TL;DR: Differences in binding to viral peptide antigens by HLA may be the major factors underlying genetic differences between HIV controllers and progressors, and genome-wide association results implicate the nature of the HLA–viral peptide interaction as the major factor modulating durable control of HIV infection.
Journal ArticleDOI
Transcriptional analysis of HIV-specific CD8 + T cells shows that PD-1 inhibits T cell function by upregulating BATF
Michael Quigley,Florencia Pereyra,Björn Nilsson,Filippos Porichis,Catia Fonseca,Quentin Eichbaum,Boris Julg,Jonathan L. Jesneck,Jonathan L. Jesneck,Kathleen Brosnahan,Sabrina Imam,Kate Russell,Ildiko Toth,Alicja Piechocka-Trocha,Douglas V. Dolfi,Jill M. Angelosanto,Alison Crawford,Haina Shin,Douglas S. Kwon,Jennifer Zupkosky,Loise M. Francisco,Gordon J. Freeman,E. John Wherry,Daniel Kaufmann,Daniel Kaufmann,Bruce D. Walker,Benjamin L. Ebert,Benjamin L. Ebert,Benjamin L. Ebert,W. Nicholas Haining +29 more
TL;DR: It is shown that PD-1 coordinately upregulates a program of genes in exhausted CD8+ T cells from humans and mice that includes upregulation of basic leucine transcription factor, ATF-like (BATF), a transcription factor in the AP-1 family that inhibit T cell function.
Journal ArticleDOI
Consistent Cytotoxic-T-Lymphocyte Targeting of Immunodominant Regions in Human Immunodeficiency Virus across Multiple Ethnicities

Davis,James I. Mullins,Philip J. R. Goulder,Bruce D. Walker,Christian Brander +27 more
TL;DR: Factors that contribute to the immunogenicity of these highly targeted and relatively conserved sequences in HIV that may represent promising vaccine candidates for ethnically heterogeneous populations are identified.
Journal ArticleDOI
HIV-Specific Cytolytic CD4 T Cell Responses During Acute HIV Infection Predict Disease Outcome
Damien Z. Soghoian,Heiko Jessen,Michael Flanders,Kailan Sierra-Davidson,Sam Cutler,Thomas Pertel,Srinika Ranasinghe,Madelene Lindqvist,Isaiah Davis,Kimberly Lane,Jenna Rychert,Eric S. Rosenberg,Alicja Piechocka-Trocha,Abraham L. Brass,Jason M. Brenchley,Bruce D. Walker,Bruce D. Walker,Hendrik Streeck +17 more
TL;DR: The data demonstrate that HIV-specific CD4 T cell responses can be used during the earliest phase of HIV infection as an immunological predictor of subsequent viral set point and disease outcome and suggest that expansion of granzyme A+ HIV- specific cytolytic CD4T cell responses early during acute HIV infection contributes substantially to the control of viral replication.
Journal ArticleDOI
TCR clonotypes modulate the protective effect of HLA class I molecules in HIV-1 infection

Douek,Daniel Kaufmann,Todd M. Allen,Bruce D. Walker,Bruce D. Walker +27 more
TL;DR: The efficacy of such so-called 'protective alleles' is modulated by specific TCR clonotypes selected during natural infection, which provides a functional explanation for divergent HIV-1 outcomes.
