Woods,Jinghai James Xu,Kathy Yarborough,Jan G. Hengstler +94 more
TL;DR: This review encompasses the most important advances in liver functions and hepatotoxicity and analyzes which mechanisms can be studied in vitro and how closely hepatoma, stem cell and iPS cell–derived hepatocyte-like-cells resemble real hepatocytes.
Journal ArticleDOI
Multilineage communication regulates human liver bud development from pluripotency
J. Gray Camp,Keisuke Sekine,Tobias Gerber,Henry Loeffler-Wirth,Hans Binder,Malgorzata Gac,Sabina Kanton,Jorge Kageyama,Georg Damm,Daniel Seehofer,Lenka Belicova,Marc Bickle,Rico Barsacchi,Ryo Okuda,Emi Yoshizawa,Masashi Kimura,Hiroaki Ayabe,Hideki Taniguchi,Takanori Takebe,Takanori Takebe,Barbara Treutlein +20 more
TL;DR: It is found that liver bud hepatoblasts diverge from the two-dimensional lineage, and express epithelial migration signatures characteristic of organ budding, and a striking correspondence between the three-dimensional liver bud and fetal liver cells is found.
Journal ArticleDOI
Hepatocyte pyroptosis and release of inflammasome particles induce stellate cell activation and liver fibrosis
Susanne Gaul,Susanne Gaul,Aleksandra Leszczynska,Fernando Alegre,Fernando Alegre,Benedikt Kaufmann,Casey D. Johnson,Casey D. Johnson,Leon A. Adams,Alexander Wree,Georg Damm,Daniel Seehofer,Carolina Jimenez Calvente,Davide Povero,Tatiana Kisseleva,Akiko Eguchi,Akiko Eguchi,Matthew D. McGeough,Hal M. Hoffman,Pablo Pelegrín,Ulrich Laufs,Ariel E. Feldstein +21 more
TL;DR: It is demonstrated that primary mouse and human hepatocytes can undergo pyroptosis upon NLRP3 inflammasome activation with subsequent release of NLRP1-YFP HEK cells that amplify and perpetuate inflammaome-driven fibrogenesis.
Journal ArticleDOI
Gene networks and transcription factor motifs defining the differentiation of stem cells into hepatocyte-like cells
Patricio Godoy,Patricio Godoy,Wolfgang Schmidt-Heck,Karthick Natarajan,Baltasar Lucendo-Villarin,Dagmara Szkolnicka,Annika Asplund,Petter Björquist,Agata Widera,Regina Stöber,G Campos,Seddik Hammad,Agapios Sachinidis,Umesh Chaudhari,Georg Damm,Thomas S. Weiss,Andreas K. Nussler,Jane Synnergren,Karolina Edlund,Barbara Küppers-Munther,David C. Hay,Jan G. Hengstler +21 more
TL;DR: In this article, the authors performed a transcriptomic study with HLC derived from human embryonic and induced stem cells (ESC, hiPSC) from three different laboratories and identified gene networks representing successful and failed hepatocyte differentiation.
Journal ArticleDOI
Cell sources for in vitro human liver cell culture models.
TL;DR: This review gives an overview of the major human cell sources under investigation for in vitro liver cell culture models, including primary human liver cells, liver cell lines, and stem cells.
