Abstract: We investigate the effects of 12-week interval training of moderate- or high-intensity exercise on blood lipids and plasma levels of adiponectin. Thirty-four obese adolescent females [age = 15.9 ± 0.3 years; BMI and BMI-Z-score = 30.8 ± 1.6 kg/m2 and 3 ± 0.3, respectively], were randomized to high-intensity interval training (HIIT, n = 11), moderate-intensity interval training (MIIT, n = 11), or a control group (CG, n = 12). Maximal oxygen uptake ( $$\mathop V\limits^{.} {\text{O}}_{{2{\text{peak}}}}$$ ), maximal aerobic speed (MAS), plasma lipids and adiponectin levels were measured in all subjects before and after training. Following the training program, in both training groups, body mass, BMI-Z-score, and percentage body fat (% BF) decreased, while $$\mathop V\limits^{.} {\text{O}}_{{2{\text{peak}}}}$$ and MAS increased. Low-density lipoprotein cholesterol, high-density lipoprotein cholesterol, and adiponectin levels were positively altered (−12.6 and −7.4 %; 6.3 and 8.0 %; 35.8 and 16.2 %; high to moderate training program, respectively). Waist circumference, triglyceride and total cholesterol decreased only in HIIT group (−3.5; −5.3 and −7.0 %, respectively, in all P < 0.05). Significant decrease in the usual index of insulin resistance (HOMA-IR) occurred in HIIT and MIIT groups (−29.2 ± 5.3 and −18.4 ± 8.6 %, respectively; P < 0.01). The results show that HIIT positively changes blood lipids and adiponectin variables in obese adolescent girls, resulting in improved insulin sensitivity, as attested by a lower HOMA-IR, and achieving better results compared to moderate-intensity exercise.

Abstract: Cell death is a fundamental ingredient of life Thus, not surprisingly more than one form of cell death exists Several excellent reviews on various forms of cell death have already been published but manuscripts describing interconnection and interdependence between such processes are uncommon Here, what follows is a brief introduction on all three classical forms of cell death, followed by a more detailed insight into the role of p53, the master regulator of apoptosis, and other forms of cell death While discussing p53 and also the role of caspases in cell death forms, we offer insight into the interplay between autophagy and apoptosis, or necrosis, where autophagy may initially serve pro-survival functions The review moves further to present some details about less researched forms of programmed cell death, namely necroptosis, necrosis and mitoptosis These “mixed” forms of cell death allow us to highlight the interconnected nature of cell death forms, particularly apoptosis and necrosis The interdependence between apoptosis, autophagy and necrosis, and their significance for cancer development and treatment are also analyzed in further parts of the review In the concluding parts, the afore-mentioned issues will be put in perspective for the development of novel anti-cancer therapies

Abstract: Soil salinity and drought are among the environmental stresses that most severely affect plant growth and production around the world. In this study the rhizospheres of Salicornia plants and bulk soils were collected from Sebkhet and Chott hypersaline ecosystems in Tunisia. Depiction of bacterial microbiome composition by Denaturing Gradient Gel Electrophoresis unveiled the occurrence of a high bacterial diversity associated with Salicornia root system. A large collection of 475 halophilic and halotolerant bacteria was established from Salicornia rhizosphere and the surrounding bulk soil, and the bacteria were characterized for the resistance to temperature, osmotic and saline stresses, and plant growth promotion (PGP) features. Twenty Halomonas strains showed resistance to a wide set of abiotic stresses and were able to perform different PGP activities in vitro at 5% NaCl, including ammonia and indole-3-acetic acid production, phosphate solubilisation, and potential nitrogen fixation. By using a gfp-labelled strain it was possible to demonstrate that Halomonas is capable of successfully colonising Salicornia roots in the laboratory conditions. Our results indicated that the culturable halophilic/halotolerant bacteria inhabiting salty and arid ecosystems have a potential to contribute to promoting plant growth under the harsh salinity and drought conditions. These halophilic/halotolerant strains could be exploited in biofertilizer formulates to sustain crop production in degraded and arid lands.

Abstract: Raman spectra of potassium, sodium, and ammonium sulfates (K2SO4, Na2SO4, and (NH4)2SO4) are reported and analyzed. These sulfates have been investigated under two states: solid (anhydrous and hydrated) salts and aqueous solutions. The effects of monovalent ions (K+ , Na+ , and NH4+) and hydration on the position of Raman lines assigned to internal vibrations of sulfate anion SO42- are discussed. In solid salts, the line position of each Raman peak is shown to decrease with increasing radius of the cation. The main ν1 mode of sulfate molecule is particularly affected. It is emphasized that this sensitivity in solid sulfates vanishes in aqueous solutions. As a consequence, this mode can be probed by Raman spectroscopy as the main signature of SO42- to determine its concentration within a single calibration.

Abstract: BACKGROUND Mimiviridae Mimivirus, including the largest known viruses, multiply in amoebae. Mimiviruses have been linked to pneumonia, but they have never been isolated from patients. To further understand the pathogenic role of these viruses, we aimed to isolate them from a patient presenting with pneumonia. METHODS We cultured, on Acanthamoeba polyphaga amoebae, pulmonary samples from 196 Tunisian patients with community-acquired pneumonia during the period 2009-2010. An improved technique was used for Mimivirus isolation, which used agar plates where the growth of giant viruses is revealed by the formation of lysis plaques. Mimivirus serology was tested by microimmunofluorescence and by bidimensional immunoproteomic analysis using Mimivirus strains, to identify specific immunoreactive proteins. The new Mimivirus strain genome sequencing was performed on Roche 454 GS FLX Titanium, then AB SOLiD instruments. RESULTS We successfully isolated a Mimivirus (LBA111), the largest virus ever isolated in a human sample, from a 72-year-old woman presenting with pneumonia. Electron microscopy revealed a Mimivirus-like virion with a size of 554 ± 10 nm. The LBA111 genome is 1.23 megabases, and it is closely related to that of Megavirus chilensis. Furthermore, the serum from the patient reacted specifically to the virus compared to controls. CONCLUSIONS This is the first Mimivirus isolated from a human specimen. The findings presented above together with previous works establish that mimiviruses can be associated with pneumonia. The common occurrence of these viruses in water and soil makes them probable global agents that are worthy of investigation.

Abstract: The association between plant and plant growth promoting bacteria (PGPB) contributes to the successful thriving of plants in extreme environments featured by water shortage. We have recently shown that, with respect to the non-cultivated desert soil, the rhizosphere of pepper plants cultivated under desert farming hosts PGPB communities that are endowed with a large portfolio of PGP traits. Pepper plants exposed to bacterial isolates from plants cultivated under desert farming exhibited a higher tolerance to water shortage, compared with untreated control. This promotion was mediated by a larger root system (up to 40%), stimulated by the bacteria, that enhanced plant ability to uptake water from dry soil. We provide initial evidence that the nature of the interaction can have a limited level of specificity and that PGPB isolates may determine resistance to water stress in plants others than the one of the original isolation. Abstract: Conservation agriculture based on no-tillage (CA/NT) becomes a current tendency to enhance crop productivity by improving: (i) soil quality, (ii) water management and (iii) organic supply. In Tunisia, the applicability of CA was poorly documented. This study was carried out to evaluate changes in soil organic matter (SOM) and water properties generated by seven (NT7) and three (NT3) years of applying NT as compared to conventional tillage (CT). On three adjacent large plots, a field experiment was established since the growing season 2000/01 under rain-fed conditions on a clay loam soil in a dry subhumid zone of North Tunisia. In NT3 plot, a cultivation of wheat/fava bean sequence was investigated and residues biomass of 500 kg ha−1 and 450 kg ha−1 were left on the soil surface for wheat and fava bean, respectively. In NT7 plot, a cultivation of wheat/sulla sequence was investigated and residues biomass of 500 kg ha−1 and 700 kg ha−1 were left on the soil surface for wheat and sulla, respectively. However for the CT plot, a moldboard plowing at approximately 20 cm depth was managed for continuous wheat production. The selected key parameters evaluated for a 0–30 cm soil profile were SOM, bulk density (BD), total porosity (TP), macro-porosity (MP), micro-porosity (mP), soil water retention at field capacity (FC) and at permanent wilting point (PWP), available water content (AWC) and soil moisture content (SMC). NT7 have increased significantly (p < 0.05) levels of SOM at the top 30 cm soil layer, which in turn enhanced BD and TP of soil as compared to CT. However, improvement of these parameters was restricted to 0–10 cm layer with NT3. Both NT3 and NT7 have increased considerably (p < 0.05) mP at 0–30 cm thereby increasing soil water content at FC and PWP, whereas MP was remarkably reduced by NT treatments as compared to CT but reduction was less important in NT7 plot. AWC was significantly (p < 0.05) higher in NT7 across the 30 cm profile while NT3 and CT have similar effect to each other. SMC was improved at different soil depths by NT system during the whole agricultural season and the highest content was found with NT3. Soil moisture values increased with increasing soil depth in both NT3 and NT7 indicating a good water infiltration induced by these treatments. Hence, water storage properties in rooting zone could be enhanced by NT7 and NT3 but water availability for plants was more important with NT7.

Abstract: The antioxidant activity and the chemical composition of methanol extracts from peel and pulp belonging to two species of Tunisian prickly pears Opuntia ficus indica (spiny and thornless forms) and Opuntia stricta have been studied. The antioxidant capacity was measured by DPPH radical scavenging activity. The total phenolic compound (TPC) and the total flavonoid content were determined by the Folin-Ciocalteu method and colorimetric method, respectively. The phenolic compounds were identified and quantified by high-performance liquid chromatography (HPLC) coupled with an electrospray ionization mass spectrometry (ESI-MS). The results showed that O. stricta fruits present the best antioxidant activities than the two forms of O. ficus indica, while the TPC was more important in O. ficus indica than in the O. stricta fruits. The peels have higher flavonoids than pulp, and the thornless variety has more flavonoid than the spiny. The RP-HPLC and ESI-MS analysis detected two classes of phenolic compounds and betalain pigments. Isorhamnetin derivatives are the dominant flavonol glycoside identified in O. ficus indica (spiny: 65.25 μg·g(-1); thornless: 77.03 μg·g(-1)) and O. stricta peels (19.22 μg·g(-1)).

Abstract: Since the establishment of the ‘Tunisian Chemical Group’ in Gabes city, Tunisia, no serious investigations were carried out about the heavy metal pollution in Gabes gulf. In the present study, the contents of four heavy metals were assessed in four mollusk species (two gastropods, Gibbula ardens and Patella caerulea , and two bivalves, Pinctada radiata and Pinna nobilis ), collected from twelve coastal stations. The results obtained showed generally that high concentrations of heavy metals were recorded in the central area of Gabes gulf, nearer to Gabes city; the low concentrations were in contrast found at the edges of this gulf, which is probably due to the chemical pollution generated from the huge industry of phosphoric acid in Gabes city. Comparing the results found with the four examined species, the lowest concentrations were noted with the two bivalve species P. radiata and P. nobilis . The highest heavy metals’ concentrations, noted during this study, are comparable to the findings of other authors in other areas, but they are considerably beyond the standards. A remediation action is necessarily needed either by reducing the amount of heavy metals in the phosphgypums quantities thrown directly in the sea or by stopping completely this harmful industry to protect the marine life in the area. Abstract: BACKGROUND: Leishmania (L.) are intracellular protozoan parasites able to survive and replicate in the hostile phagolysosomal environment of infected macrophages. They cause leishmaniasis, a heterogeneous group of worldwide-distributed affections, representing a paradigm of neglected diseases that are mainly embedded in impoverished populations. To establish successful infection and ensure their own survival, Leishmania have developed sophisticated strategies to subvert the host macrophage responses. Despite a wealth of gained crucial information, these strategies still remain poorly understood. MicroRNAs (miRNAs), an evolutionarily conserved class of endogenous 22-nucleotide non-coding RNAs, are described to participate in the regulation of almost every cellular process investigated so far. They regulate the expression of target genes both at the levels of mRNA stability and translation; changes in their expression have a profound effect on their target transcripts. METHODOLOGY/PRINCIPAL FINDINGS: We report in this study a comprehensive analysis of miRNA expression profiles in L. major-infected human primary macrophages of three healthy donors assessed at different time-points post-infection (three to 24 h). We show that expression of 64 out of 365 analyzed miRNAs was consistently deregulated upon infection with the same trends in all donors. Among these, several are known to be induced by TLR-dependent responses. GO enrichment analysis of experimentally validated miRNA-targeted genes revealed that several pathways and molecular functions were disturbed upon parasite infection. Finally, following parasite infection, miR-210 abundance was enhanced in HIF-1α-dependent manner, though it did not contribute to inhibiting anti-apoptotic pathways through pro-apoptotic caspase-3 regulation. CONCLUSIONS/SIGNIFICANCE: Our data suggest that alteration in miRNA levels likely plays an important role in regulating macrophage functions following L. major infection. These results could contribute to better understanding of the dynamics of gene expression in host cells during leishmaniasis.

Abstract: blaVEB-1 is an integron-located extended-spectrum β-lactamase gene initially detected in Escherichia coli and Pseudomonas aeruginosa strains from south-east Asia. Several recent studies have reported that VEB-1-positive strains are highly resistant to ceftazidime, cefotaxime and aztreonam antibiotics. One strategy to overcome resistance involves administering antibiotics together with β-lactamase inhibitors during the treatment of infectious diseases. During this study, four VEB-1 β-lactamase inhibitors were identified using computer-aided drug design. The SWISS-MODEL tool was utilized to generate three dimensional structures of VEB-1 β-lactamase, and the 3D model VEB-1 was verified using PROCHECK, ERRAT and VERIFY 3D programs. Virtual screening was performed by docking inhibitors obtained from the ZINC Database to the active site of the VEB-1 protein using AutoDock Vina software. Homology modeling studies were performed to obtain a three-dimensional structure of VEB-1 β-lactamase. The generated model was validated, and virtual screening of a large chemical ligand library with docking simulations was performed using AutoDock software with the ZINC database. On the basis of the dock-score, four molecules were subjected to ADME/TOX analysis, with ZINC4085364 emerging as the most potent inhibitor of the VEB-1 β-lactamase.

Abstract: Whether sex chromosomes are differentiated is an important aspect of our knowledge of dioecious plants, such as date palm (Phoenix dactylifera). In this crop plant, the female individuals produce dates, and are thus the more valuable sex. However, there is no way to identify the sex of date palm plants before reproductive age, and the sex-determining mechanism is still unclear. To identify sex-linked microsatellite markers, we surveyed a set of 52 male and 55 female genotypes representing the geographical diversity of the species. We found three genetically linked loci that are heterozygous only in males. Male-specific alleles allowed us to identify the gender in 100% of individuals. These results confirm the existence of an XY chromosomal system with a nonrecombining XY-like region in the date palm genome. The distribution of Y haplotypes in western and eastern haplogroups allowed us to trace two male ancestral paternal lineages that account for all known Y diversity in date palm. The very low diversity associated with Y haplotypes is consistent with clonal paternal transmission of a nonrecombining male-determining region. Our results establish the date palm as a biological model with one of the most ancient sex chromosomes in flowering plants.

Abstract: The localised corrosion resistance of austenitic stainless steels is strongly influenced by the quality of finished surface. EDM machining induces substantial changes by the high thermal gradients generated by electric sparks. Experimental techniques such as roughness measurement, scanning electron microscopy (SEM), energy dispersive microanalysis (EDX) and X-ray diffraction technique, reveal micro-geometrical, microstructural, chemical and mechanical changes. These changes lead to white and heat-affected layers with a depth less than 100 μm. The white layer is a melted material characterised by dendritic structure and constituted by austenite, chromium carbide and e-carbide. The heat-affected layer is characterised by very large grain size comparatively to the bulk material. Electrochemical test coupled with metallographic examinations using SEM reveals a weakening of the resistance to pitting and intergranular corrosion comparatively to diamond polished surface. This weakening is correlated to differences in structure and chemical composition of white layer. Susceptibility to stress corrosion cracking has been attributed to the field of tensile residual stresses resulting from thermal effects. The removal of the white layer material by polishing or wire brushing restores the corrosion resistance of the AISI316L SS.

Abstract: Through its impact on photosynthesis and morphogenesis, light is the environmental factor that most affects plant architecture. Using light rather than chemicals to manage plant architecture could reduce the impact on the environment. However, the understanding of how light modulates plant architecture is still poor and further research is needed. To address this question, we examined the development of two rose cultivars, Rosa hybrida'Radrazz' and Rosa chinensis'Old Blush', cultivated under two light qualities. Plants were grown from one-node cuttings for 6 weeks under white or blue light at equal photosynthetic efficiencies. While plant development was totally inhibited in darkness, blue light could sustain full development from bud burst until flowering. Blue light reduced the net CO(2) assimilation rate of fully expanded leaves in both cultivars, despite increasing stomatal conductance and intercellular CO(2) concentrations. In 'Radrazz', the reduction in CO(2) assimilation under blue light was related to a decrease in photosynthetic pigment content, while in both cultivars, the chl a/b ratio increased. Surprisingly, blue light could induce the same organogenetic activity of the shoot apical meristem, growth of the metamers and flower development as white light. The normal development of rose plants under blue light reveals the strong adaptive properties of rose plants to their light environment. It also indicates that photomorphogenetic processes can all be triggered by blue wavelengths and that despite a lower assimilation rate, blue light can provide sufficient energy via photosynthesis to sustain normal growth and development in roses.

Abstract: Aim To compare efficacy (intraocular pressure (IOP) reduction) and safety of preservative-free latanoprost (T2345) to benzalkonium chloride (BAK)-preserved latanoprost (BPL; Xalatan) in ocular hypertension (OHT) or primary open angle glaucoma (POAG) patients. Methods Prospective, international, multicentre, randomised, investigator-masked, parallel–group trial. After a wash-out period, POAG or OHT patients, previously managed by BPL monotherapy, randomly received T2345 or BPL (one drop into the affected eye(s)) once daily from D0 to D84. Change in IOP was measured at 09:00 (±1 h) from D0 to D84 in the worse eye. Results Mean IOP reduction (D0–D84) was −8.6±2.6 mm Hg (−36%) on T2345 and −9.0±2.4 mm Hg (−38%) on BPL, confirming non-inferiority of T2345 to BPL. Non-inferiority of T2345 was observed from D15. The most frequent ocular adverse event, drug intolerance, was reported in 1 (0.5%) patient on T2345 versus 4 (2.1%) patients on BPL. Moderate to severe conjunctival hyperaemia was less frequent on T2345 than on BPL at D42 (20.2% vs 30.6%; p=0.003) and D84 (21.4% vs 29.1%; p=0.02). Upon instillation, the global subjective ocular symptom score was significantly lower on T2345 than BPL on D42 (0.15 vs 0.41; p=0.001) and D84 (0.18 vs 0.46; p=0.001). Conclusions Preservative-free latanoprost has the same efficacy as BPL, with improved local tolerance.

Abstract: Fas/Fas ligand (FasL) system is one of the key apoptotic signaling entities in the extrinsic apoptotic pathway. De-regulation of this pathway, i.e. by mutations may prevent the immune system from the removal of newly-formed tumor cells, and thus lead to tumor formation. The present study investigated the association between −1377 G/A (rs2234767) and −670 A/G (rs1800682) polymorphisms in Fas as well as single nucleotide polymorphisms INV2nt −124 A/G (rs5030772) and −844 C/T (rs763110) in FasL in a sample of Iranian patients with breast cancer. This case-control study was done on 134 breast cancer patients and 152 normal women. Genomic DNA was extracted from whole blood samples. The polymorphisms were determined by using tetra-ARMS-PCR method. There was no significant difference in the genotype distribution of FAS rs2234767 polymorphism between cases and controls. FAS rs1800682, FASL rs5030772, and FASL rs763110 genotypes showed significant associations with an increasing risk of breast cancer (odds ratio OR = 3.18, P = 0.019; OR = 5.08, P = 0.012; OR = 2.40, P = 0.024, respectively). In conclusion, FAS rs2234767 was not associated with breast cancer risk. Though, FAS rs1800682, FASL rs5030772, and FASL rs763110 polymorphisms were associated with the risk of breast cancer in the examined population.

Abstract: Detection and identification of induction machine faults through the stator current signal using higher order spectra analysis is presented. This technique is known as motor current signature analysis (MCSA). This paper proposes two higher order spectra techniques, namely the power spectrum and the slices of bi-spectrum used for the analysis of induction machine stator current leading to the detection of electrical failures within the rotor cage. The method has been tested by using both healthy and broken rotor bars cases for an 18.5 kW-220 V/380 V-50 Hz-2 pair of poles induction motor under different load conditions. Experimental signals have been analyzed highlighting that bi-spectrum results show their superiority in the accurate detection of rotor broken bars. Even when the induction machine is rotating at a low level of shaft load (no-load condition), the rotor fault detection is efficient. We will also demonstrate through the analysis and experimental verification, that our proposed proposed-method has better detection performance in terms of receiver operation characteristics (ROC) curves and precision-recall graph.

Abstract: Purpose – The aim of this article is to examine perceptions and expectations of two major stakeholders: students and employers of the importance of skills and attributes for securing entry-level employment in accounting. Design/methodology/approach – The authors conduct an empirical study using a list of 78 items, starting by comparing employers’ expectations with students’ perceptions concerning the set of technical and non technical skills required from accounting graduates to pursue a career in the accounting profession This study involving data collection from 81 accounting students in five business schools in Tunisia and 48 practitioners. Findings – Findings indicate that employers are seeking graduates who possess a diverse range of non-technical skills. However, accounting students perceive that it is rather technical skills which determine their abilities to pursue a career in the accounting profession. Practical implications – The paper’s findings are of interest to several different parties, primarily the professional accounting body, universities and students. Originality/value – The paper describes an empirical study, conducted on a developing country, which provides a better understanding of new skills expected for future accountants in this changing business environment. The paper constitutes also a meaningful contribution to the accounting education literature, as it examines an interesting subject that has not yet been investigated in the Tunisian context.

Abstract: A total of 119 lactic acid bacteria (LAB) were isolated, by culture-dependant method, from rhizosphere samples of olive trees and desert truffles and evaluated for different biotechnological properties. Using the variability of the intergenic spacer 16S-23S and 16S rRNA gene sequences, the isolates were identified as the genera Lactococcus, Pediococcus, Lactobacillus, Weissella, and Enterococcus. All the strains showed proteolytic activity with variable rates 42% were EPS producers, while only 10% showed the ability to grow in 9% NaCl. In addition, a low rate of antibiotic resistance was detected among rhizospheric enterococci. Furthermore, a strong antibacterial activity against plant and/or pathogenic bacteria of Stenotrophomonas maltophilia, Pantoea agglomerans, Pseudomonas savastanoi, the food-borne Staphylococcus aureus, and Listeria monocytogenes was recorded. Antifungal activity evaluation showed that Botrytis cinerea was the most inhibited fungus followed by Penicillium expansum, Verticillium dahliae, and Aspergillus niger. Most of the active strains belonged to the genera Enterococcus and Weissella. This study led to suggest that environmental-derived LAB strains could be selected for technological application to control pathogenic bacteria and to protect food safety from postharvest deleterious microbiota.
