Journal ArticleDOI
Mutation in the α-synuclein gene identified in families with Parkinson's disease
Mihael H. Polymeropoulos,Christian Lavedan,Elisabeth Leroy,Susan E. Ide,Anindya Dehejia,Amalia Dutra,Brian L. Pike,Holly Root,Jeffrey Rubenstein,Rebecca Boyer,Edward S. Stenroos,Settara C. Chandrasekharappa,Aglaia Athanassiadou,Theodore Papapetropoulos,William G. Johnson,Alice Lazzarini,Roger C. Duvoisin,Giuseppe Di Iorio,Lawrence I. Golbe,Robert L. Nussbaum +19 more
TL;DR: A mutation was identified in the α-synuclein gene, which codes for a presynaptic protein thought to be involved in neuronal plasticity, in the Italian kindred and in three unrelated families of Greek origin with autosomal dominant inheritance for the PD phenotype.
Journal ArticleDOI
A clinical genetic study of Parkinson's disease: evidence for dominant transmission.
Alice Lazzarini,Richard H. Myers,T. R. Zimmerman,Margery H. Mark,Lawrence I. Golbe,Jacob I. Sage,William G. Johnson,Roger C. Duvoisin +7 more
TL;DR: The cumulative risk of Parkinson's disease in relatives of patients seen consecutively for 1 year and the proportion of secondary cases of PD as a function of pedigree completeness was assessed, suggesting significant familial aggregation in a subset of randomly ascertained families.
Journal ArticleDOI
Restless Legs Syndrome Confirmation of Linkage to Chromosome 12q, Genetic Heterogeneity, and Evidence of Complexity
Alex Desautels,Gustavo Turecki,Jacques Montplaisir,Lan Xiong,Arthur S. Walters,Bruce L. Ehrenberg,Kateri Brisebois,Amelie K. Desautels,Yves Gingras,William G. Johnson,Elio Lugaresi,Giorgio Coccagna,Daniel L. Picchietti,Alice Lazzarini,Guy A. Rouleau +14 more
TL;DR: The results support the presence of a major restless legs syndrome-susceptibility locus on chromosome 12q, which has been designated as RLS1, and suggest that at least one additional locus may be involved in the origin of this prevalent condition.
Journal ArticleDOI
The relationship between CAG repeat length and age of onset differs for Huntington's disease patients with juvenile onset or adult onset.

Adrienne Cupples,Marie Saint-Hilaire,Steven M. Hersch +96 more
TL;DR: In this article, a two-segment exponential regression model was proposed for age-of-obstinence analysis of the CAG repeat expansion in the HD gene, and a plot of natural log-transformed age of onset against CAG-repeat length revealed this segmental relationship.
Journal ArticleDOI
Studies of penetrance and anticipation in five autosomal-dominant restless legs syndrome pedigrees.
Alice Lazzarini,Alice Lazzarini,Arthur S. Walters,Arthur S. Walters,Kathleen Hickey,Giorgio Coccagna,Elio Lugaresi,Bruce L. Ehrenberg,Daniel L. Picchietti,Mitchell F. Brin,E. Scot Stenroos,Tracy Verrico,William G. Johnson +12 more
TL;DR: In two of the five analyzed pedigrees, there is statistical support for anticipation and variations in penetrance and anticipation suggest possible genetic heterogeneity.
