Discovery of alkoxy benzamide derivatives as novel BPTF bromodomain inhibitors via structure-based virtual screening

TL;DR: By integrating docking-based virtual screening with biochemical analysis, this study identified a novel selective BPTF bromodomain inhibitor DCB29 with the IC50 value of 13.2 ± 1.6 μM by homogenous time-resolved fluorescence resonance energy transfer (HTRF) assays and presented great potential as a powerful tool for BPTF-related biological research and further medicinal chemistry optimization.
Journal ArticleDOI

A conserved hydrophobic core at Bcl-xL mediates its structural stability and binding affinity with BH3-domain peptide of pro-apoptotic protein

TL;DR: The BH3-domain peptide binding affinity and pore forming propensity of Bcl-x(L) were correlated to its death-repressor activity, which provides new information to help study the regulatory mechanism of anti-apoptotic proteins.
Journal ArticleDOI

Discovery of potent DOT1L inhibitors by AlphaLISA based High Throughput Screening assay.

TL;DR: A HTS platform based on AlphaLISA method for screening and discovery of DOT1L novel inhibitor is developed, through which compound 3 and its analogues as potent DOT1l inhibitors with promising MLL-rearranged leukemia therapeutic application are discovered.
Journal ArticleDOI

Combinatorial Pharmacophore Modeling of Multidrug and Toxin Extrusion Transporter 1 Inhibitors: a Theoretical Perspective for Understanding Multiple Inhibitory Mechanisms

TL;DR: A combinatorial pharmacophore (CP) model for Multidrug and toxin extrusion 1 (MATE1/SLC47A1) inhibitors was developed based on a data set including 881 compounds and indicated that the small inhibitors matching HHR1 and DRR involve in competitive inhibition, while the relatively large inhibitors matching AAAP are responsible for the noncompetitive inhibition by locking the conformation changing of MATE1.
Journal ArticleDOI

Outline Design of ITER Radial X-Ray Camera Diagnostic

TL;DR: The radial X-ray camera (RXC) as mentioned in this paper was designed to measure the poloidal profile of plasma Xray emission with high spatial and temporal resolution, and its primary diagnostic role includes measuring low (m,...
