TL;DR: The COVID Human Genetic Effort established to test the general hypothesis that life-threatening COVID-19 in some or most patients may be caused by monogenic inborn errors of immunity to SARS-CoV-2 with incomplete or complete penetrance finds an enrichment in variants predicted to be loss-of-function (pLOF), with a minor allele frequency <0.001.

TL;DR: This article reported very rare, biochemically deleterious X-linked TLR7 variants in 16 unrelated male individuals aged 7 to 71 years (mean: 36.7 years) from a cohort of 1,202 male patients aged 0.5 to 99 years with unexplained critical COVID-19 pneumonia.

TL;DR: In this article , the molecular and cellular determinants of critical COVID-19 pneumonia were reviewed and the TLR3- and TLR7-dependent production of type I interferons by respiratory epithelial cells and plasmacytoid dendritic cells, respectively, is essential for host defence against SARS-CoV-2 infection.

TL;DR: The AIM-IgM association plays a critical role in the obesity-associated autoimmune process, and the increase of multiple IgG autoantibodies observed in obese wild-type mice was abrogated.

Homozygosity for TYK2 P1104A underlies tuberculosis in about 1% of patients in a cohort of European ancestry
Gaspard Kerner,Gaspard Kerner,Noé Ramírez-Alejo,Yoann Seeleuthner,Yoann Seeleuthner,Rui Yang,Masato Ogishi,Aurélie Cobat,Aurélie Cobat,Etienne Patin,Lluis Quintana-Murci,Stéphanie Boisson-Dupuis,Stéphanie Boisson-Dupuis,Stéphanie Boisson-Dupuis,Jean-Laurent Casanova,Laurent Abel,Laurent Abel,Laurent Abel +17 more
TL;DR: A strong enrichment of P1104A homozygotes in a British sample of 620 patients with TB (1%), relative to 114,473 controls, suggests that homozygosity for the P 1104A TYK2 variant may underlie TB in ∼1% of European patients.
