstruction depends on the combined contribution of induced activation of Nrf2 upregulating its downstream gene products and Hsp70 response. Impaired ability to mount the biological response to the prevailing oxidative stress leading to renal injury was shown in prolonged obstruction.

Abstract: Two autochthonous yeasts from the northwest region of Argentina, Kloeckera apiculata mc1 and Saccharomyces cerevisiae mc2, were used as pure or mixed starter cultures in microvinification trials conducted in Malbec red must. Also, the effect of Oenococcus oeni X2L was evaluated. S. cerevisiae mc2 showed adequate growth and fermentative activity in single and composite fermentations, producing standard concentration of ethanol. The amount of esters was higher in fermentations conducted using mixed yeast starters. Independent of the timing of inoculation of O. oeni, this malolactic bacterium completely depleted malic acid. Sensory evaluation indicated that young wines fermented with mixed yeast cultures and sequential inoculation of O. oeni were preferred, achieving the highest scores for positive descriptors and they allowed better control of the sensory quality. Consequently, this study proposes inclusion of autochthonous K. apiculata mc1 as an adjunct culture to S. cerevisiae mc2 during Malbec must fermentation to improve the organoleptic properties of red wines. Furthermore, sequential inoculation of O. oeni X2L should be carried out after completion of the alcoholic fermentation to enhance sensory characteristics.

Abstract: In this review, we examine the plant group Solanum sect. Lycopersicon – a clade of 13 species, including the domesticated tomato (Solanum lycopersicum L.) and its wild relatives – along with four allied species in the immediate outgroups Solanum sects. Juglandifolia and Lycopersicoides. We summarize the geographic distribution and morphological characters of these plant groups, describing their evolutionary relationships in the context of a new taxonomic revision at the species level of all these groups. We provide an overview of the role that wild tomato species have played in the development of cytogenetic stocks, in classical and molecular genetic studies as well as in crop improvement through traditional and advanced tools. We discuss how the very narrow genetic basis of cultivated tomato germplasm has forced tomato geneticists and breeders to rely on the wealth of genetic variation present in the wild relatives to address the many breeding challenges. The numerous molecular mapping studies conducted using interspecific crosses have clearly demonstrated that the breeding value of exotic (wild) tomato germplasm goes far beyond its phenotype. These studies also show that we are still far from being able to fully exploit the breeding potential of the thousands of accessions stored in seed banks around the world, in addition to those that may still be found in natural habitats. Over the past decades, tomato breeders have been at the forefront of establishing new principles for crop breeding based on the use of wild species to improve modern cultivars. In this respect, among all model systems, the wild and domesticated species of the tomato clade have pioneered development of novel populations such as “exotic libraries.” These genetic resources, combined with the increasing knowledge deriving from the many “omics” tools, including the tomato genome sequence, are expected to further improve the efficiency with which wild tomato relatives will contribute to the improvement of this important crop.

Abstract: A simple technique based on ultrasound-assisted emulsification microextraction in situ derivatization (USAEME-ISD) is proposed for the one-step derivatization, extraction, and preconcentration of bisphenol A (BPA) in beverage samples prior to gas chromatography-mass spectrometry (GC-MS) analysis. BPA was in situ derivatized with acetic anhydride and simultaneously extracted and preconcentrated by using USAEME. Variables affecting the extraction efficiency of BPA were evaluated. Under optimal experimental conditions, the detection limit (LOD) was 38 ng L(-1) with a relative standard deviation (RSD) value of 11.6%. The linear working range was 100-1250 ng L(-1), and the coefficient of estimation (r(2)) of the calibration curve was ≥0.9971. The robustness of the proposed methodology was probed by developing a recovery study at two concentrations (125 and 500 ng L(-1)) over different beverage samples. This study led to a satisfactory result achieving recoveries of ≥82%, which showed acceptable robustness for determination of nanograms per liter of BPA in samples of food safety interest.

Abstract: In the present work, an ionic liquid (IL) lighter than water was employed as extraction solvent in a dispersive liquid-liquid microextraction (DLLME) methodology. An original flow injection system for online microextraction and preconcentration of cobalt (Co) based on the use of tetradecyl(trihexyl)phosphonium chloride (CYPHOS® IL 101) was designed. Cobalt was complexed with 4-(2-pyridylazo)-resorcinol (PAR) reagent at pH 4.8 and then, the IL-DLLME procedure was developed by dispersing CYPHOS® IL 101 with acetone in an aqueous solution containing Co-PAR complex. Different pyridylazo dyes were evaluated for Co preconcentration in terms of their molecular structure, stability and acid-base equilibrium. Online extraction of Co-PAR into the IL and separation of the dispersed IL enriched phase were accomplished with a microcolumn containing Florisil material. Cobalt was removed from the microcolumn with a 10% (v/v) HNO3 acidified-acetone solution and finally measured by electrothermal atomic absorption spectrometry (ETAAS). The detection limit achieved after preconcentration of 2 mL of sample solution was 8 ng L-1. The precision for 10 replicate determinations at the 1 µg L-1 Co level was 5.1% relative standard deviation (RSD), calculated from the peak heights obtained. The method was successfully applied to Co determination in water samples as well as ophthalmic and parenteral solutions. For the first time, an IL-based microextraction technique was applied for metal determination in these complex samples, where Co recovery varied between 97.9 and 103%.
