High-throughput fingerprinting of human pluripotent stem cell fate responses and lineage bias.

TLDR
A high-throughput platform to screen hPSCs in configurable microenvironments in which it is demonstrated that short-term responses predict definitive endoderm induction efficiency and can be used to rescue differentiation of cell lines reticent to cardiac induction.
Abstract
This paper describes a platform for high-throughput image-based profiling of human pluripotent stem cells (hPSCs). It is used to quantify differentiation bias and endogenous signaling levels in hPSC lines.

Quality cell therapy manufacturing by design

TL;DR: It is argued here that a solution to this challenge is to design cell manufacturing processes according to quality-by-design (QbD) principles, which integrates scientific knowledge and risk analysis into manufacturing process development and is already being adopted by the biopharmaceutical industry.

A stepwise model of reaction-diffusion and positional information governs self-organized human peri-gastrulation-like patterning

TL;DR: A high-throughput in vitro system allowing the induction of peri-gastrulation-like fates in geometrically confined hPSC colonies reveals that a two-step process underlies the observed self-organization and subsequent fate acquisition.

A Simple Statistical Parameter for Use in Evaluation and Validation of High Throughput Screening Assays.

TL;DR: A screening window coefficient, called "Z- factor," is defined, which is reflective of both the assay signal dynamic range and the data variation associated with the signal measurements, and therefore is suitable for assay quality assessment.

Core transcriptional regulatory circuitry in human embryonic stem cells.

TL;DR: Insight is provided into the transcriptional regulation of stem cells and how OCT4, SOX2, and NANOG contribute to pluripotency and self-renewal and how they collaborate to form regulatory circuitry consisting of autoregulatory and feedforward loops.

Cell shape, cytoskeletal tension, and rhoa regulate stem cell lineage commitment

TL;DR: It is demonstrated that cell shape regulates commitment of human mesenchymal stem cells to adipocyte or osteoblast fate and mechanical cues experienced in developmental and adult contexts, embodied by cell shape, cytoskeletal tension, and RhoA signaling, are integral to the commitment of stem cell fate.

Highly efficient neural conversion of human ES and iPS cells by dual inhibition of SMAD signaling

TL;DR: Noggin/SB431542-based neural induction should facilitate the use of hES and hiPS cells in regenerative medicine and disease modeling and obviate the need for protocols based on stromal feeders or embryoid bodies.

Maintenance of pluripotency in human and mouse embryonic stem cells through activation of Wnt signaling by a pharmacological GSK-3-specific inhibitor

TL;DR: It is shown that activation of the canonical Wnt pathway is sufficient to maintain self-renewal of both HESCs and MESCs, and the use of GSK-3-specific inhibitors such as BIO may have practical applications in regenerative medicine.
